Abstract

In this study of teratogenic toxicity tests was carried out on the ethanol extract of tamarindus leaves (Tamarindus Indica Linn) on rats (Rattus norvegicus) during the organogenesis period. This study aims to know the safety and toxicity of the administration of ethanol extract of tamarind leaves (Tamarindus indica L.) to fetus rats (Rattus norvegicus) with macroscopic parameters including number, resorption, weight, and skeleton of the fetus. 15 animals were divided into 3 groups, each group consisted of 5 rats. Group I was treated with 0.5% Na CMC as normal control,  group II was treated with ethanol extract of tamarind leaves with a dose of 14 mg / 200 g BW for 10 days and group III was given a simvastatin tab at a dose of 10 mg / kg BW. Treatment was given orally during the organogenesis period on the 5th to 15th day of pregnancy. On the 16th day, laparatomy of the rat and fetus was removed from the uterus. The results showed that the suspension of ethanolic extract of tamarind leaves did not cause teratogenic occurrence in fetal rats, and the control of simvastatin caused teratogenic due to resorption, less fetal weight, hand and foot defect, and death in the fetus, compared to the normal control group.

Abstract

Burns are damage or loss of skin tissue caused by contact with heat sources such as hot water, fire, chemicals, electricity, friction and radiation. Sea lotus (Strychnos ligustrina Blume) is known to contain compounds that have the potential to heal burns such as flavonoids, saponins and tannins. This study aims to determine the activity of ethanol extract of bidara de mer leaves on burn healing as a function of reduction in burn diameter, wound discoloration, edema and crust formation. This study used 5 Oryctolagus cuniculus that received 4 types of treatment after being induced by hot plate, namely wound I (bioplacenton® gel positive control), wound 2 (HPMC 5% negative control), wound 3 (extract concentration 10%) and wound 4 (concentration of extract 10%) 25%). Apply the test sample to the burns twice a day and observe the condition of the wound daily. Burn healing as a function of wound diameter was determined based on the AUC (Area Under Curve) value and statistically analyzed using SPSS 21.0. The test results showed that the mean AUC value at 10% and 25% concentrations was 0.495 cm/day and 0.532 cm/day, respectively. The results of the statistical analysis showed that the ethanol extract of bidara sea leaves gave significant results compared to the negative control. treatment (HPMC). The conclusion obtained is that the ethanolic extract of sea bidara leaves accelerates the healing process of burns in Oryctolagus cuniculus.

Abstract

Morus alba dikenal sebagai sumber senyawa antioksidan alami yang dapat menangkal radikal bebas yang berkontribusi terhadap penyakit degeneratif. Penelitian ini bertujuan untuk mengidentifikasi aktivitas antioksidan dari ekstrak metanol dan fraksi daun M.alba menggunakan metode BCB, CUPRAC, dan FRAP. Ekstraksi dilakukan menggunakan pelarut metanol, kemudian difraksinasi dengan n-heksana, etil asetat, n-butanol, dan air. Metode BCB digunakan untuk menentukan IC50, metode CUPRAC untuk mengukur kapasitas antioksidan dalam GAEAC (Gallic Acid Equivalent Antioxidant Capacity), dan metode FRAP untuk menilai kemampuan reduksi ion besi dalam FeEAC (Ferri Sulfate Equivalent Antioxidant Capacity). Pengujian aktivitas antioksidan pada metode FRAP dilakukan dengan reagen dapar asetat, TPTZ, dan FeCl? yang diukur menggunakan microplate reader pada panjang gelombang 595 nm berdasarkan kemampuan sampel dalam mereduksi ion besi. Metode CUPRAC menggunakan reagen CuCl? 10 mM, neocuproine 7,5 mM, dan ammonium sulfat 1 M. Hasil uji BCB menunjukkan bahwa fraksi n-heksana memiliki aktivitas antioksidan tertinggi dengan IC50 sebesar 64,85 ppm dibandingkan dengan etil asetat (437,23 ppm). Pada CUPRAC, ekstrak metanol menunjukkan kapasitas tertinggi (342,74 µM/g), diikuti oleh etil asetat (342,43 µM/g). Uji FRAP menunjukkan fraksi etil asetat memiliki aktivitas tertinggi (809,28 mmol/g), diikuti oleh metanol (761,03 mmol/g), dan n-butanol (613,03 mmol/g). Ekstrak metanol, fraksi etil asetat, dan n-butanol menunjukkan kemampuan signifikan dalam mereduksi ion Cu²? dan Fe³? dibandingkan dengan n-heksana dan air. Hasil ini menunjukkan bahwa aktivitas antioksidan ekstrak M. alba bervariasi tergantung pada fraksi dan metode yang digunakan. Oleh karena itu, pemilihan metode yang tepat sangat penting dalam mengevaluasi potensi antioksidan dari suatu sampel.

Abstract

Simvastatin is a drug acting on 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase enzyme leading to decrease of lipid level in plasma. Simvastatin is associated with pleiotropic effects such as cardioprotective, hepatoprotective, and nephroprotective effect. This study aimed to observe effect of supplementation of trigona honey on parameters of liver function (SGPT and SGOT) and kidney function (urea) in albino rats (Rattus norvegicus) given 40 mg/kg simvastatin. Twenty-four male albino rats were divided into 6 groups (n=4). Each group was administered different treatments for 15 days orally. Group I was put as health control without any treatment, group II was given sodium carboxymethylcellulose (1% b/v) as negative control, group III was given simvastatin at the dose of 40 mg/kg, group IV was administered simvastatin (40 mg/kg) and trigona honey (6.5% v/v), while group V and VI were administered simvastatin (40 mg/kg) and ubiquinone (1.43 mg/kg); and simvastatin (40 mg/kg), trigona honey (6.5% v/v), and ubiquinone (1.43 mg/kg), respectively. Upon the treatments, level of SGOT, SGPT, and ureum was determined. The data were analyzed by using Analysis of Variance (ANOVA) and Least Significant Difference tests (p=0.05). According to the analysis, it was concluded that supplementation of trigona honey in rats administered simvastatin showed significantly lower level of all parameters than groups of simvastatin and controls.
