Abstract

Avian Influenza (AI) or Bird Flu is a fatal zoonotic disease caused by highly pathogenic avian influenza(HPAI) virus of H5N1 sub-type. The disease is still endemic in Indonesia. This study was conducted toinvestigate AI cases in chickens in Bali. Virus isolation was performed in 9 day-old embryonated chickeneggs, and then followed by serologic testing by haemaglutination (HA) and Haemaglutination Inhibition(HI) assay using standard microtiter procedure. All of the samples were further tested with reversetrancriptasepolymerase chain reaction (RT-PCR). All work has been done in the Biomedical and MolecularBiology Laboratory, Faculty of Veterinary Medicine, Udayana University, Denpasar, during the period2009-2011. A total of ten samples were examined A total of ten chicken samples consisting of 6 fieldsamples and 4 meat samples have been confirmed to be AIV H5N1. All field cases showed clinical signsand gross pathology that were typical to the infection of avian influenza. The result indicates that AI casesare still prevalent among chickens in Bali.

Abstract

The administration of Pregnant Mare Serum Gonadotrophin (PMSG) hormone to stimulate superovulation has been proven to improved fetal prenatal growth, birth weight, mammary gland growth anddevelopment, milk production, litter size, as well as pre and post weaning growth. This study was conductedto apply the administration of PMSG hormone technology in small-holder farms. One hundred ewes weredivided into two groups. Group I (control: 50 ewes) ewes without administration of PMSG and Group 2(treatment: 50 ewes) ewes with administration of PMSG to stimulate super ovulation and improveendogenous secretion of pregnant hormones. The application of PMSG increased the numbers of lamb bornby 26.8% (69 vs. 87) with average litter size of 20.7% (1.38 vs. 1.74). The lamb weight at birth was alsoincreased by 25.7% (2.76 ± 0.33 vs. 3.47 ± 0.35 kg). The milk production of the dam was also increased by29.7% (0.64 ± 0.25 vs. 0.91 ± 0.22 L/ewe/day). Lambs born to ewes administered PMSG had better weaningweight 22.1% (12.32 ± 2.44 vs. 15.04 ± 1.11 kg/ewe). The number of survival lamb to reach weaning age washigher compared to the control group. The weight at weaning age was almost twice (628.4 kg vs. 1202.8 kg)compared to the control group. Using simple economic analysis calculation, the application of this hormonetechnology could increase gross revenue by Rp 294.280- per ewes. It was concluded that the administrationof PMSG is economically feasible to be applied in to the small-holder farms.

Abstract

Antioxidant activity of clove leaf was investigated. The clove leaves (Eugenia aromatica) wereprepared with reflux extraction using methanol, water, and ethanol. Total activities of the extractsand reducing power were measured with thiocyanate method and reducing potential method. Resultshowed that the highest total antioxidant activity was observed in methanol extract. It appearsthat the ability of this extract for partitioning at the interface of emulsion in tested oxidationsystem was the highest among the other extracts, therefore it had the best activity to inhibitoxidation. The type of phenolic compounds of this extract appeared to be responsible for the highestradical scavenging capacity. The same phenomenon occurred for reducing power, methanol extracthad the highest reducing power, thereby suggesting that each extracts comprised different type ofphenol based on different polarity of reflux used for extraction. Total antioxidant activity and thehighest reducing power obtained from methanol extract. Both are closely related to total phenolcontent of the clove leaves.

Abstract

The studies have reported that ginger have many activities such as antiemesis, anti-inflammatory,anti-bacterial and anti-parasites. Therefore, this study was conducted to evaluate antiviral effect of essentialred ginger oil againts Avian Influenza (AI) in ovo using hemagglutination test (HA). Avian Influenzaviruses were treated with 0,01%, 0,1% and 1% of essential red ginger oil, and then inoculated in chickenembryonated egg via allantoic sac. Allantoic fluid was harvested using for HA test . Result of this studyshows that application of 1% of essential red ginger oil results in the reduction of titer HA . Interestingly,essential oil shows antiviral activity revealed HA titre 20 whereas the titre HA AI which AI virus treatedwith 0,01% and 0,1% essential red ginger oil, the HA titer was 25. The conclution of this study proved thatessensial oil 1% of the red gingger is the best concentration as antiviral activity .

Abstract

The aim of this study was to evaluate the effectiveness of ethanol turmeric extract ointment in woundhealing process of streptozotocin-induced diabetic mice. Ethyl acetate and hexane fractions of ethanolturmeric extract were studied for their wound-healing properties in the formulation of ointment. Sixty micewere divided into 4 groups : group I was control (normal control, without treatment); group II was diabeticgroup (diabetic, without treatment); group III was diabetic and treated with ethyl acetate fraction ointment,group IV was diabetic and treated with hexane fraction ointment. The diabetic groups were i.p injectedwith 40mg/kg of streptozotocin and all groups were received incision 2 cm on their back skin. The ointmentsof ethyl acetate and hexane fraction were given topically twice a day. Three mice from each groups werenecropsied at 2nd, 4th, 7th, 14th and 21st days post incision (pi) for gross pathological and histopathologicalevaluation of the injured skin. Gross examination revealed that the ethyl acetate and hexane fractionointment groups showed better result on wound-healing process compared to the diabetic group.Microscopically, the ethyl acetate and hexane fraction ointment groups showed faster neovascularizationand reepithelialization compared to the diabetic group. In comparison with the diabetic group, the ethylacetate and hexane fraction ointment groups showed fewer neutrophils infiltration which indicated antiinflammatory activities of ethyl acetate and hexane fractions. Based on the macroscopic and microscopicobservation, the ointments of ethyl acetate and hexane fraction have properties to promote wound healingin diabetic mice.

Abstract

In order to study the distribution of Newcastle disease virus (NDV) following infection, chickenswere experimentally infected with visceretropic velogenic NDV isolate. Monoclonal antibodies (mAbs)against the NDV LaSota vaccine strain were then produced to detect viral antigen in the infectedorgans. The mAbs were firstly tested for their specificity by enzyme linked immunosorbent assay(ELISA) using NDV and normal allantoic fluids as antigens. Eight mAbs specific against NDVwere isolated and two mAbs were used for immunodetection of NDV antigen in chicken’s tissues.By immunohistochemistry labeled streptavidin-biotin (LSAB) staining NDV–antigen was detectedin paraffin embedded tissues of NDV-infected chickens. NDV antigen was not detected in noninfected chickens. In the infected chickens, high intensity of NDV antigen was detected in thelymphoid tissues, lung and intestine. The NDV antigen with a lesser intensity was detected in thebrain, trachea, liver and myocardium. This study shows that although viscerotropic velogenicNDV isolate can infect almost all organs, the main target of infection are lung, intestine andlymphoids tissues
