Abstract

ABSTRACTPhalaenopsis orchid is one of famous genera in Orchidaceae family which have high economic value. Phalaenopsis breeding needs to be developed intensively to fulfill market demand for new varieties. The objective of this study was to obtain the information of  morphological and biochemical diversity of a collection of Phalaenopsis orchid which will be useful for Phalaenopsis breeding. Plant materials used were 10 genotypes of Phalaenopsis with various petal colours of white, yellow, pink, deep pink, purple, or combination of yellow and brown. Morphological diversity of 10 genotypes were analyzed based on qualitative and quantitative characters following UPOV guidance for Phalaenopsis, while biochemical diversity was based on pigment content such as chlorophyl, anthocyanin, and carotene in plant roots, leaves and flower petals. The result of this research showed morphological diversity of 10 genotypes used was relatively high, with similarity of 55%. In that level of similarity, genotypes were grouped into two cluster. Biochemical analysis showed that most of genotypes were different for anthocyanin and carotenoid content for each plant organ. Genotype which showed high content of anthocyanin in leaves was Phal. hybrid ‘PH37’ while high content of carotene in leaves was Phal. hybrid ‘PIROUI. High content of anthocyanin in petals were found in Phal. hybrid ‘PH37’ and Phal. esmeralda, and high content of carotene were in Phal. cornucervi and Phal. cornucervi Red. Root with high anthocyanin content was found in Phal. hybrid ‘PIROU1’. There were significant correlation (0.975) for anthocyanin content between leaves and petals, petals and roots (0.953) on Phal.hybrid ‘PH37’, and between leaves and roots (0.874) on Phal. hybrid ‘PIROU1).Keywords: anthocyanin, carotene, chlorophyll, Phalaenopsis, similarity

Abstract

ABSTRACTBambara groundnut (Vigna subterranea L. Verdc.) is an important underutilized legume crop in Indonesia. The aim of this research is to study genetic diversity of bambara groundnut from Sukabumi and Sumedang, West Java, Indonesia. This study used 107 bambara groundnut accessions, which consisted of 57 accessions from Sukabumi and 50 accessions from Sumedang. We use five simple sequence repeat (SSR) to analyze the accessions. Totally nine alleles were detected, with a mean of 1.8 alleles per locus. Allelic and gene diversities were higher in Sumedang (1.8 alleles per locus and 0.119) than in Sukabumi population (1.4 alleles per locus and 0.020), respectively. We constructed a phylogenic tree by Neighbor-Joining analysis based on genetic distances (DA) and showed the tree divided bambara groundnut accessions into two broad groups according to the origin of samples (Sukabumi and Sumedang). Results from the phylogenic tree are in line with those from the population structure analysis. Keywords: allele diversity, bambara groundnut, genetic distance, under utilized legume

Abstract

Bacterial soft rot disease because of Dickeya sp. infection is the main problem in Phalaenopsis production in Indonesia, but the percentage of infected plants has never been recorded in detail. Isolation and characterization of Pto gene from Phalaenopsis could be useful to support breeding for resistance Phalaenopsis. Encoding serine-threonine kinase, Pto gene confers resistance to bacterial infection of Pseudomonas syringae in tomato. The objectives of this study were to isolate, sequence and characterize fragment of Pto gene from 20 genotypes of Phalaenopsis (16 species and 4 hybrids) and to evaluate their molecular diversity. Genomic fragments of Phalaenopsis were amplified using Pto specific degenerate primers; and the PCR amplicons were sequenced. Searching the identity of determined sequences was done using BLAST against all accessions in NCBI GenBank DNA database and in Conserve Domain Database. PCR amplification using Pto specific primers produced a single DNA fragment of ~500 bp. The determined nucleotide sequences from the amplicon were ~449 bp. The nucleotide sequences of the amplicons from 20 Phalaenopsis genotypes showed high sequence identity to Pto from Musa acuminata. Translation of the amplicon results in ~149 amino acid residues. Comparison of the translated polypeptides identify indicated there were low variations of Pto gene among accessions since they contain the PTO catalytic domain and the Serine/Threonine kinases, sub family of Interleukin-1 Receptor Associated Kinase (STK_IRAK) which are the conserved domains for PTO.Keywords: catalytic domain, disease resistance, fragment Pto gene, phylogenetic, RGA

Abstract

ABSTRACT            Developing new rice  varieties adaptive to heat stress is important to maintain high rice production in anticipating global warming effects. This research was aimed to find selection characters and to select the best segregant in early generation based on agronomic characters for adaptation of rice to heat stress. The experiment was carried out in September 2014 to Januari 2015. About 210 F2 segregants generated trough hybridization between IPB 4S (sensitive parent) and Situ Patenggang (tolerant parent), 20 individuals of IPB 4S, and 20 individuals of Situ Patenggang, were used as genetic control. All genetic materials were exposed to high temperature stress by growing in a green house of Bogor Agricultural University. The average temperature in the research period was 24 °C and 42.09 °C, respectively for minimum and maximum temperature.  The results showed that charachters of productive and total tiller number, total grain number, and seed weight had high heritability, high coeffiecient of genetics variability-with an additive gene action. That charachters could be proposed as selection criteria in early generation for rice breeding of adaptation to high temperature stress. The characters could be applied  in either single trait or multiple traits selection. Based on multiple traits selection we found 50 the best F2 segregation.Keywords: differential selection, early generation, heat tolerant, heritability, gene action, variability genetics coeffiecient

Abstract

Endosperm quantity in kopyor coconut determines the quality of kopyor coconut. However, data of the best parental based on their progeny test is unavailable yet. In this study, we employed polycross approach and SSR molecular marker to estimate general combining ability of the best pollen contributor for the breeding purpose of this trait. Totally 151 kopyor coconuts were collected from the naturally open-pollinated population of kopyor Tall coconut Agom Lampung. The harvested nuts were rated for their endosperm quantity (scored 1-9). The analysis identified 31 paternal trees of the 53 progenies. The TJ-185 with the highest GCA has been considered as a promising paternal tree for development of high endosperm quantity of kopyor coconut.Keywords: combining ability, molecular marker, paternal trees, polycross, tall coconut
