Abstract

A research was conducted to determine genetic variability and structure of ebony either within provenances or within trees in the same provenance using isozyme analyses. Results of this study are expected to show genetic variability of ebony with different provenances. For the purpose of the study, five provenances of ebony (Maros,Barru, Sidrap, Malili, and Mamuju) were prepared. The isozyme analysis using electrophoresis was applied to determine the genetic variability. The obtained data were statistically analyzed using multivariate and dendrogram analyses with Numerical Taxonomy System (NTSYS) Program applying unwighted pairgroup method and arrithmetic average (UPGMA) approach. Results showed that the variability of genetic provenances of ebony were less than any other tree species. Among the existing provenances, Barru dan Mamuju showed higher genetic variability compared to other provenances. Ebony provenance was generally found to homozygously increase or to performinbreeding. The genetic variability of ebony was mostly derived from the variability in population (95.4%). Grouping individual trees based on their provenances indicated that Malili provenance showed closed relationship to Maros provenance, whilst Mamuju provenance showed closed relationship to Barru and Sidrap provenances.

Abstract

Sengon (Paraserianthes falcataria (L) Nielsen) is a forestry plant often developed for industrial purposes in Indonesia. Sengon is a fast-growing species and has a relatively short-cutting period. Sengon wood is widely used in the production of carpentry wood, plywood raw materials, and pulp. The purpose of this study was to optimize the transformation of the entC and pmsB genes into sengon plants using the embryo injury method through Agrobacterium tumefaciens as an intermediary and to determine the efficiency of the transformation using the injury to sengon embryos. This research was conducted from September 2022 to March 2023. The research was conducted at the Genetically Modified Organism (GMO) Tissue Culture Laboratory, Recombinant Protein (Microbe) Laboratory, and Greenhouse of the Genetic Engineering Center, National Research and Innovation Agency (BRIN) Cibinong, Bogor, Java West. The research procedure consisted of several steps, namely Preparation of MS Media, Preparation of LB Media, Preparation of Sengon Seed Infection Using Agrobacterium tumefaciens, Sterilization and Planting of Sengon Seeds, Acclimatization, and Molecular Analysis (including Sengon DNA isolation, PCR Amplification, and Electrophoresis). Transformation efficiency was calculated by the positive strain of hpt PCR bands compared to the number of transformed embryos and multiplied by 100%. In the results of the PCR analysis, it was found that one plant sample carried the hpt marker, which indicates the integration of the entC and pmsB genes. which when analyzed using transformation efficiency calculations, the data obtained for the entire transformation stage was 0.8%, where the success of the transformation obtained was still classified as low. Factors that cause the low success of transformation include the method of transformation, the growth phase of bacteria, genetic material, and culture aseptic conditions.

Abstract

Pengabdian ini bertujuan untuk meningkatkan pengetahuan dan kemampuan pemasaran produk secara online bagi para petani dan peternak di desa Buntu Mondong, Kabupaten Enrekang, dengan menggunakan situs web integrasi WhatsApp. Kegiatan yang dilakukan terdiri dari: 1) Pelatihan penggunaan website e-commerce, 2) Sosialisasi Pengenalan Teknologi, 3) Persiapan dan Observasi, dan 4) Pemantauan dan penilaian program. Alat penilaian yang digunakan adalah observasi langsung terhadap proses pengenalan teknologi dan kuesioner pra dan pasca tes. Untuk mengukur pertumbuhan pengetahuan dan kemampuan, data dianalisis secara deskriptif. Hasil dari layanan ini menunjukkan peningkatan yang signifikan dalam pengetahuan dan kemampuan teknis petani dan peternak berdasarkan situs web integrasi aplikasi WhatsApp. Semua anggota mitra juga mengalami peningkatan dalam hal selain itu Mitra juga mengalami peningkatan  secara mendiri  mengelola websitenya melalui ponsel dalam menjual produknya.

Abstract

Diosphyros celebica Bakh., also known as Sulawesi ebony, is an endemic to central and northern Sulawesi. Information about pollen dispersal patterns of D. celebica have not been previously investigated. This study was aimed to determine pollination type, percentage of selfing and outcrossing, as well as distance of pollen dispersal of D. celebica. This study was conducted at experimental forest of Universitas Hasanuddin, Maros District, South Sulawesi Province, Indonesia. One hundred and sixty six individuals of D. celebica consisted of adult trees and seedlings were analyzed in this study. Ninety four adult trees were selected to become parent trees. The trees were mapped with GPS coordinates . All samples were genotyped using four SSR markers loci. Parental analysis and determination of pollen dispersal patterns were carried out using Cervus 3.0.3. Results indicated that the evaluated markers were effective for assigning candidate of male parents to all evaluated seedlings. Donated pollens could come from male parents in any directions relative to female parent positions. Pollen dispersal pattern showed outcrossing pollination among different male parents (pollen donated trees). The results indicated that seeds were produced predominatly by outcrossing. Pollen dispersal reached up to 166 m. Pollen related processes were linked through female parents, pollinators availability and ecological environment. Simultaneous use of progeny genotyping, spatially explicit analysis of environmental variables and outcomes of plant–animal interactions, were the key elements for an expanded approach to gene flow analysis considering dispersal via pollen and seeds. Research on pollen dispersal of D. celebica should be carried out in other forest types, such as mixed forest and highland forest.
