Abstract

ABSTRACTBrek is a consumption fish belong to barbs group that widely distributed, one of them is in Serayu river. The aims of this study was to reveal the characteristics of meristic, morphometric, and habitat of the species. The study was conducted on the Serayu river especially in Banjarnegara, Central Java Province. Sampling was conducted on May 2012 to June 2013. The area of study was divided into three zones and the zones was categorized based on Reservoirs at Mrica area these are upper zone, reservoir zone, and below the reservoir zone. Fishing gear used are cast net, elektorfishing, and gill nets. Samples of fish were caught preserved in 4-10% formalin solution and then observed in the laboratory. Morphometric characters observed were 24 characters. The results were standardized to standard length, then analyzed with discriminant methods. The structure of the scales and number of ridges on the back of the last dorsal fin spines also observed. The results meristic characters are 4.8 dorsal fin, 3.5 anal fin, 1.14-16 pectoral fin, and predorsal 10-13 scales, also there were no spots on the caudal peduncle. The results of discriminant analysis showed known five characters distinguishing three species near 100%. The habitats have rocky bottom, rapid current, and divide into two groups.Keywords: barb fish, meristic, morphometric, Serayu river

Abstract

ABSTRACTThe Newcastle Disease (ND) virus causes the most important disease in chicken, and very contagiuos in poultry in many countries. In Indonesia the Newcastle Disease or is called tetelo became endemic in chicken currently, the ND virus of genotype VII caused morbidity 100% and mortality 80% in chicken. The fusion and haemaglutinin-neuraminidase protein are involved in adhesion to the surface of hert cells to antibodies. HN and F protein induced by the ND vaccines have neutralizing effects. The aim of this study was to compare the antibody level and protection of several ND vaccine genotypes (genotype I, II, VI and VII) in chicken against ND virus chicken / Indonesia/GTT/11 genotype VII. Our result showed that all genotype ND vaccines (I, II, VI dan VII) produce 100% protection in chicken from clinical and mortality, but the challenge virus excretion in chickens vaccinated with ND vaccine VII genotypes were significantly different (p <0.05) to other groups. Response of antibody in chicken vaccinated ND genotype VII show highest mean titer (GMT 122) compered other ND vaccine. In onclusion, the ND virus chicken/Indonesia/ GTT/11 genotype VII is good to be used as inactive vaccine seed, which appropriate to the field virus circulating at this time.Keywords: virus, Newcastle Disease, genotype, vaccine

Abstract

ABSTRACTVegetation Analysis of Lowland Forest in Batanta Island, Raja Ampat, Papua. A vegetationanalysis of Batanta lowland forest has been made by setting up 17 plots of each 30-m x 30-mdistributed in 3 study sites were Yenanas (5 plots), Yensawai (7 plots) and Wailebet (5 plots).All trees (dbh. e”10 cm) within all of 17 plots were measured, and determined their positions,and identify their species. In total there were 171 tree species recorded within plots andbelonging to 108 genera and 40 families. Pometia pinnata was the most common speciesfollowed by Anthocephalus macrophyllus, Pangium edule, Toxotrophis illicifolius, andKoordersiodendron pinnatum. Almost all of common species such as Pometia pinnata,Anthocephalus macrophyllus, Celtis hidebrandii and Intsia bijuga were observed as theemergent and/or canopy trees. According to ordination analysis there were five communitytypes, Aporusa–Pometia, Antocephalus-Toxotrophis, Sterculia-Grewia, Ficus-Antocephalus,and Duabanga-Pterocymbium communities. Abstract

Tempuyung (Sonchus arvensis L.) is known as an important medicinal plant used as a diuretics andantihypertensives. This plant is widely distributed in Indonesia. Genetic diversity of tempuyung is importantinformation as a database for further research especially in medicinal plant standardization. The objective of thisstudy was to analyse genetic characterization of tempuyung based on SRAP (Sequence-related amplifiedpolymorphism) molecular markers. Thirteen samples were collected from 8 different locations and amplified using5 primer SRAP combinations. Similarity matrix was calculated using Dice coefficient. Unweighted Pair GroupMethod Using Arithmetic Mean (UPGMA) cluster analysis was performed to develop a dendrogram. The resultindicates that there was a genetic variation among tempuyung accessions and divided into 4 clusters with similarityindex of 0,7719. Citeureup and Turen3 accessions were the most closely similar with similarity index of 0,8936. Inconclusion, SRAP markers may serve as an efficient and effective tools to analyze the genetic diversity amongtempuyung accessions.Keywords: genetic characterization, tempuyung (Sonchus arvensis L.), SRAP

Abstract

The Effect of Concentrate Supplementation in The Ration on Performance, Carcass and Non-Carcass Weight of Javan Porcupine (Hystrix javanica F. Cuvier, 1823). This study was conducted at Small Mammal Captivity, Zoology Division, Research Center for Biology,Indonesian Institute of Sciences, Cibinong, Bogor Regency. Eight porcupines consisted of 6 females and 2 males were used to know the effect of concentrate supplementation in the ration on performance, carcass and non carcass weight of javan porcupine. Ration treatment consisted of control ration (PI) and control ration plus concentrate comersial such as koi fish pellet (PII).Four porcupines (3 female and 1 male) were given control ration (PI) and three porcupines (3 female and 1 male) were given ration PII. A completely randomized design was used in this research. Data collected were nutrient consumption, average daily gain, feed conversion, slaughter weight, weight of cascass and non-carcass. The data were analyzed by analysis of variance. The result showed supplementation of koi fish pellet in the ration didn’t significantlyinfluence (P>0.05) performance of javan porcupine, but significantly (P<0.05) increase consumption on calcium and phosphor. Weight and percentage of carcass and non-carcass of javan porcupine were not affected by ration treatment. Average of weight and percentage of porcupine’s carcass of PI were 4,505.08 g and 59.67% and those of PII were 3,957.55 g and 56.54%, while average of weight and percentage of porcupine’s non-carcass of PI were 2,979.05g and 39.46% and those of PII were 2,774.60 g and 39.64%.Keywords: Concentrate, ration, performace, carcass, non-carcass

Abstract

ABSTRACTThe Parasitics wasp superfamily Chalcidoidea (Insecta : Hymenoptera) potentialas biological control agents for pests of Eggplant (Solanum melongena) and BitterMelon (Momordica charantia). The recent contribution on diversity of parasitics waspChalcidoidea for biological control perspective against the pests of the two economiccrops was discussed, with emphasis on the most significant steps for the selection ofcandidate biological control agents. The survey was carried out in Bogor, West Javausing Malaise traps, Yellow pan traps and a Swept net. The crops are the most economicallyimportant in agriculture not only in West Java but also in Indonesia. Meanwhile the pestcontrol of those two crops has so far been used only by the insecticides and would affectto the health of the production of the crops when the vegetables are consumed freshly. Ofthe 78 species within 8 familes of Chalcidoidea are found in both crops, 7 species can beclassified as potential biological control agents. With the object of biological control ofthese 7 species, a survey was carried out to establish the presence and importance oftheir natural enemies on those crops in Java.Key words : Bitter Melon, Chalcidoidea, Eggplant, Parasitoids, Pest control.

Abstract

ABSTRACTSomatic cell manipulation of Artemisia annua L. was conducted by induction of polyploid plants with Colchicine and Oryzalin in order to increase level of artemisinin. Polyploid plantlets were multiplied on MS medium without plant growth regulators. After acclimation processes, plants were grown in the field for agronomic performance observation. Survival rate of plantlets was recorded. Agronomic performance of plants was observed by recording height of plants, number of branches, leaf biomass, stomatal characteristics, and artemisinin content. The results showed that survival rate of the plantlets from Colchicine and Oryzalin treatments were ranging from 13.40 to 33.33% and 11.11 to 41.67%, respectively. Growth rates of plant height and plant branching were not significantly different between diploid and tetraploid plant both from Colchicine and Oryzalin treatments, except to triploid plants from Colchicine treatment. Averages of plant height from Colchicine and Oryzalin treatments were ranging from 10.0 to 220.0 cm and from 35.0 to 186.0 cm, respectively. The averages number of branches per plant of polyploid plants from Colchicine and Oryzalin treatments were ranging from 3 to 66 and from 11 to 63, respectively. Averages of dry leaves biomass between diploid and tetraploid plant from Colchicine and Oryzalin treatments were also not significantly different. They were ranging from 12 to 64 g/plant and from 11 to 62 g/plant, respectively. However, tetraploid clones have bigger size of stomata and produced more artemisinin than the diploids.Keywords: Artemisia annua L, Colchicine, Oryzalin, Polyploids, Acclimation, Agronomic performance

Abstract

 ABSTRACTExopolysaccharides (EPS) are water soluble polymers synthesized by three strains of lactic acid bacteria (LAB) namely EPS-S1, EPS-S2 and EPS-S7. Some exopolysaccharides (EPSs) produced by lactic acid bacteria (LAB) present potential health-beneficial properties, such as immune stimulation and antioxidative activities. Antioxidants function in several ways including preventing the formation of radicals, scavenging free radicals, formation of hydrogen peroxide and lipid peroxides. The Objectives of this research were to evaluate the antioxidant activities of exopolysaccharides (EPS) extracts from lactic acid bacteria (LAB). The extracts were investigated by using in vitro assays induced tersier-Butilhidroperoksida (t-BHP) 6 µg.ml-1 red blood cell (RBC) for their effects on lipid peroxidation malondealdehyde (MDA) level and  activities of some antioxidant enzymes which includes catalase (CAT) and superoxide dismutase (SOD). Six groups consisted of 3 replicated per group; (I) normal control, RBC (II) negative control, RBC+ t-BHP (III) positive control, RBC+t-BHP+vitamin-C, and (IV-VI) samples, RBC+t-BHP+EPS (S1, S2 and S7) extracts. The results showed that exopolysaccharides (EPS) of three strains of lactic acid bacteria (LAB) increased catalase (CAT) and superoxide dismutase (SOD) enzyme activity and decreased malondealdehyde (MDA) content, exopolysaccharides (EPS)-S2 most potential to increase the activity of catalase (CAT; 77.46%) and superoxide dismutase (SOD; 33.87%) enzyme and inhibit the increase malondealdehyde (MDA; 32.19%) levels. Keywords: exopolysaccharide, lactic acid bacteria, malondealdehyde (MDA), superoxide dismutase (SOD), catalase (CAT)
