Abstract

Abstract

This study was conducted to see the effect of growth and antioxidant activity of microalgae Oocystis sp before and after adding NaOCl to Growmore medium combined with urea. Antioxidant activity test was conducted by calculating IC50 spectrophotometrically with the 1,1-diphenyl-2-picrylhydrazyl (DPPH) method. Oocystis sp extract was obtained by sonication and maceration process with methanol solvent. The results showed the best growth curve for the growth of microalgae Oocystis sp in a mixture of Growmore medium 0.02% b/v and Urea 0.002% b/v. These results also showed a decrease in Oocystis sp growth in a mixture of Growmore medium 0.02% b/v and Urea 0.002% b/v with the lowest growth decrease in the addition of NaOCl with a concentration of 0.01%. The IC50 value of Oocystis sp extract in methanol solvent has high antioxidant activity in Growmore 0.02% b/v + Urea 0.002% b/v medium, which is 55.5862 mg/L. It can be concluded that NaOCl affects the growth and antioxidant activity of microalgae Oocystis sp, but at certain concentrations NaOCl experienced a negative effect. Keywords: Microalgae, Oocytis sp, Oxidative Stress, Growth, Antioxidant

Abstract

Abstract

The Environmental pollution caused by industrial waste including oil spills have become a global issue that requires effective and environmentally friendly solutions. Bioremediation used by bacteria immobilized has been develop as a promising method for pollutant degradation, because it can increase the stability and activity of microorganism under various condition in the environment. This study is a systematic review to evaluate various immobilized technique including adsorption, entrapment, adsorption-embedding, cross linking and the techniques effectiveness in hydrocarbon, crude oil, and heavy metals remediation. This review collates a vast amount of existing literature on the myriad contaminants treated using immobilized bacteria. Based on the reviewed article, the immobilization techniques such as adsorption, adsorption-embedding, entrapment and cross-linking were frequently reported to enhance degradation performance, particularly in crude oil bioremediation. The techniques consistently demonstrated high effectiveness in pollutant degradation across different environmental conditions. The environmental factors, including pH, pollutant concentration and surfactant availability have an important role in the success of bioremediation. Although, this technology enhances bacterial resilience and biodegradation efficiency, the challenges such as the hight cost of immobilization materials and limitations in extreme environment application remain a problem. The optimization techniques and immobilized materials has the potential to provide a sustainable solution for pollutant bioremediation in the environment

Abstract

Organic fertilizers are widely used in agriculture as an environmentally friendly alternative to improve soil fertility and crop productivity. However, their safety remains a concern due to the potential contamination by pathogenic bacteria, particularly Escherichia coli and Salmonella sp., which may pose risks to human health. This study aimed to analyze the presence of E. coli and Salmonella sp. in solid organic fertilizer samples based on the requirements of the Indonesian National Standard (SNI 7763:2024). Laboratory analyses were conducted at the Balai Standardisasi dan Pelayanan Jasa Industri (BSPJI) Palembang using standard microbiological methods, including presumptive and confirmatory tests, followed by Most Probable Number (MPN) estimation. The results confirmed the presence of E. coli colonies with a characteristic metallic green sheen on Eosin Methylene Blue Agar (EMBA), with an estimated concentration of 30 MPN/g, still below the SNI threshold of 100 MPN/g. In contrast, Salmonella sp. was not detected in the samples, as shown by negative growth on Salmonella Shigella Agar (SSA), with an estimated value of <3.0 MPN/g. These findings indicate that the tested organic fertilizer samples comply with microbiological safety standards, suggesting that the production and handling processes were adequate to minimize pathogenic contamination. Continuous monitoring and strict adherence to hygienic practices are recommended to ensure the consistent quality and safety of organic fertilizers in agricultural applications.
