Abstract

Actinomycetesis one of the microorganisms which have the ability of enzymes hydrolases such as cellulose, amylase, protease, and manannase. Manufacturer of enzymes of the hydrolase can be used as a candidate probiotic. A candidate probiotic should have a good resistance in different environmental conditions including pH and salinity.  This research is conducted to filter the candidate’s probiotics from bacteria Actinomycetes that tolerance salinity and has the activity of the enzyme hydrolase. Testing the activity of the enzyme was preformed qualitatively on the variation of pH 4, 7, and 9.8.  Theability of Atinomycetesproduce enzymes hydrolases is indicated by the colony on media containing the substrate.From the test results, cellulase enzyme activity with the largest enzymatic index value is 3.5 AF2 isolates pH 9.8. Amylase enzyme activity with the largest enzymatic index value is 3.66 RH1 isolates pH 7. Protease enzyme activity with the largest enzyme index value is 0.66, AF2 isolates at pH 7. Mananase enzyme activity with the largest enzymatic index value is 0.85, namely AF2 isolates pH 4. In testing the effect of salinity test isolates were able to grow at variations in the concentration of NaCl 0, 3, and 6%.

Abstract

Hydrolase enzymes are a class of enzymes used to break down the organic substrates into simpler molecules. Hydrolase enzymes play a role in biocontrol, industry, and public health. One of the hydrolase enzyme producers is bacteria. Serratia marcescens strain MBC1 is a collection in the Microbiology Laboratory of the Department of Biology, Faculty of Mathematics and Natural Sciences, University of Lampung which enzymatic activities haven’t been known yet. This research aimed to determine the enzymatic activity of S. marcesescens strain MBC1 that can be used as a candidate for biocontrol agents, biosurfactant producers, industry, or in the health sector. The screening was carried out using a qualitative method plate assay using selective solid media for each test, with three replications for each test. The data obtained were analyzed statistically using analysis non-parametric kruskall-wallis test. The lipolytic activity of the S. marcescens strain MBC1 showed high efficacy with an enzymatic index of 5.52 and amylase, cellulases, protease, mannanase, and chitinase with enzymatic indexes, respectively, 4.17, 1.50, 0.69, 0.35, and 0.27. This enzyme activity of this isolate can be used as a candidate for biological control agents,biosurfactant producers, industry, or the health sector.

Abstract

Malaria is a tropical disease that endangers public health which is caused by the plasmodium parasite, and transmitted through the bite of a female Anopheles sp. mosquito. Malaria is a major health problem in Indonesia due to its high risk of transmission and drug resistance. This encourages a study to look for other antimalarial alternatives by utilizing natural ingredients. The purpose of this study is to identify the functional groups found in Eucheuma cottonii secondary metabolites using methanol 70% as the solvent. This study is included in the experimental research. The functional group of methanol extract of Eucheuma cottonii was evaluated using Fourier Transform Infrared (FT-IR) Nicolet iS 10 spectrometers. The FT-IR analysis identified the O-H functional group at 3347.1 cm-1, C≡C at 2094.8 cm-1, C=C at 1632.6 cm-1, phenol or O-H bend at 1401.5 cm-1, aromatic C-O at 1192.7 cm-1, and a C-C group at 1043.7 cm-1. The results of phytochemical analysis on Eucheuma cottoni revealed the presence of saponins. The functional groups formed in the 70% methanol extract of Eucheuma cottonii seaweed are the O-H, C≡C, C=C, phenol or O-H bend, aromatic C-O, and C-C groups which are flavonoid compounds in the flavanone group which have the potential to be antimalarial. Furthermore, the presence of the O-H, C=O, C-O, C=C, or C-H functional groups is a characteristic of a flavonoid compound. Therefore, it can be concluded that the identified functional groups indicate the existence of flavonoid molecules from the flavanone group, which potential as antimalarial properties.

Abstract

Bakteri merupakan kandidat yang berpotensi sebagai larvasida Aedes aegypti. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak Serratia marcecsens strain MBC1 terhadap kematian larva instar III Aedes aegypti. Penelitian ini merupakan penelitian eksperimental dengan dua macam bentuk pengujian, yaitu uji larvasida untuk menentukan mortalitas dan uji lanjut untuk melihat perkembangan larva setelah perlakuan larvasida. Larva instar III diberi perlakuan dengan konsentrasi 125, 250, 500, dan 1.000 ppm. Uji larvasida diamati selama 24-72 jam. Uji lanjut larva instar III  diberi perlakuan dengan konsentrasi 500 dan 1.000 ppm serta diamati sampai semua larva mati. Kontrol positif menggunakan Abate® 1% dan kontrol negatif menggunakan air sumur. Hasil analisis probit didapatkan bahwa nilai LC50 dan LC90 ekstrak S. marcescens adalah 66.426,02 dan 749.001,41ppm. Larva instar III mati pada 18 hari setelah perlakuan ekstrak S. marcescens strain MBC1. Hasil dari penelitian ini menunjukkan bahwa larvasida ekstrak S. marcescens mempengaruhi lamanya siklus hidup dan mati sebelum mencapai stadium dewasa.     Bacteria have potencial as larvicide of Aedes aegypti. The purpose of this study is to determine the effect of Serratia marcescens strain MBC1 against larval third instars of Aedes aegypti. This research was an experimental study with two kinds of testing. The larvicide test to determine the mortality of the larval and the continuous test to observe the development of the larval after treatment with the larvacide. The third instars of mosquito larval were given bacterial extract in four concentrations (125, 250, 500, and 1000 ppm), and observed mortality during 72 hours. The continuous larvicide test was observed at two concentrations (500 and 1,000 ppm). The observation is completed when all of the larvae died. The positive control used Abate® 1% and the negative control used natural water. The treatment was repeated 4 times. The results show that LC50 and LC90 of S. marcescens extract are 66,426.02 dan 749,001.41 ppm.  All of the larvae in the continuous test die after 18 days have been givenan extract of S. marcescens strain MBC1. The experiment shows that the larvicide of extract of S. marcescens influences the length of the life cycle and larvae of Ae. aegypti die before reaching the adult.

Abstract

The Sars-Cov2 virus, also known as the Covid-19 pandemic, has led in a rise in the usage of immune-boosting supplement items. This situation resulted in higher prices and a scarcity of stock on the market. This program seeks to motivate and educate UPT Liwa Botanical Garden (KRL) employees about the necessity of utilizing herbs to boost body ’s immune system during the Covid-19 outbreak. The activity's target audience is workers, who interact often with tourists from various locations. Twenty persons participated, including KRL officials, Lumbok Seminung Kehati Park Managers, and KRL apprentice students. This activity takes the form of providing content, question and answer sessions, demonstrations on how to make herbal beverages, and training assessment using pre and post-test questionnaires. The results of the training implementation revealed an increase in participants' motivation and understanding of the importance of herbal functions and the skills of making herbal drinks, as evidenced by their enthusiasm in the discussion both before and after the demonstration of making herbal drinks.
