Abstract

This research was conducted to determine the potential of phytochemical contents in producing antibacterial activity from 70% ethanol extract of Balik Angin leaves  (Alphitonia incana (Roxb.) Teijsm . & Binn. ex Kurz) against Escherichia coli bacteria in silico and in vitro. In silico assay of 20 compounds contained in Alphitonia species against the DNA gyrase receptor using PLANTs. Anti-E.coli activity assay of Balik Angin leaves macerated with 70% ethanol solvent using the Well diffusion method. In silico studies show that β-Sitosterol, Stigmasterol, Rutin, Kaempferol 3-O-rutinoside, and Zizyphoiside C have binding energies with DNA gyrase that can exceed Ciprofloxacin. The antibacterial assay results at a highest concentration of 26.56 mg/ml can provide an average inhibitory zone diameter of 12.75 ± 0.2 mm with a minimum inhibitory concentration (MIC) of 1.66 mg/ml (5.87 ± 1.5 mm). The 70% ethanol extract of Balik Angin leaves has the potential as an anti-Escherichia coli, supported by the content of phytochemical compounds that can act as DNA gyrase inhibitors.

Abstract

Abstract

Background: Acne is a skin infection that may be induced by Propionibacterium acnes. Treatment of acne infection from natural ingredients native to Kalimantan such as Balik Angin or Alphitonia incana (Roxb.) Teijsm. & Binn. ex Kurz can be an alternative therapy. Prior research indicates that leaves macerated with methanol and 70% ethanol solvents can inhibit the growth of P. acnes in the medium category with a Minimum Inhibitory Concentration (MIC) of 3.2%. Objectives: The research purpose is to compare the effect of the different solvents of soxhlet extraction on the antibacterial activity of Balik Angin leaves in inhibiting the growth of P. acnes. Material and Methods: Balik Angin leaves were extracted using the Soxhlet with two different solvents, which were methanol and 70% ethanol, then followed by phytochemical screening. The antibacterial activity assay was carried out by using well diffusion with each sample concentrations of 25.6%; 12.8%; 6.4%; 3.2%; 1.6%; 0.8%; 0.4%; and 0.2%. Results: Both extracts contained the same phytochemical groups, which were phenols, flavonoids, alkaloids, tannins, saponins, and triterpenoids. The antibacterial activity test results showed that both extracts had the same MIC of 0.8%. however, the 70% ethanol extract of Balik Angin leaves was able to provide a larger inhibition diameter average of 9.61±0.35 mm compared to the methanol extract of 9.20±0.22 mm. Nonetheless, there was no significant difference (p>0.05) between both concentrations, when compared to the range of 1.6-25.6%. Conclusions: The conclusion of this research is that differences in Soxhlet extraction solvents in Balik Angin leaves can affect its activity as an anti-Propionibacterium acnes agent.

Abstract

Kelakai root contains alkaloids, saponins, tannins, and flavonoids. This compound is known to have medicinal properties such as wound healing. The effectiveness of wound healing increased by using hydrogel to deliver the active substance. The use of polymers played a role in delivering the active substance. This study aimed to formulate hydrogels for wound healing with various concentrations of polyvinyl alcohol (PVA) and hydroxy propyl methyl cellulose (HPMC) K4M. Preparation of hydrogel by the solvent casting method used 0.2% Kelakai root extract. The physical characteristics of hydrogel investigated before and after the stability test included organoleptic, weight uniformity, thickness, pH, moisture content, and hedonic test of Kelakai roots extract hydrogel.  The optimum formula was tested for wound healing activity. The data obtained were then analyzed using Wilcoxon testswith p0.05 considered significant. Physically, Kelakai root extract hydrogel met the test requirements. The results of the wound-healing activity showed that the hydrogel extract of the Kelakai root was able to heal the wounds within seven days. There was a significant difference in wound healing time with p=0.014. The optimum formula obtained based on the stability test was F4 with a 1:4 ratio of PVA and HPMC.
