Abstract

1. Sekularisasi: Weber mengantisipasi bahwa spiritual capital itu perlahan-lahan makinmenyusut berkat terjadinya sekularisasi. Fondasi kapitalisme modern bukan lagi EPmelainkan kemajuan dan terobosan di bidang Iptek. Akibatnya profit dan wealthmenjadi tujuan mutlak yang terlepas dari panduan etis religious. Ringkasnya, bahayamunculnya materialism dan hedonism: motif kenikmatan duaniawi menggesermotiuf Protestant tentang keselamatan surgawi.2. Kapitalisme global dewasa ini bertumpu pada prinsip maksimalisasi produksi &maksimalisasi konsumsi. Akibatnya terjadilah krisis ekologis yang paarah.3. EP menandaskan bahwa sukses financial seseorang itu tergantung dari watak dankebajikan moral masing-masing individu ybs. Akibatnya cenderung ada pandanganyang negative terhadap orang-orang yang dianggap tak produktif (khususnyamiskin & menganggur): Mereka sering dianggap sebagai orang yang kurangmempunyai mental dan moral yang baik. Padahal mungkin mereka adalah korbanstructural.

Abstract

EKB meneliti proses kematian dari para pasien yang mengalami “natural death”kibat usia tua dan penyakit terminal. Akibatnya, uraiannya mungkin tidak seluruhnya berlaku bagi  orang yang mengalami “violent death,” yakni para korban yang mati mendadak sebagai korban kejahatan, kecelakaan atau serangan fatal seketika penyakit tertentu. Tambahan lagi, para pasien yang diamati EKB itu mendapat perawatan medis dan bimbingan psiko-spiritual saat menempuh perjalanan akhir hidupnya. Mungkin tidak setiap orang bisa mendapoat fasilitas  dan keberuntungan semacam itu. Kendati demikian, uraian EKB trentang tanda & tahap kematian itu tetaplah berguna sebagai bahan refleksi pribadi kita dan panduan bagi interaksi kita dengan orang-orang yang berada di ambang maut, agar  merejka bisa meninggakl dengan tenang dan bermartabat.EKB juga menandaskan bahwa kelima tahap itu berlaku dan berguna bukan saja bagi orang yang dihadang oleh krisis kematian, melankan juga bagi memahami setiap krisis kehidupan yang dialami manusia pada umumnya. Krisis adalah “lifechanging events,” saat dan peristiwa yang membawa perubahan yang besar dalam cara hidup kita, misalnya: cacat, lumpuh, perceraian, patah hati, drop-out, PHK, bankrut, menopause, pensiun dan pikun etc. Dinamika bertahap yang serupa, dengan intensitas yang berbeda, mungkin bisa terjadi saat kita mencoba untuk bangkit dari aneka krisis semacam itu.

Abstract

Rice stem borer (Scirpophaga sp.) is considered as one of the primary pests causing yield decrease in Indonesia. Genetic engineering is considered as one of the most effective way to improve the resistance of rice plants to yellow stem borer since no resistance gene has been found in rice and its wild relatives. A double T-DNA binary vector p2TDNAcryIAb wich carry two independent T-DNA was used to transform Indonesian elite rice cv Ciherang using Agrobacterium-mediated transformation to generate transgenic rice that free from selectable marker genes. One T-DNA containing a selectable marker hygromycin phosphotransferase (hpt) gene and the other carrying the cry1Ab gene. As many as 37 putative transgenic plants from 14 embryos were generated. PCR analysis demonstrated that 22 out of 37 putative transgenic rice contain cry1Ab gene. All Ciherang rice carrying the cry1Ab gene expressing Cry1Ab protein as shown by the immunostrip assay. Further work will be carried out to determine the transgenes copy number, to select marker free transgenic rice, and to evaluate the resistance of transgenic rice against neonates larvae of yellow stem borer.Keyword: double T-DNA, transgenic rice, Cry1Ab, transformation

Abstract

Transcription factors have an important function in regulating gene expression and plant responses to stresses. The ERF, bZIP, WRKY, MYB, and NAC are stress inducible transcription factors. The OsNAC6 is a member of the NAC transcription factor family in rice and its expression is induced by abiotic stresses, wounding and blast disease. Characterization of OsNAC6 gene sequences would give a better undersanding on how OsNAC gene functions biologically. The objectives of this research are to isolate the OsNAC6 cDNA from Nipponbare, Batutegi, and Rojolele cultivars, to characterize their DNA sequences, and to compare their sequences to other NAC genes from other plants available in GenBank DNA databases. Isolated cDNA and sequencing of the fragments resulted in a 912 bp DNA sequences. Translation of the sequences yielded a protein consisted of 303 amino acid residue. Blast analysis of amino acid sequences indicated identity of isolated cDNA from three Indonesian rice cultivars are the OsNAC6 gene. Deduced amino acid residues from amplified cDNAs of Nipponbare, Batutegi, and Rojolele cultivars shared 100% sequence identities to rice OsNAC6 (Acc. # BAA89800), 71-100% sequence identity to a number of OsNAC protein from Oryza sativa and 63-83% sequence identity to NAC protein from other plants.

Abstract

Liberibacter bacteria are one of the pathogenic bacteria in citrus plants. Siam Citrus Plant in Setapok Village, Singkawang City, suspected to be infected with Liberibacter spp. bacteria by showing patterns of symptoms of chlorosis accompanied by patches. This study aims to detect Liberibacter spp. on Siam Citrus leaves with symptomatic chlorosis with patches. Leaf sampling uses the roaming method in one of the citrus plantations in Setapok Village, Singkawang City. Detection of Liberibacter spp. done by PCR (Polymerase Chain Reaction) technique. Detection of bacterial DNA through several stages namely, isolation of bacterial plasmid DNA, 16S rDNA gene amplification with a thermocycler machine, and visualization using a set of electrophoresis devices. Electrophoresis results show the presence of DNA band lines, with a size of around 1100 bp for OI1 and OI2c primers, and DNA bands of 703 bp for A2 and J5 specifics. The results showed that there were Liberibacter spp. in the symptomatic chlorosis of Siam Citrus leaves accompanied by patches.

Abstract

Sengon (Paraserianthes falcataria (L) Nielsen) is a forestry plant often developed for industrial purposes in Indonesia. Sengon is a fast-growing species and has a relatively short-cutting period. Sengon wood is widely used in the production of carpentry wood, plywood raw materials, and pulp. The purpose of this study was to optimize the transformation of the entC and pmsB genes into sengon plants using the embryo injury method through Agrobacterium tumefaciens as an intermediary and to determine the efficiency of the transformation using the injury to sengon embryos. This research was conducted from September 2022 to March 2023. The research was conducted at the Genetically Modified Organism (GMO) Tissue Culture Laboratory, Recombinant Protein (Microbe) Laboratory, and Greenhouse of the Genetic Engineering Center, National Research and Innovation Agency (BRIN) Cibinong, Bogor, Java West. The research procedure consisted of several steps, namely Preparation of MS Media, Preparation of LB Media, Preparation of Sengon Seed Infection Using Agrobacterium tumefaciens, Sterilization and Planting of Sengon Seeds, Acclimatization, and Molecular Analysis (including Sengon DNA isolation, PCR Amplification, and Electrophoresis). Transformation efficiency was calculated by the positive strain of hpt PCR bands compared to the number of transformed embryos and multiplied by 100%. In the results of the PCR analysis, it was found that one plant sample carried the hpt marker, which indicates the integration of the entC and pmsB genes. which when analyzed using transformation efficiency calculations, the data obtained for the entire transformation stage was 0.8%, where the success of the transformation obtained was still classified as low. Factors that cause the low success of transformation include the method of transformation, the growth phase of bacteria, genetic material, and culture aseptic conditions.

Abstract

The increasing demand for clean water services in Majalengka Regency, especially in the strategic area of West Java International Airport (BIJB), necessitates optimal performance from PDAM Majalengka. This study aims to evaluate and optimize SPAM technology implementation to enhance water distribution services. Using quantitative analysis through surveys and statistical testing with SPSS, findings indicate that key factors such as water pressure, continuity, quality, pricing, and complaint handling significantly influence customer satisfaction. Recommendations include upgrading distribution networks, optimizing existing water sources, and adopting advanced SPAM technologies to meet future demands.

Historically, the original impulse of capitalism is to be the emancipatory social power to liberate mankind from the fetter of Religious Leviathan reigning in the dogmatic Medieval Era. Ironically, after its historical global triumph, it becomes an Economic Leviathan of its own. Abstract

Driven by his method of correlation, Paul Tillich critically analyzes the social condition of modern society which is characterized by the growing hegemony of capitalism. Capitalism is viewed not simply as a certain economic arrangement of society but as the embodiment of the religion of reason, namely, the historical project of humanity to realize its utopian goals and longings by the power of science and technology alone. However, by forgetting the awareness of the inherent finitude of reason cultivated by a genuine religious consciousness, mankind is threatened to lose sight of the demonic dimension built-in structurally in the capitalist mode of wealth creation.

Abstract

This article investigates the deeper foundation for the rapidly emerging and widespread consciousness in the business world on tbe nature and demand of the corporate social responsibility. Ethical norms and values should no longer be considered as externalistics, annoying additional costs and burdens to business interprices, but the very inner motivation for doing business at all, namely, to bring betterment to the world-wide human condition. That enduring noble al could only be accomplished if there is a comprehensive understanding on the kind of rationality that should permeate the so-called market society. In addition to the instrumental rationality that presently makes the market fluorish, we should also integrate what Max Weber called affective rationality and normative rationality.
