Abstract

ABSTRACTGenetic improvement of taro (Colocasia esculenta L.) is one of important research program to increase productivity and give better cultivation on the marginal land. Induction of polyploid plants is a method useful to increase genetic diversity. The aim of this research was to induce polyploid taro by oryzalin treatment. Polyploidy was induced from in vitro diploid taro ‘bentul’ using oryzalin at 7.5; 15; 30; 60 and 75 ?M soaked for 3 days. Regenerated shoots were grown on MS medium containing 2 mg/l BAP, 1 mg/l thiamine and 2 mg/l adenine. Shoot growth was recorded four weeks (subculture-0), eight weeks (subculture-1) and twelve weeks (subculture-2) after treatments. The results showed that survival rate of treated shoots was 100%. Higher oryzalin concentration reduced the proliferation of shoots, petiole length, numbers of leaves as well as the numbers of roots. Ploidy levels analysis determined by flowcytometer for 122 plantlets were investigated. The results indicated that control shoots were diploid. All treated shoots were polyploids. Oryzalin at 60 ?M gave 50% of tetraploid planlets, 30 ?M of oryzalin gave 5.71% hexaploids, 60 ?M of oryzalin gave 9.09% octaploids. All acclimatized plantlets gave 100% survival rate. Keywords : Colocasia esculenta L., taro, in vitro induced polyploidy, oryzalin

Abstract

ABSTRACTThe analysis of palm and sole arschitexture dermatographic traits have been done for two sub genera of Hylobates and Symphalangus in Indonesia. A dermatographical study was conducted to specimens from Indonesia Archipelago on genera Hylobates and Symphalangus, which are deposited in Museum Zoologicum Bogoriense-LIPI. A total of 101 specimens of Hylobates (78 specimens) and Symphalangus (23 specimens) were examined on palm and sole dermatographic traits. The discriminant canonical analyses showed that the Indonesian gibbon has two differences in function space of dermatograph types. By plotting these data, the percentage of the two groups that correctly classified was calculated to be 90.63%. Only nine specimens of Hylobates and Symphalangus were incorrectly classified, seven (9.5%) specimens Hylobates were classified as Symphalangus, and two specimens Symphalangus (9.1%) were classified as Hylobates. The majority of loops type dermatograph in this study indicated that the traits direction to ulnar, radial, and proximal between Hylobates and Symphalangus are significantly different (X2 >0.05, df=3; X2 = 13.58, 10.03 and 7.91 respectively). Keywords: Architecture Dermatographic, Hylobates, Symphalangus

Abstract

Triploid banana breeding could be achieved by crossing tetraploid to diploid banana cultivars. The availaibility of tetraploidbananas is naturally very limited. One of methods to produce tetraploid bananas is inducing tetraploid plants usingchemicals that inhibit spindle fiber formation during mitosis such as oryzalin or colchicine. The present research is aimed toproduce and characterize tetraploid Pisang Mas Lumut (Musa acuminata, AAAA genome) induced by in-vitro oryzalintreatment from diploid Pisang Mas Lumut. Fifteen in vitro-cultured shoots were treated wih oryzalin at of 60μM for 7days in a liquid MS basal medium with addition of 2 mg/l BA. After 6 sub-cultured, 54 shoots and 104 plantlets wereproduced. Forty seedlings were then survived and grew well and healthy. Ploidy identification of induced Pisang MasLumut was conducted using Flowcytometer at seedling stage and repeated at reproductive phases for 2 cycles. The resultsshowed that oryzalin treatments produced tetraploids at a frequency of 30%, diploid at 65% and mixoploid at 5%. Thetetraploid plants have longer and thicker pseudostems, produce fewer suckers, drooping and larger leaves, and broader fruitscompared to its diploid control at harvest time. The tetraploid plants have similar genetic properties with their diploidcontrols as shown by genetic identity of 0.90-0.97.Key words: Pisang Mas Lumut, Musa acuminata, AA, oryzalin, tetraploid
