Abstract

Plantaricin IIA-1A5 is a bacteriocin produced by Lactobacillus plantarum IIA-1A5 which is isolated from Indonesian beef, and it inhibits activity of Gram negative and positive pathogenic bacteria. However, preparation of the antibacterial agent for further applications or studies is costly due to the usage of a bacterial medium. Therefore, this study was aimed to investigate the feasibility of cheese whey as a growth medium for production of the bacteriocin. The growth curve of Lactobacillus plantarum IIA-1A5 in producing antimicrobial compounds was found to occur in the logarithmic phase with an incubation time of 28 and 32 hours. Purification of plantaricin IIA-1A5 produced peptides with a molecular weight of 9.59 kDa consisting of whey and whey+ (20g/L sucrose, 12.5 g/L tryptone and 7.5 g/L yeast extract); thus, the peptide was grouped as class IIa (<10 kDa) bacteriocin. The protein concentration of plantaricin IIA-1A5 with whey+ (sucrose, tryptone, and yeast extract) treatment and whey treatment was 1883.17 mg/ml and lower than 325.58 mg/ml, respectively. Based on the antimicrobial activity test using a paper disc method, plantaricin IIA-1A5 demonstrated antimicrobial activity against Staphylococcus aureus ATCC 25923; the whey and whey+ treatment yielded 38.02 IU/dL and 321 IU/dL, respectively, while antimicrobial activity against Escherichia coli ATCC 25922 using whey and whey+ treatment yielded 44.85 IU/dL and 172.08 IU/dL, respectively. The effectiveness of the antimicrobial properties of plantaricin IIA-1A5 in the whey medium is proven through the results of this study. In short, the whey is appropriate growth medium for bacteriocin production.

Abstract

Metagenomic approaches are highly valuable in food microbiology, particularly for the investigation of traditional fermented products such as dangke buffalo milk cheese from South Sulawesi, Indonesia. Buffalo milk, a primary raw material, contains diverse biochemical and bioactive components that are produced by microbial activity during fermentation. Unlike conventional culture-based techniques, metagenomics enables the comprehensive characterization of microbial communities directly from food matrices. This study aimed to identify the key microbial taxa that contribute to the dangke quality of fermentation and to understand the factors influencing it. Bacterial 16S rRNA genes were amplified using primers 27F and 1492R under optimized polymerase chain reaction conditions. DNA concentrations were measured with NanoDrop and Qubit instruments, and sequencing was conducted using Oxford Nanopore Technology with MinKNOW software (v23.04.5). Metagenomic analysis revealed Enterococcus faecium as the dominant lactic acid bacterium across all dangke samples (P1–P3), emphasizing its crucial role in fermentation and probiotic potential. These findings suggest that E. faecium can enhance the quality and functional properties of dangke. Moreover, metagenomic tools can support the development of standardized fermentation practices and microbial safety assessments for traditional dairy products.

Abstract

The aim of the research was to evaluate the effect of fermented sausages using probiotic bacterial L. plantarum IIA-2C12 to the gastrointestinal tract of rats. The research was conducted from March to June 2014. Research was conducted at Breeding and Genetics Laboratory, Faculty of Animal Husbandry Laboratory Large Ruminants and UPH Faculty of Veterinary Medicine, Bogor Agricultural University. This study was divided into two stages, 1) subculturing (fermented sausage and beef satay) starter culture, the manufacture of casein, beef satay and sausage fermentation and, 2) in vivo evaluation. Rats were divided into three groups: casein (P1), fermented sausages (P2) and beef satay (P3). This research used a pattern completely randomized design with three treatments type of feed is feed casein, fermented sausage and beef satay. The results showed that based on the amount of lactic acid bacteria, fermented sausage produced met the criteria as a probiotic food. Fermented sausages could increased the population of LAB on the 20th day mucose (T2). Treatment of feed had no significant effect (P> 0.05) in the number of E. coli in the rat liver, but on the 10th day termination (T1), the rats P1 showed the number of BAL were significantly (P <0.05) higher than P2. Fermented sausage increased the population of LAB, decreased E. coli population in mucosa until 20th day in mucosa.

Abstract

In Indonesia, broiler meat is available abundant mainly in traditional market. Unfortunately, this kind of meat is perishable most physical, organoleptic, and microbiological deterioration. To conserve consumers from low quality of meat, this study was conducted to evaluate the physical, organoleptic, and microbiological quality of meat. Meat evaluated was obtained from three traditional markets determined purposively, namely A market, B market, and C market. The physical quality measured were pH and aw; the microbiological quality were total plate count and E. coli number; the organoleptic were meat color, texture, aroma that value in hedonic and hedonic rank test. This study showed that physically the meat was not really decrease although it had pH higher than normal pH of meat. The value of aw reached was 0.84-0.85. The microbial contaminant was higher than the number of microbial allowed. However, organoleptic value indicated that meat were acceptable for panelists.

Abstract

This study was conducted to determine the effect of different temperatures storage on the value of pH, water holding capacity, water activities, Total Plate Count and Escherichia coli contamination of beef meatballs during storage. The result showed that the highest pH value was in the freezer storage (5.83 ± 0.023). The highest value of water holding capacity was obtained at treatment dingingerator storage (23.33 ± 7.026 % water-free), while the results of the analysis of variance showed that effect of different temperature storage was not signiicant (P <0.05) to the water holding capacity. Aw values in this study ranged from 0.854 to 0.908. Aw values at refrigerator and freezer storage were signiicantly higher (P> 0.05) than the meatballs in fresh condition. The analysis of variance showed that treatment of different temperature storage had no signiicant effect (P <0.05) for Total Plate Count (TPC). Mean of TPC (log10 cfu/g) ranged between 5,30 + 0,62 to 5,86 + 0,65. E. coli contamination was found in meatballs stored at freezer storage. Treatment of different temperature storage was signiicantly affect water activity and was not effect on pH, water holding capacity and microbiological content (TPC and E. Coli).

Abstract

Dietary changes can increase the risk of degenerative diseases. The risk can be reduced by consuming healthy food, food that not only meets the needs of nutrition but also provide health benefits. Food that has both of these functions is called a functional food. Components that can be added into the food in order to have functional value is probiotic bacteria. Lactobacillus plantarum IIA-2C12 that isolated from local beef at Bogor was added in the process of  meat processing into fermented sausage and than tasted by in vivo. The aims of this study was to examine the influence of the probiotic fermented sausage using L. plantarum IIA2C12 as probiotica cultures on growth status of rats. This research was conducted in two stages, first bacterial starter culture refreshment and sausage making; second testing in rats (in vivo). The initial population of L.plantarum IIA-2C12 was 9. 27 log cfu mL  and after processing into fermented sausage bacteria became 8. 67 log cfu/g so that sausage could be categorized as a probiotic food. Feeding fermented sausages and sausages for 10 days had no significant effect (P>0.05) body and organ weights of rats. While the fermented sausage had highest feed convertion ratio than kasin and nonfermented sausage.

Abstract

It is well known that yoghurt is one of the fermented milk products. The addition of carbondioxide into yoghurt is an inovation for development of new product. The purpose of this researchs was to formulate and produce carbonated probiotic yoghurt also evaluate its functional properties. This research was divided into three step including formulation, evaluation of carbonated probiotic yoghurt during 18 days of refrigerated storage, and evaluation two best formulation for viability of BAL on gastric’s pH and the bile salt’s. The results showed that the addition of carbondioxide had no affect significantly on pH, total acid and the viabilities of lactic acid bacteria (LAB) during storage. On hedonic test, it affected significantly on smell, viscosity, and general appearance. Based on the results of organoleptic tests and performance during storage, formulas I and II were selected as two best formulas. On testing of two best formulas on resistance to acid condition, LAB from both formula’s were able to survive in the population about 8 log 10  cfu/mL. On testing resistance to bile salt, LAB from both formula’s were decreasing up to 5.5 log 10  cfu/mL. The loss occurred, the two formulas can still be considered as probiotic fermented milk.

Abstract

Red fruit pasta are reported containing active components that are important to health, so the opportunities to be developed into a functional food ingredients. The purpose of this study is utilize pasta as natural dye in beef sausage and based on chemical quality and sensory. Given some active compounds are known to display as natural dye for food.  In this study, beef sausage was added by red fruit pasta in various concentrations: (0%, 7%, 9%, and 11%) were added on beef sausage. The observed variables were chemical qualities (water content, protein content, fat content, ash content and carbohydrate content) and sensory (colour, taste, texture, and aroma). The result showed that the addition of red fruit pasta on beef sausage significantly affected on water content and protein content. Organoleptic testing (hedonic and hedonic quality testing) showed that the panelists remarkably prefered all beef sausages with addition of red fruit pasta

Abstract

This research aimed to study and evaluate the nutritional composition of sie reuboh treated with different concentrations of palm vinegar (Arenga pinnata) and kaffir lime leaves (Citrus hystrix) addition. The four treatments for sie reuboh were P0 (0 mL palm vinegar and 0 kaffir lime leaves), P1 (100 mL palm vinegar and 10 kaffir lime leaves, P2 (100 mL palm vinegar and 10 kaffir lime leaves). The parametes are nutrition content (water content, ash content, fat content, protein content and carbohydrates content) and organoleptic (hedonic qualiy test and hedonic test). The result showed that different concentrations of palm vinegar and kaffir lime leaves addition significantly affected (p<0.05) ash content and carbohydrates content of sie reuboh. Ash content in this study ranged from 3.11 % to 2.43 % and carbohydrates content ranged 3.89 % to 5.59 %. The result of organoleptic showed that were significantly affected (p<0.05) on hedonic quality test (flavor of palm vinegar, flavour kaffir lime leaves and taste). The value of palm vinegar flavor has 3.17 to 3.83, the value of kaffir lime leaves has 3.00 to 3.57 and the taste value has 3.30 to 3.77. Different level of palm vinegar and kaffir lime leaves not affected at moisture content, protein content, fat content, hedonic quality (color and tenderness) and hedonic (color, flavor, taste, tenderness and overall appearance).

Abstract

This study was conducted to evaluate physical, microbiological, and organoleptic quality of layer eggs marketed around IPB campus Darmaga, Bogor. Egg samples were obtained from three market groups (supermarket, retailer, and traditional market). Each group includes three markets determined purposively, namely SM-1, SM-2, and SM-3 for supermarkets; BR-1, BR-2, and BR-3 for retailers; and TM-1, TM-2, and TM-3 for traditional markets. Physical qualities measured were egg weight, yolk index, albumen index, air sac height, pH and Aw. Microbiological quality was expressed as colony forming unit with Total Plate Count (TPC) method and the number of Escherichia coli colony formed. Organoleptic properties evaluated were hedonic and hedonic ranking test. All physical and microbiological qualities of eggs showed no significant differences among markets origin. This study confirmed that eggs excluded from SNI 3926-2008 (Indonesia National Standard for consumption egg) and other scientific findings. However, the low physical and microbiological qualities did not cause any refusing on the acceptability of sensorial properties by panelists.
