Abstract

Latar Belakang: Ayam kampung merupakan salah satu kebutuhan protein hewani yang sering dicari masyarakat. Pemeliharaan ayam kampung dengan sistem dibiarkan lepas menjadi predisposisi terinfeksi oleh parasit gastrointestinal. Tujuan: Penelitian ini bertujuan mengkaji perubahan sel epitel dan hipertrofi sel goblet pada saluran pencernaan ayam kampung yang terinfeksi cacing cestoda Raillietina spp. secara alami. Metode: Penelitian ini menggunakan tujuh preparat arsip histopatologi usus halus ayam kampung yang terinfeksi cacing cestoda. Sampel usus halus ayam kampung diambil dari dua pasar yang berbeda, yaitu pasar Pluit di Jakarta Utara dan pasar Kebayoran Lama di Jakarta Selatan. Hasil: Hasil pengamatan histopatologis pada preparat usus ditemukan deskuamasi epitel vili dan proliferasi sel-sel kripta yang diakibatkan oleh infeksi cacing cestoda. Sel goblet yang mengalami hipertrofi paling banyak ditemukan pada sampel preparat yang diambil dari pasar Kebayoran Lama, Jakarta Selatan (p>0,05). Simpulan: Dapat disimpulkan bahwa deskuamasi vili dan jumlah hipertrofi sel goblet terjadi akibat infeksi cacing cestoda pada mukosa usus halus ayam kampung.

Abstract

Background Dieffenbachia seguine (Jacq.) Schott, commonly known as the dumb cane, is a tropical ornamental plant known for its toxic and proinflammatory properties. Its potential as a natural inducer of inflammation warrants further investigation, particularly via the intranasal route. Objective This study aimed to evaluate the proinflammatory effect of D. seguine leaf extract nanoemulsion by measuring the levels of interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α) in bronchoalveolar lavage fluid (BALF) following intranasal administration in rats. Methods Wistar rats (n= 24) were divided into six groups: water for injection (WFI, negative control); emulsion base; lipopolysaccharide (LPS, positive control); and D. seguine extract nanoemulsions at concentrations of 1%, 2%, and 4%. Each rat received an intranasal instillation, and IL-6 and TNF-α concentrations in the BALF were quantified using ELISA. Results Only the 4% D. seguine nanoemulsion group showed a significant increase in IL-6 and TNF-α levels compared to the control. Lower concentrations (1% and 2%) did not induce significant cytokine elevation. All treatment groups showed significantly lower cytokine levels than the LPS group. Conclusion D. seguine extract nanoemulsion at 4% concentration demonstrates potential as a natural inflammation-inducing agent via the intranasal route, although its effect remains lower than that of LPS.

Abstract

Bovine Herpesvirus (BoHV) is a member of Herpesviridae family that acts as pathogenic virus causing infectious bovine rhinotracheitis (IBR) among cattles, resulting in economic loss for cattle industry. BoHV-1 infection in cows is closely related to abortion, respiratory infection, reduced milk production, infertility, and low birth weight. The aim of this study was to identify and characterize the molecular of BoHV-1 and other virus types, as well as the possible presence of other Herpesviridae family using PCR to amplify DNA terminase gene. Four out of 210 nose swab samples were positive for herpes virus on DNA terminase gene. Further characterization of samples showed 99-100% similarity to BoHv-1 and BoHV-6 sequence. Genetic distance between genera BoHV-1 and BoHV-6 is 0.518 and within genera was 0.001 and 0.044. According to phylogenetic tree analysis of DNA terminase gene, the analyzed sequence clustered into 2 genera, namely Varicellovirus which is identical to BoHV-1 and Macavirus which is identical to BoHV-6. The study provides scientific information on molecular characteristics of Herpesviridae family, especially BoHV-1 which is prevalent in Indonesia with the highest density in the central ranches in Aceh province

Abstract

Kemiri (Aleurites moluccana L.) merupakan tanaman yang memiliki manfaat dari setiap bagiannya khususnya biji kemiri yang diolah untuk menghasilkan minyak. Penelitian ini bertujuan menghitung tingkat toksisitas minyak kemiri pada mencit dengan menggunakan penentuan nilai lethal dose 50 (LD50). Penelitian ini menggunakan rancangan percobaan acak lengkap. Sebanyak 20 ekor mencit betina dibagi menjadi lima kelompok yaitu kelompok kontrol negatif dan kelompok perlakuan yang diberikan minyak kemiri dosis 5, 10, 15, dan 20 g/kg BB secara peroral. Pengamatan gejala toksisitas pada mencit pascaperlakuan dilakukan selama 14 hari. Parameter yang diamati berupa mortalitas, gejala klinis, dan respon fisiologis mencit. Hasil pada pengamatan dengan parameter mortalitas hewan uji menunjukkan bahwa tidak ada mencit yang mengalami kematian sampai dengan dosis 20 g/kg BB. Parameter respon fisiologis seperti suhu tubuh dan frekuensi nafas berada di rentang normal, sedangkan frekuensi denyut jantung berada di bawah rentang normal. Kesimpulan penelitian ini yaitu minyak kemiri termasuk kedalam kategori praktis tidak toksik.

Abstract

African swine fever is a viral disease that causes hemorrhagic fever in domestic pigs and wild swine worldwide. The first case of ASF in Indonesia was reported in North Sumatra Province in September 2019. A total of 20 archival samples of positive ASF collected from the Animal Disease Investigation Center of Medan during the outbreak investigation took place from 2019 to 2023, were used in this study. The partial B646L gene (p72), the full sequence of the E183L gene (p54) and the central variable region (CVR) of the B602L gene were amplified, purified, then sequenced. Web-based BLAST program and MEGA XI software were used to analyze the nucleotide sequencing results. The results of molecular and phylogenetic characterization analysis revealed that the ASF virus that infected pigs in North Sumatra Province in 2019-2023 was belonged to genotype II. Analysis of the three genes (B646L, E183L and B602L) did not show changes in the nucleotide and amino acid sequences in the isolates in the last 5 years. No novel variants were found in the CVR gene B602L. CVR analysis showed that these ASF virus strain belonged to subgroup XXXII. The results of this study revealed that the ASF virus in North Sumatra Province has high homology with ASF virus isolates previously detected in China, Vietnam and East Leste in 2019.
