Abstract

Children under 5 years of age are particularly vulnerable to malaria. Malaria has caused 445,000 deaths worldwide. Currently, rapid diagnostic tests (RDTs) are the fastest method to diagnose malaria. However, there are limitations that exist such as low sensitivity in detecting infections with low parasitemia. Practical, non-invasive and high ability tests to detect parasite are needed to find specific biomarkers for P. falciparum infection to determine the potential of P. falciparum 4 thioether in breathomics analysis by GC-MS as a practical non-invasive method in diagnosing malaria in pediatrics. Literature reviews from Google Scholar and ProQuest were published no later than the last 5 years. The concept of breathomics is that the breath's volatile organic compounds (VOCs) profile is altered when the health condition changes. Breath samples from individuals infected with P. falciparum malaria were taken by exhalation. Through GC-MS analysis, it was found that 4 thioether compounds (allyl methyl sulfide (AMS), 1-methylthio-propane, (Z) -1-methylthio-1-propene and (E) -1-methylthio-1-propene) underwent a significant change in concentration during the infection. Based on experiments conducted on mice and humans, the breathomics method is known to be able to detect parasitemia levels up to <100 parasites/µL, has a sensitivity level of about 71% to 91% and a specificity of about 75% to 94%. The discovery of 4 thioether compounds by GC-MS is a strong indication of malaria, because it has the potential for high sensitivity and specificity, and the detection power exceeds the ability of RDTs.

Abstract

Background Nematode infections result in 60.000 deaths each year. The majority of these infections are dominated by Ascaris Lumbricoides, which is asymptomatic and is currently treated with albendazole. However, prolonged use increases the risk of drug resistance. Research on herbal anthelmintics is very much needed. Herbs like C. asiatica and M. pudica (a wild tropical plant), have proven to possess antiparasitic properties due to their anthelmintic content. Methods This research used in-vitro experiments to compare the effectiveness of M. pudica and C. asiatica herbal extracts and their concentrations on the death time of A. suum worms. Herbal extracts were obtained through remaceration techniques, and each herb was made in concentrations of 10%, 20%, and 50%. Each experimental group was given 5 A. suum worms, which were observed every hour for 24 hours. Results A total of 53.3%, 86.6%, and 93.3% of the worms died in the C. asiatica group using 10%, 20%, and 50% concentration. Meanwhile,  60%, 73.3%, and 100% of the worms died in the M. pudica group using the same concentrations. By using two-way anova to compare the effectiveness of each concentration and also to compare the two plants, it could be concluded that there was no significant difference between C. asiatica and M. pudica either in their respective concentrations (p=0.0588) or between herbs (p=0.1670). The post-hoc test carried out also showed that there was no difference in the lethal effect of the two extracts. Conclusions Both extracts exhibited similar anthelmintic effects (p > 0.05), with LC50 of 8.91% for C. asiatica and 9.33% for M. pudica.

Program Studi Kedokteran, Fakultas Kedokteran dan Ilmu Kesehatan, Universitas Katolik Indonesia Atma Jaya (FKIK UAJ) menyelenggarakan kegiatan edukasi berupa workshop pembuatan apusan tinja dengan pewarnaan lugol dan demo preparat parasitologi. Kegiatan dirancang untuk memberikan gambaran mengenai pembelajaran dalam program studi kedokteran terutama saat berada di tahap preklinik. Ilmu parasitologi merupakan salah satu ilmu yang wajib dikuasai oleh lulusan kedokteran. Hal tersebut relevan karena sebagai salah satu negara beriklim tropis, penyebaran penyakit infeksi akibat parasit masih banyak ditemukan di Indonesia. Kegiatan edukasi dilakukan di laboratorium FKIK UAJ diikuti oleh siswa/i SMA Pahoa Gading Serpong. Evaluasi keberhasilan kegiatan dilakukan dengan membandingkan nilai pre-test dan post-test menggunakan aplikasi Quizizz. Hasil perbandingan nilai pre-test dan post-test terdapat kenaikan sebesar 27,7%. Hal tersebut menunjukkan adanya peningkatan pengetahuan umum siswa/i di bidang parasitologi. Kegiatan yang telah dilakukan memberikan dampak yang baik terhadap program pengenalan kampus FKIK UAJ dan meningkatkan pengetahuan siswa/i terhadap bidang ilmu kedokteran khususnya parasitologi. Sehingga kegiatan serupa dapat dilakukan secara berkesinambungan.
