Abstract

Cymbidium sp. is one of the most popular orchid desired in public, it means high productivity is needed in its fulfillment. One of the obstacle of Cymbidium sp. orchid in order to fullfilment of productivities are the stress and orchid’s seed size which is very small. Vesicular Arbuscular Mycorrhizae is a symbiont fungus that can help absorption of nutrients in stress phase. VAM help Cymbidium sp. to maintenance and increase the productivity. This research aimed to determined VAM species and colonization in rhizospher of Cymbidium sp. orchid. This research has been conducted from November 2020 untill April 2021. Spore isolation was conducted by using wet shievings method, meanwhile VAM colonization was conducted by using roots staining techniques. The result showed there were 4 genus consisted by 6 different type of spores which has been found. There was Glomus (2 types), Dentiscutata (2 types), Acaulospora (1 type), and Gigaspora (1 type). Colonization of VAM in sample plant has been known by presence of VAM special structure, there were vesicular and internal hyphae.

Abstract

Isolation and identification of arbuscular mycorrhizal fungi (AMF) in cashew plants was carried out in Datah Village, Abang District of Karangasem Regency. This study aims to determine the diversity of mycorrhizal spores in the cashew rhizosphere and AMF infections in the roots of cashew plants. The result in microscopic observations found 5 genera of AMF spores such as Glomus, Racocetra, Entrophospora, Septoglomus, and Acaulospora and the population of the genus Glomus were found most dominant in the rhizosphere of cashew plants. The result of staining root with trypan blue obtained the internal structure of AMF in the form of vesicles, arbuscules and hyphae found in root tissue of cashew plants. The result of AMF DNA amplification with primer NS31 and AML2 showed 560 bp bands in electrophoresis with 1% agarose gel determined AMF infection in root of cashew plants.

Abstract

The research was coducted at Laboratory of Biotechnology Faculty of Agriculture Udayana University. The purpose of this research were to investigate moleculer detection of CVPD pathogen in its vector, other phytophagous insects on citrus cv, Siam and kemuning (Murraya paniculata L. Jack.). The result of this research showed that adults and nymphs of Diaphorina citri Kuw. could transmit CVPD disease, but the pathogen of CVPD could not be transovarially transmited. As a host of CVPD disease , kemuning could be source of infection. The adult of Toxoptera citricidus Kirkaldy and Maleuterpes dentipes Hell. could not potensial transmited CVPD disease.

Abstract

Citrus Vein Phloem Degeneration (CVPD) disease is a major obstacle in the effort to develop and increase the production of citrus fruits in Bali. The study on the polymorphism of CVPDr DNA fragment shows that the CVPDr DNA fragment is resistant factor againt CVPD disease. This study try to elaborate the difference in resistance led to differences in plant nutrients deficiencies in the citrus plant with CVPD disease. . Besides, there are also difference in the quality of fruit due to CVPD disease attacks such as water content, vitamin C content and antioxidants in citrus fruits, color, flavor, taste and texture and fruit into small, hard and sour taste.

Abstract

Reishi (Ganoderma lucidum (Curtis) P. Karst) and shiitake (Lentinula edodes (Berk.) Pegler) mushrooms have high economic value and contain bioactive metabolites that provide heatlh benefits. Shiitake mushrooms have a delightfully unique taste as an edible mushroom. Mushroom production can be increased by utilizing microbes known as MGPB (Mushroom Growth Promoting Bacteria). This study aimed to determine the presence of bacteria associated with shiitake and reishi mushrooms in the baglog and determine how these bacteria influence the growth of shiitake and reishi mycelium. Bacteria was isolated from the mushroom baglog then were purified and centrifuged to obtain the cell-free extract. Furthermore, the bacterial cell-free extracts were poured on PDA media, then shiitake and reishi mycelium were inoculated to test the effect of bacteria isolates on fungal growth. The results showed that three isolates have the potential as MGPB in reishi mushroom, i.e., isolate S1C, S1D, and S2A. Reishi mycelium grown on isolate S2A achieved the fastest maximum growth on day 6th. In shiitake mushroom, it was seen that all isolates showed a negative effect on mycelium growth. Shiitake mycelium in isolate S1C, S1D, S2A, and S2B were seen to form a clear zone on PDA medium.

Abstract

Bulung Sangu (Gracilaria spp) as many other macroalgae, is a source of beneficial phytochemical for health. Phytochemicals in macroalgae are widely used as active ingredients in cosmetic preparations. The study was aimed to determine the antioxidant properties of Bulung Sangu formulated in topical preparation. Bulung Sangu was macerated in ethanol 96% following by extract formulation in cream preparation in two concentrations: 5% and 10%. Antioxidant activity was determined against DPPH radical, compared to ascorbic acid. Percentage of reducing power in five concentration series (10, 20, 30, 40, and 50 µm/ml) statistical analyzes using one-way ANOVA with 95% of confidence level, following by IC50 measurement. Absorbance was read in wavelength of 517 nm. Concentration of 5% and 10% cream extract exhibited IC50 of 13,091 µg/ml and 11,867 µg/ml, respectively. These values are lower than IC50 of ascorbic acid (18,505 µg/ml), indicating the strong antioxidant properties of Bulung Sangu extract cream which is more potential than ascorbic acid. Bulung Sangu exhibit a potential source for being developed as medicine and cosmetics in topical preparation.

Abstract

Arbuscular mycorrhizal fungi live in a symbiotic mutualism with plant roots and helps plant absorbnutrients and also able to live in various places. The study aimed to identify arbuscular mycorrhizal fungiin rizhosphere of several grass and cacao through microscopic method based on morphologicalcharacteristics and was conducted from December 2013 to March 2014. The methods used in this studywere spore isolation using wet sieving and root staining techniques. The results showed that arbuscularmycorrhizal fungi spores found in Imperata cylindrica L. were spores of Glomus (Glomales: Glomeaceae)with its vesicular and hyphae structure. The presence of spores and structures in Paspalum notatum wereAcaulospora (Glomales: Acaulosporaceae) and Gigaspora with its arbuscules, vesicules and hyphae structure.Spores and structures found in the Pennisetum purpureum were belong to genus of Glomus with hyphaeand arbuscular structure. Spores and structures of mycorrhizal fungi in Cyperus rotundus are spores of thegenus of Gigaspora (Glomales: Gigasporineae) and spores of Glomus with internal hyphae structure. Whilespores and mycorrhizal structures in Cacao were found two types of spore belong to genus of Glomus withhyphae and vesicular structures.

Abstract

Phenol is an organic compound that is toxic to humans and the environment. This compound was contained in the hospital liquid waste and petrochemical-based industrial sectors. This study aims to obtain hydrocarbonoclastic bacterial isolates that act as biodegradation agent in waste handling. Samples obtained from the biofilter tank of liquid waste treatment Bhayangkara Hospital Denpasar,Bali, Indonesia then carried out isolation and inoculation of bacteria in selective media NA (Nutrient Agar) containing Phenol. The bacteria obtained was performed the viability test on the media with various Phenol concentration levels, obtained three isolates of bacteria capable of growing to the concentration Phenol 500 mg / L, i.e. isolates IB2, IB5, and IB10. Therefore, it was performed the degradation ability and rate test on those three isolates. After 72 hours of incubation periods, the change of Phenol concentrations was analyzed using the folinciocalteau method and examined using the spectrophotometer with 660 nm of wavelength. Based on the analysis of final phenol content, the IB2 isolate has the highest degradation capability in the concentration of 600 mg/L (153,4 mg) with a degradation rate of 2,13 mg/h. IB5 isolates have the highest degradation capability in the concentrations of 700 mg/L (109,45 mg) with a degradation rate of 1,52 mg/h. Meanwhile, IB10 isolates show the lowest ability and degradation rate in both concentrations. Through morphological identification and Gram stain, three isolates belong to the Gram-negative rod-shaped bacteria, which has similarities with the Pseudomonas genus.

Abstract

Training on ginger wilt diseases was held at Taro village, Gianyar regency on 18 st July 2013. The activitiesaimed how to know control of ginger wilt diseases. The training was attended by 25 participants from local farmergroups of Taro village. The methods used in activities were lectures, demonstration and practice in the gingerfield. All participants participated enthusiastically and hope they have the next intensive training again.

Abstract

Training on jumping plantlice of citrus (Diaphorina citri Kuw) the vector of CVPD (Citrus Vein Phloem Degeneration) has been held at Taro village, Gianyar regency on 30st July 2012. The activities aimed to know bioecology of D. citri, the vector of CVPD. The training was attended by 32 participants from local farmer groups of Taro village. The method used in activities were lectures, demontration and practical in the citrus field. All participants participited anthusiastically and hope the have the next intensive traning of the citrus culture.
