Abstract

The aims of Genetic study of pinus identified stand in Unhas Experimental Forest is to analyses of genetic characteristics of stand, based on RAPD (Random Amplified Polymorphic DNA) marker. The study was conducted in Biotechnology and Tree Breeding Laboratory, Forestry Faculty, Hasanuddin University. The method are DNA isolation, Primers selection and RAPD analyses. DNA analyses of Pine with ten RAPD marker showed number of alel varietied and polymorphic. Coefficient of similarity in population have number of 0.15-0.73.Highest genetic distance is 0.9630 and lowest genetic distance is 0.2698. Number of genetic diversity of Pine in Experimental Forest Hasanuddin University is 0,489 and categorized highly.

Abstract

Apart from being a key to identifying wood, the characteristics of the anatomical structure of cell walls can be used as a reference in determining the quality of wood, especially for its use. However, the characteristics of wood in the same species are sometimes found to be different due to the growing place is unidentical. Therefore, this study aimed to identify the cell wall characteristics of Jabon Merah wood from Wajo provenance. The wood was collected from Tangkoro Village, Pitumpanua District, Wajo Regency, South Sulawesi. It was then prepared in slicing and maceration samples. The structures of cell walls observed in the preparations were a pit, perforation plate, and tylosis. The results showed the Jabon Merah had inter vessel pit and perforation plate types, namely polygonal alternating and scalariform, respectively. Also, it had a tylosis in the vessel.

Abstract

This research aimed to identify the mycorrhiza types that associated in roots and soil on  Palaquium sp. plant. This  research has implemented on December 2017 until February 2018. This research procedures made by two stages includes collecting soil and root samples on Palaquium sp plant on  Ko'mara Community Forest, Takalar District. Therefore, the second step is isolation, identification, and observing colonization of mycorrhiza spores in the Integrated Laboratory and Biotechnology and Tree Breeding Laboratory, Faculty of Forestry, Hasanuddin University, Makassar. The results showed types of spore that found were Glomus, Gigaspora and Acaulospora. Glomus spore has highest relative abundance in root samples whereas on soil samples were Glomus, Gigaspora, and Schleroderma with Glomus spores has highest relative abundance in. The colonization percentage on site was intermediate.

Abstract

Kegiatan pembesaran ikan kakap putih di beberapa Kabupaten di Sulawesi Selatan yang menggunakan benih yang diproduksi dari balai perikanan di Sulawesi Selatan dan Ambon telah berkembang selama beberapa tahun terakhir. Akan tetapi, benih yang dihasilkan dari kegiatan pembenihan tersebut memiliki kualitas yang masih rendah yang diduga karena keragaman genetik yang rendah. Penelitian ini menggunakan metode Random Amplified Polymorphic DNA, RAPD untuk menganalisis variasi genetik ikan kakap putih tipe liar dari Bulungan Kalimantan Utara dan ikan hasil domestikasi dari panti pembenihan di Ambon yang dibesarkan di tambak rakyat di Desa Borongkalukua Kabupaten Maros Sulawesi Selatan. Hasil penelitian menunjukkan bahwa ikan kakap putih tipe liar memiliki nilai heterozigositas yang lebih tinggi dibandingkan dengan ikan kakap putih hasil domestikasi. Jarak genetik antar individu-individu di dalam populasi tipe liar lebih tinggi dibandingkan dengan jarak genetik individu-individu hasil domestikasi. Dendogram dan pohon filogenetik yang dibentuk berdasarkan evolutionary dissimilarity menunjukkan bahwa pada tingkat ketidakmiripan 10% ikan kakap putih tipe liar dari Bulungan Kalimantan Timur membentuk klaster yang terpisah dengan ikan kakap putih hasil domestikasi dari Ambon yang dipelihara di tambak Dusun Borongkalukua Kabupaten Maros. Keragaman genetik ikan hasil domestikasi lebih rendah dibandingkan ikan tipe liar dan menunjukkan pola evolusi yang pholyphyly Hasil yang diperoleh tersebut menunjukkan bahwa marka RAPD dapat digunakan untuk menganalisis perubahan keragaman genetik antara populasi ikan kakap putih tipe liar dan hasil domestikasi serta bermanfaat sebagai informasi dalam menyusun strategi pemanfaatan wild population sebagai sumber induk dan konservasi plasma nutfah ikan kakap putih.

Abstract

Soil fungi, which produce hormones that occupy the rhizosphere ecosystem, have roles as agents to promote plant growth. This study aims to isolate rhizosphere fungi in mahogany in the Experiment Forest of Universitas Hasanuddin, and identify the fungal isolates obtained based on morphological characters. The research began with rhizosphere soil sampling, isolation, and identification based on morphological characters. The rhizosphere microbial isolation on mahogany stands in the Unhas Educational Forest obtained 18 fungal isolates with varied morphological characteristics, both in colony color and elevation. Microscopic observations resulted in 4 genera, namely Rhizopus, Gliocladium, Trichoderma, and Aspergillus. The results showed that there were superior fungi isolates that could produce growth hormones and could be applied to forest plant seeds. The formula with these select fungi can give the seeds optimal growth in forest plant seeds.

Abstract

Bitti (Vitex cofassus) is one of the most important tree species in Sulawesi, which is used as material for phinisi boats. The aim study was to study the genetic diversity of Bitti in the genetic resources area in Bone Regency. This study used provenance Bulukumba, Muna, Kolaka, and North Luwu proof which was analyzed based on microsatellite markers. The resulting study showed evidence that provenance Muna has a Ho value 0f 0,65 or a relatively high genetic diversity value in comparison to provenance Bulukumba, Kolaka, and North Luwu.

Abstract

Parentage analysis has been used to evaluate pollen dispersal in ebony (Diosphyros celebica Bakh.). The objectives of this research were to evaluate (i) the dispersal of pollen, and (ii) the distance of pollen travel in ebony Lasitae provenances. The finding of this activities should be beneficial to support breeding of this ebony tree. There were 14 progeny arrays were harvested from 3 female parents. There were 32 ebony trees surrounding the female parents were analyses as the potential male parents for the progenies. Ebony tress were mapped according to their GPS position. All samples were genotyped using four SSR marker loci. Parentage analysis was done using CERVUS version 2.0 software. Results of the analysis indicated the evaluated markers were effective for assigning candidate male parents to all evaluated seedlings. There is no specific direction of donated pollen movement from assigned donor parents to the female ones. The donated pollens could come from assigned male parents in any directions relative to the female parent positions. Pollen dispersal pattern of ebony female parent occured in outcrossing pollination among its different donated trees. Based on the progenies analysis, ebony female parent is a dominantly outcrossing pollination species. Distances of pollen travel reach to 18 until 269 meter.Key words: Ebony, ssr marker, pollen dispersal.

Abstract

Diosphyros celebica Bakh., also known as Sulawesi ebony, is an endemic to central and northern Sulawesi. Information about pollen dispersal patterns of D. celebica have not been previously investigated. This study was aimed to determine pollination type, percentage of selfing and outcrossing, as well as distance of pollen dispersal of D. celebica. This study was conducted at experimental forest of Universitas Hasanuddin, Maros District, South Sulawesi Province, Indonesia. One hundred and sixty six individuals of D. celebica consisted of adult trees and seedlings were analyzed in this study. Ninety four adult trees were selected to become parent trees. The trees were mapped with GPS coordinates . All samples were genotyped using four SSR markers loci. Parental analysis and determination of pollen dispersal patterns were carried out using Cervus 3.0.3. Results indicated that the evaluated markers were effective for assigning candidate of male parents to all evaluated seedlings. Donated pollens could come from male parents in any directions relative to female parent positions. Pollen dispersal pattern showed outcrossing pollination among different male parents (pollen donated trees). The results indicated that seeds were produced predominatly by outcrossing. Pollen dispersal reached up to 166 m. Pollen related processes were linked through female parents, pollinators availability and ecological environment. Simultaneous use of progeny genotyping, spatially explicit analysis of environmental variables and outcomes of plant–animal interactions, were the key elements for an expanded approach to gene flow analysis considering dispersal via pollen and seeds. Research on pollen dispersal of D. celebica should be carried out in other forest types, such as mixed forest and highland forest.
