Abstract

Identification of Viruses that Causes Mosaic Diseases in Melons(Cucumis melo L.) in Bali The aims of this research was to identify of some virus that infects melons in Bali. The first step of this research was to take samples in Jembrana Regency, TabananRegency, Gianyar Regency, Denpasar City, and Badung Regency. The next step wasdoing virus detection at Laboratory of Plant Diseases, Faculty of Agriculture, UdayanaUniversity. Variations of symptoms found in the field were mosaic, yellow leaves,malformations, and burning spots on the leaves. The percentage of virus attacks inGianyar and Badung was the highest percentage (60.00%), Jembrana (57.14%),Denpasar (42.85%), and Tabanan (37.50%). To know type of virus that causes thisdisease, molecular identification was conducted. Extraction was the first step to gettotal RNAs from symptomatic leaves. Total RNAs was transcribed intoComplementary (c) DNAs using reverse transctiption-polymerase chain reaction (RTPCR)technique. Polymerase chain reaction (PCR) was conducted using cDNA and three primer pairs of CABYV-489-F/Pol-G-R, MABYV-262-F/Pol-G-R andCIFOR/CIREV. Primers would be use to amplified a part of coat protein. CABYV489-F/Pol-G-Rand MABYV-262-F/Pol-G-R is specific primer, and CIFOR/CIREV would be use to amplified gene of Potyvirus. Based on the results of molecular with PCR it was found that melons in Bali were positively infected with viruses from thePotyvirus group with a size of around 683 bp in samples from Gianyar and Denpasar.

Abstract

ABSTRACT Study of Anti fungi Extract of Some Types of Marine Biota to the Apergillus flavus LINK. and Penicillium sp. LINK. The purpose of this study is to know the effectiveness of marine biota, namely Aglaophenia sp., Eucheuma cottonii, Sargassum sp., Gracilaria sp., and Ulva sp. in impeding the growth of Aspergillus flavus and Penicillium sp. fungi which is isolated from contaminated corn seed. The results of this study shown that the crude extract of marine biota is very effective in impeding the growth of Aspergillus flavus and Penicillium sp. fungi. Extract which is capable forming the greater impeding zone is Aglaophenia sp, that is 68,20 mm to Penicillium sp. with very strong category of impeading power. Aglaophenia sp. ekstract shown a greater empeading power to the colony of fungi, that is 49,40% to Aspergillus flavus and 49,20% to Penicillium sp. Abstract

The Effects of Processing Delays on Storage Ability of Soybean Seed (Glycine max L. Merril) Soybeans are the third important food crop in Indonesia after rice and maize. Proper handling or seed processing will affect the quality of the seeds obtained. Generally soybean seed can be stored for 3-4 months with the correct handling process. Delays in seed processing affect the appearance of seed quality both physical and physiological. Weather factors and facility limitations are the reasons for delaying seed processing after harvest. Soybean seed storage is important because soybean seeds are likely to be stored before the seeds are planted. This study aims to determine the effect of delayed processing that can be tolerated to store ability of soybean seeds. The experimental desain used was Completely Randomized Design (RAL) by a single factor with a processing delay which is consisting of five treatments and five repetitions. The observed variables were moisture content (%), germination (%), vigor saving test (%), growthability test (%), and electrical conductivity test (?mhos cm-1 g1).The results showed that delays of processing time after harvest in soybean seeds can effect the shelf life. The longer the processing time was delayed, the lower the storage ability of the seed. Seeds without delay in processing time can maintain seed viability for eight weeks of storage with the lowest electrical conductivity of 2,47 ? mhos cm-1 g1. Processing one day delays and two day delay after harvest can be tolerated because the seeds produced have a germination greater than 80% and still meet the standard of seed quality.

Abstract

The Effect of Lateral Branch Trimming Against Results and Quality of LongBean Seeds (Vigna sinensis L.) This study aims to determine the effect of the number of leaves that are left on the lateral branch roasting on the results of young pods of long bean plants. Thistudy uses a randomized block design (RBD). Treatment of the number of leaves lefton lateral branch pruning includes: total lateral pruning, leaving 2 leaves, leaving 4eaves, leaving 6 leaves and leaving 8 leaves on lateral branch pruning. This researchwas repeated 5 times, so as many as 25 experimental units were obtained. Variablesobserved included: main stem length, total number of leaves per plant, total leaf areaper plant, fresh weight and oven dry weight of plant parts above ground, number ofharvest pods per plant, fresh pod weight per plant, weight of oven dried pods perplant and the weight of fresh pods per hectare. The results showed that more leaveswere left on the lateral branch pruning causing the number of leaves, leaf area,weight of the plant part above the ground, and oven dry weight of the plant part onhe ground to increase. Total lateral pruning pruning treatment resulted in the numberof crop harvest pods, fresh pod weight, and oven pod dry weight with the highestaverage value. The total lateral pruning treatment resulted in the highest per hectarefresh pod weight of 25.59 tons.

Abstract

Identification and Characterization of Genetic Resources of Local Fruits in GianyarThe local fruits is every kinds of fruits are cultivated and developed by farmers in Bali, while the local fruit products is every types and derivative results which derived from fresh fruits or already processed. This study conducted with the aims to identify the types of genetic resources of local fruits in Gianyar, to compile the genetic resources of the local fruits based on the morphological characters and utilization, superior fruit, harvest seasons and the geographical map of the diversity distribution of the local fruits. This study was conducted in February-July 2015 throughout Sub-district in the Gianyar District, namely: Payangan, Tegallalang, Tampaksiring, Ubud, Blahbatuh, Gianyar and Sukawati. The study consists of three phases, namely; Secondary data collection, field surveys for the types of genetic resources and distribution, and identify of the morphological characters and agronomy, utilization, superior fruit, harvest seasons and the distribution map of the local fruits. The results of this study shown, was found 45 species and 41 sub species of fruits which are spread throughout Sub-districts, and the three of seeded commodities namely; Citrus, Mangoes and Papaya.

Abstract

ABSTRACT The Identification of Traditional Settlement Patterns of Setu Babakan Betawinese Cultural Village, District of Srengseng Sawah, Subdistrict of Jagakarsa, South Jakarta, the Province of  DKI JAKARTA The objective of the research is to show an evidence that the form of traditional settlement patterns of Betawinese does still exist in DKI Jakarta. The method used is descriptive quantitative, in which data source was derived from the literature study, observations, structured interviews, and questionnaires (30 respondents). The results of the research showed that Setu Babakan Betawinese Cultural Village settlement patterns uses clustered settlement pattern which follows the circular shape of the Lake/Setu Babakan, and has the characteristic of clustered spread distribution pattern. The former elements in this settlement are physical, economical and social cultural, and they are inter-related. This settlement has the area about 289 ha (67 ha owned by the Local Government) with the percentage of built-up space by 61.17% and 38.83% undeveloped area (include water bodies). The spatial patterns in Setu Babakan was not visible any philosophical history or any clear layout. The existing vegetations in the settlement are of particularly those plants with the characteristic of Betawi producing plants, such as Durio zibetinus Var. Sitokong, Baccaurea racemosa and Sandoricum loetjape. The results of research in the field, 43% of existing plants in the settlements has functioned as a producing plants. This Betawinese settlement is a typical “suburban Betawinese settlement”, which has two types of traditional houses, Rumah Gudang and Bapang/Kebaya and do not have the wind direction and specific orientation in placement. Abstract

The Attempt to Improve the Quality of Balinese Grapevine Fruits (Vitis vinifera L. Var Alphonso Lavallee) With GA Application from Bamboo Shoot Extract at The Blooming Phase Balinese table grape is one of the superior fruit in Bali. Balinese table grape less salable than imported grapes because of the lack of the sweetness, small fruit size and the presence of the seeds are the weaknesses of Balinese table grapes quality. This study aims to determine the effectiveness of the bamboo shoots extract at the blooming phase to improve fruit quality. The experimental design used was a Randomized Block Design with 5 treatments and 5 replications. The treatments are different concentration of bamboo shoot extract including without bamboo shoot extract (control), 25%, 50%, 75% and 100% of bamboo shoot extract. The observed variables were fruitset length, number of fruit per fruitset, fruit weight per fruitset, weight per fruit, number of seeds per fruit, total soluble solids and organoleptic test. The result showed that 100% bamboo shoot extract concentration was able to improve the quality of Balinese grapevines with fruitset length of 20.93 cm increased by 157.3% compared to control, the number of seeds 1.36 decreased by 54% compared to control, and total soluble solids 8.79oBrix increased by 122.8% compared with controls.

Abstract

In vitro culture of citrus plant Pamelo (Citrus maxima Merr.) Using Explant seeds as well as the detection of the presence of Bacteria Liberobacter asiaticus at Shoots are produced This research was begun with doing sample approachment in Kapal Village, Mengwi Sub-District, Badung regency and Sumerta Klod Village, East Denpasar Sub-District, Denpasar City and the culture in vitro was done in Implementing Technical Unit (UPT) Sumberdaya Genetika dan Biologi Molekuler Laboratory since October 2018 until January 2019. This research aim to recognize the response of pamelo orange's seed explant rather it will grow in based MS media which has been richen and got pamelo orange seed that is free from the infection of liberobcter asiaticus. The approachment of sample was based on the visualization of the plant, the plant which shows any indication of CVPD disease next will be known as 1st Sample and 2nd sample would be the plant which shows no indication of cvpd disease. The seed of each sample be cultured with MS media which has been richen, was maintained for 8 weeks after it's planted (mst). By the results of this research, it concludes that there's seed explant from both samples which grows without showing any indication of cvpd disease, but the result of DNA amplification from 1st sample that has been cultured in vitro after got UV transluminator visualization shows the existence of 1160 bp DNA ribbon which is belong to Liberobcter asiaticus. This shows that the sample has been infected by Liberobacter asiaticus, whereas can't be found in 2nd sample.

Abstract

ABSTRACT Utilization of Trichoderma sp. and Grafting to Controlthe Mayor Diseases of Tomato (Licopersicum esculentum Mill.) in Bangli Village, Baturiti, Tabanan Control of plant diseases is one of the constraints in the cultivation of tomatoes. Control method performed by farmers generally use synthetic pesticides, however that cause environmental pollution. The use of Trichoderma sp. and grafting is an environmentally friendly technology in controlling plant diseases. The purpose of this study was in order to determine the ability of Trichoderma sp. and the grafting between the eggplant EG203 strain as rootstock and scions tomatoes as to control major diseases and improve tomato production. This study used a factorial randomized block design with six treatments and four replications.Diseases were found in field tomatoes are late blight (Phytophthora infestans) and yellow curly leaf disease (Tomato Yellow Leaf Curl Virus). Statistical analysis showed that Trichoderma sp. and grafting can reduce the disease severity of leaf blight and increase the production of tomato plants, but was unable to control the disease yellow leaf curl. Average of disease severity of leaf blight on grafting with screen and Trichoderma sp. was lower (61.11 and 62.03%) when compared to treatment without grafting and without Trichoderma sp. (82.99 and 75.47%). Average of yields on treatment grafting with screen and Trichoderma sp. was higher (3912.50 and 3822.22 g/plant) compared to treatment without grafting and without Trichoderma sp. (2858.33 and 3280.55 g/plant). Keywords: disease of tomato, Trichoderma sp., and grafting.
