Abstract

Cucumber mosaic virus (CMV) merupakan patogen penting pada Cucurbitaceae, diantaranya melon. Pengelolaan virus di lapangan cukup sulit dilakukan. Salah satu upaya untuk menekan infeksi virus yaitu melalui penyehatan tanaman menggunakan bakteri agens hayati. Penelitian ini bertujuan menguji potensi isolat bakteri Bacillus cereus (BC), Chromobacterium haemolyticum (CH), Ralstonia pickettii (RP), dan Pseudomonas flourescens (PF) dalam menekan infeksi CMV pada tanaman melon. Pengujian pengaruh bakteri terhadap performa kecambah dilakukan melalui perlakuan perendaman benih dalam suspense bakteri di laboratorium.  Efikasi bakteri dalam menekan infeksi CMV dilakukan dengan cara perendaman benih dan penyemprotan tanaman dengan suspense bakteri dalam percobaan di rumah kaca. Pengamatan dilakukan terhadap peubah penyakit dan agronomi, dan aktivitas enzim peroksidase. Hasil penelitian menunjukkan perlakuan bakteri BC menunjukkan performa kecambah terbaik di antara perlakuan bakteri lainnya. Perlakuan bakteri tidak berpengaruh terhadap masa inkubasi dan insidensi penyakit, namun secara nyata menurunkan keparahan penyakit dan akumulasi virus dibandingkan kontrol. Aktivitas enzim peroksidase lebih tinggi setelah perlakuan bakteri dan inokulasi CMV dibandingkan perlakuan sebelum inokulasi. Bakteri RP merupakan bakteri terbaik dalam menekan infeksi CMV melalui aktivitas antivirus dan induksi ketahanan dengan peningkatan aktivitas peroksidase.

Abstract

This study aimed to evaluate the effect of chitosan-based liquid organic fertilizer (LOF) at various concentrations on the growth of cocoa seedlings (Theobroma cacao L.). The research was conducted at PTPN I Regional 5, Kendenglembu Plantation, Banyuwangi, for four months using a Completely Randomized Block Design (RCBD) with five treatments: P0 (control), P1 (10 mL/L), P2 (20 mL/L), P3 (30 mL/L), and P4 (40 mL/L). Growth parameters observed included plant height, stem diameter, and number of leaves up to 12 Weeks After Planting (WAP). ANOVA analysis showed no statistically significant effect of the treatments on any parameter. However, descriptive data indicated that concentrations of 20–30 mL/L tended to produce better growth: P2 (31.25 cm) had the highest plant height, P3 (6.93 mm) the highest stem diameter, and P1 (12 leaves) the most leaves. Chitosan potentially acts as a plant growth stimulator by increasing nitrogen availability and enhancing physiological processes such as photosynthesis. In contrast, a high concentration (40 mL/L) exhibited inhibitory effects. Although the differences were not statistically significant, concentrations of 20–30 mL/L could be recommended for practical application. The lack of significant results may be attributed to suboptimal concentrations, application frequency, or environmental conditions. Further studies are recommended to modify treatments and assess additional parameters such as biomass, root development, and leaf area to better understand the efficacy of chitosan fertilizer in cocoa seedling growth.

Abstract

Termitarium Macrotermes gilvus Hagen. merupakan ekosistem kompleks yang kaya akan mikroorganisme dengan potensi bioaktivitas tinggi. Penelitian ini bertujuan mengisolasi, mengkarakterisasi morfologi koloni, dan mengevaluasi potensi bakteri yang diisolasi dari termitarium M. gilvus Hagen. sebagai agens hayati. Sampel termitarium yang telah dikumpulkan, kemudian dilakukan isolasi bakteri melalui pengenceran berseri pada media Nutrient Agar dan media selektif aktinomiset yaitu water yeast extract agar dan casamino acid yeast extract glucose agar. Karakterisasi morfologi dilakukan melalui pengamatan koloni. Evaluasi potensi sebagai agens hayati dilakukan melalui uji Gram dengan KOH3% dan uji hipersensitif. Hasil penelitian menunjukkan bahwa dari termitarium M. gilvus Hagen. berhasil diisolasi 29 isolat bakteri dengan karakteristik morfologi koloni yang beragam. Karakterisasi morfologi koloni menunjukkan variasi bentuk, elevasi, tepian, dan warna. Jumlah koloni isolat bakteri yang dapat diisolasi dan ditumbuhkan pada medium selektif aktinomiset sebanyak 12, sedangkan pada medium NA yang umum digunakan untuk mengulturkan bakteri diperolah 17 isolat. Sebelas isolat diantaranya diisolasi dari tanah sarang rayap (TS), sementara lainnya diperoleh dari fungsu comb (F/sarang rayap). Bentuk koloni bakteri yang dominan ditemukan yaitu bundar (10 isolat). Karakteristik elevasi dan tepian koloni bakteri yang paling banyak diperoleh masing-masing yaitu cembung dan siliat. Warna koloni paling dominan yaitu putih keruh sebanyak 14 isolat. Hasil pengujian jenis Gram menunjukkan bahwa 16 isolat merupakan Gram positif, dan seluruhnya bersifat negatif pada pengujian reaksi hipersensitif. Hal tersebut menunjukkan bahwa setidaknya sebanyak 16 isolat bakteri yang diperoleh potensial untuk diuji lebih lanjut sebagai agens hayati yang diharapkan dapat menjadi alternatif pengendalian OPT yang efektif dan lebih ramah lingkungan.

Abstract

This study evaluates the effectiveness of agricultural extension activities aimed at enhancing the knowledge of farmers of composting and good agricultural practices for avocados in Mulyaharja village, Bogor, West Java Province, conducted in October 2024. A total of 10 participants completed pre- and post-tests designed to measure their understanding of avocado cultivation and composting practices. Results showed a significant increase in knowledge regarding avocado cultivation, with scores rising from 64% to 79% post-intervention (P=0.049). However, the improvement in composting knowledge was minimal, increasing from 57% to 63%, which was not statistically significant (P=0.29). These findings indicate that targeted extension activities can effectively improve knowledge related to avocado farming while suggesting a need for more engaging methods to enhance composting knowledge. This research highlights the critical role of tailored extension services in promoting sustainable agricultural practices and enhancing the capabilities of farmers in the region.

Abstract

Horticultural extension services in Bogor are instrumental in promoting agricultural development by enhancing the productivity and sustainability of local horticultural practices. This study emphasizes the empowerment of women farmer groups, known as Kelompok Wanita Tani (KWT), to improve horticultural production through targeted initiatives such as providing essential production facilities and enhancing the skills of farmers and extension officers. Data collection was conducted from July to September 2024, using questionnaires and surveys to assess the knowledge of KWT members before and after extension activities. The results indicated a significant increase in knowledge related to horticultural crop cultivation, with pre-test scores rising from 69% to 87% post-training, demonstrating a 17% improvement (P0.01). This improvement in knowledge and practices is a promising sign for the future of horticultural development. Correlation analysis revealed interesting relationships among agricultural knowledge, educational background, age, and agricultural experience. This research highlights the crucial role of extension services in disseminating knowledge and best practices, ultimately improving the livelihoods of farmers in Bogor and contributing to the local economy.
