Abstract

Tissue culture succes is affected by medium composition. Murashige and Skoog (MS) basic medium composition is a medium tissue culture made from pure chemical substances which relatively expensive. Complete fertilizer commonly used as a replacement for MS basic medium is a complete fertilizer, which is relatively inexpensive and practical. Besides use suitable of basic medium composition, the addition of certain organic compound can also increase growth, development and resistance to disease of plant, such as chitosan. The research aims were to study the effect of complete fertilizer and chitosan on growth of PLBs and planlet Phalaenopsis Orchid Hybrids. The results showed that the basic medium composition complete fertilizer (N: P: K = 20:20:20) (2 g L-1) can be used as an alternative basal medium, because it can be increased the number of leaves , PLBs and plantlets higher than MS1/2 medium. Chitosan 5 ppm could not increased PLBs clump growth.

Abstract

Karakter pertambahan tinggi batang melambat merupakan salah satu tujuan dari pemuliaan tanaman kelapa sawit untuk optimalisasi umur ekonomis tanaman, mengurangi siklus replanting, dan mempermudah proses panen. Gen SMT 1 merupakan gen kunci didalam biosintesis hormon brasinosteroid yang berperan dalam pertumbuhan dan pertambahan batang tanaman. Teknik yang digunakan untuk mengidentifikasi gen yang memiliki karakter unggul tersebut adalah PCR (Polymerase Chain Reaction). Dalam proses PCR, desain primer merupakan hal yang sangat penting karena primer tersebut akan menentukan keberhasilan dalam proses amplifikasi PCR. Tujuan dari penelitian ini untuk mendapatkan kandidat primer yang akan digunakan dalam analisis keragaman genetik terkait pertambahan tinggi batang berdasarkan situs SNP. Data sekuen DNA kelapa sawit diperoleh pada laman NCBI. Pensejajaran sekuen DNA dilakukan pada sofware geneious prime untuk mengidentifikasi situs SNP. Desain primer dilakukan pada perangkat lunak WebSNAPER. Hasil penelitian menghasilkan dua kandidat pasang primer yang akan digunakan untuk melakukan analisis keragaman genetik terkait gen pengendali pertambahan tinggi batang melambat. Masing- masing primer terdiri dari primer reference dan alternate dan memiliki ukuran amplikon yang berbeda pada setiap pasang primer. The character of growth inhibitor stem is one of the goals of oil palm plant breeding to increase the economic life of the plant, reduce the replanting cycle and simplify the harvesting process. The SMT 1 gene is a key gene in the brasinosteroid hormone which plays a role in the growth and addition of plant stems. The technique used to identify genes with superior characters is PCR (Polymerase Chain Reaction). In the PCR process, the design of the primer is very important because the primer will determine the success of the PCR amplification process. The aim of this study was to obtain primer to be used in the analysis of genetic diversity related to stem height gain. Oil palm DNA sequence data were obtained in terms of NCBI. DNA sequence alignment was performed on geneious prime software to obtain SNP sites. Primer design was performed on WebSNAPER software. The results of the study obtained two candidate pairs of primers to be used for the analysis of genetic diversity related to the increase in the height of the stems of the stem. Each primer consists of reference and alternate primers and has a different amplicon size for each pair of primers.

Abstract

Teknik perbanyakan secara in vitro atau kultur jaringan mulai diaplikasikan untuk perbanyakan kelapa sawit, yang mana terbukti dapat menghasilkan tanaman kelapa sawit dalam jumlah banyak dengan waktu yang relatif lebih singkat.  Kendala utama saat fase inisiasi tanaman yaitu pencokelatan (browning) yang menyebabkan gangguan fisiologis tanaman sehingga terjadi kematian pada eksplan in vitro. Senyawa antioksidan merupakan senyawa yang dapat menghambat pencokelatan. Berbagai penelitian telah dilakukan untuk mencegah kemunculan browning pada kultur in vitro, namun setiap komoditas yang diuji memiliki senyawa antioksidan yang beragam untuk mendapatkan antioksidan terbaik yang dapat menghambat browning. Penelitian ini bertujuan untuk mendapat senyawa antioksidan terbaik untuk menghambat bahkan meniadakan terjadinya browning pada fase inisiasi kelapa sawit in vitro.  Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) non faktorial yaitu perlakuan zat antioksidan menggunakan tiga jenis zat antioksidan yaitu arang aktif (2.5 gL-1), Polyvinylpyrrolidone (PVP) (2.5 gL-1), dan asam akorbat 100 mgL-1). Data analisis ragam diuji lanjut menggunakan Duncan Multiple Range Test (DMRT) untuk data yang berbeda nyata pada taraf 5% dengan software STAR (Statistical Tool for Agriculture Research). Hasil penelitian menunjukkan antioksidan terbaik untuk menekan munculnya waktu browning dan intensitas browning adalah PVP sebesar 2.5 gL-1 pada tanaman kelapa sawit in vitro.

Abstract

Male inflorescence residue of the oil palm rich in fibre and contains cellulose content and has the potential to synthesize Carboxymethyl Cellulose (CMC). Male inflorescence is considered necessary removed in the early stages of the oil palm cultivation process to develop stem size, string and robust root systems. This activity generates plentiful of oil palm male inflorescence residues in the plantation. The aim of this research is to synthesis and characterize CMC fabricated from oil palm male inflorescence (OPMI-CMC) waste for use as an ice cream stabilizer, using a variety of treatments, which are commercial CMC 0.2%, OPMI-CMC 0.1%, 0.2%, and 0.3%. The results showed that CMC can be produced from organic waste of male flowers of oil palm plants. The water content, pH, purity, and degree of substitution meet the standard. The optimal concentration of OPMI-CMC for ice cream stabilizer is 0.3% based on melting time and overrun value. The FTIR analysis shows that the main functional group of the OPMI-CMC is highly comparable. Therefore, to possibly be employed as an alternative to organic CMC for food stabilizers, other standards must be fulfilled, and further research is required.

Abstract

Soft-rot disease (SRD) in Phalaenopsis, caused Dickeya dadantii, has resulted in significant losses in the orchid sector in Indonesia. This study aimed to evaluate the inoculation method of Dickeya dadantii and identify the resistance response of individual regenerated plantlets of Phal. amabilis from irradiated protocorm. A detached leaf assay was used to evaluate the inoculation method and resistance response of SRD. Based on the results of this study, Dickeya dadantii bacteria could only infect the leaves through wounding tissue. The density of bacteria that could infect leaf tissue was OD600 = 0.2. All dilution factors tested caused soft rot symptoms in P. amabilis. On the other hand, Vanilla planifolia only showed symptoms at a dilution factor of 10-0. Four accessions of regenerated plantlets from irradiated protocorms were resistant to SRD. They were from irradiation 5 Gy (IP 05 Gy-23, IP 05 Gy-31, and IP 05 Gy-33) and one accession from the control treatment or without irradiation (IP 0 Gy -1). These results showed that 5 Gy irradiation increased plant resistance to SRD in Phalaenopsis. A dose of 5 Gy can potentially produce mutant lines resistant to SRD in Phalaenopsis or other plants, too.

Abstract

The utilisation of oil palm empty fruit bunches (EFB) remains underexplored. Harnessing cellulolytic microorganisms for the production of cellulase enzymes offers sustainable approach to addressing waste management challenges while aligning with the principles of the Sustainable Development Goals (SDGs) and address waste management challenges. This study aims to isolate, characterize, identify, and test the potential cellulase activity of cellulolytic bacteria from EFB taken from three different locations: PO code from organic fertilizer plantations (POU1, POU2, POU3), PL code from oil palm plantations (PLU1, PLU2, PLU3), and PK code from Sulung mills (PKU1, PKU2, PKU3). This study used three isolated cultures in its testing. The research process includes sample preparation, bacterial isolation, gram staining, catalase test, hypersensitivity test, DNA amplification, bioinformatics analysis and cellulase activity analysis. The results of the bacterial isolation obtained 28 colonies. The results of the characterisation were all 3 non-pathogenic bacterial isolates, with a positive catalase test. The result of staining Gram-negative with bacilli-shaped bacteria. The amplification results obtained a band size of 1500 bp. The results of the identification obtained the species Aeromonas enteropelogenes, Nitrosomonas stercoris, and Methylobacillus caricis. The results of phylogenetic analysis showed low homology. Cellulase activity of six positive isolates with medium ability isolates code POU3 (1.3), PLU2 (1.0), PLU3 (1.0); low isolates POU1 (0.2), POU2 (0.2), PLU (0.8) and 3 negative isolates no enzyme activity PKU1 (-1), PKU2 (-1), and PKU3 (-1).

Abstract

In vocational education, field practice is not only considered as a complement, but also as the core of learning that can produce graduates who are ready to contribute directly to the world of work. Professional Job Lecture (PJL) is a mandatory courses in the sixth semester. All learning is carried out practically. This research aims to examine the level of knowledge of students who are participants in the PJL. The research was conducted on 15 August until 31 October 2023 at the Citra Widya Edukasi Oil Palm Polytechnic on all students of the Production Technology of Plantation Crops study program for the year entering 2021. The number of students in the survey was 37 people. The student has participated in PJL activities at the Subang oil palm educational plantation of the Citra Widya Edukasi Oil Palm Polytechnic for 20 days. Primary data was used in this research through filling out questionnaires. Level of confidence and knowledge of students measured by submitting statements and questions in online questionnaire. Questionnaire-filling was carried out on the first day of the PJL and 2 months after the activity was completed. The level of knowledge of students PJL about oil palm plantation management was medium to very high level based on hypothetical data categorization. The majority of students have a high level of knowledge as much as 62.86%. Effective communication based on SKKN topic for Plantation Asisstants was the highest percentage of corecct answers namely 89.86. Self-confidence was increased after PJL with the number of students by 31 people.

Abstract

Kelompok tani maduhur di Kecamatan Taman Sari, Kabupaten Bogor merupakan petani kopi. Beberapa jenis kopi ditanam diantaranya kopi arabika, robusta, liberika, dan kopi kultivar lokal yakni kopi “buhun”. Kelompok tani maduhur dalam teknis budidaya kopi masih mengalami beberapa kendala, diantaranya ketersediaan bibit unggul, kondisi agroklimat serta serangan hama dan penyakit. Hal ini menyebabkan potensi produksi dari tanaman kopi belum optimum. Sedangkan di pasaran permintaan kopi terus meningkat dan menuntut kualitas yang prima. Oleh karena itu, kelompok tani maduhur diberikan pendampingan dan inisiasi pusat pengembangan kopi khususnya di Kecamatan Tamansari, Kabupaten Bogor. Hasil diskusi dan wawancara dari petani kopi menunjukkan minimnya pengetahuan teknis budidaya kopi. Tingkat pengetahuan petani masih terbilang minim sehingga hasil produksi yang diperoleh belum mencapai target. Kesimpulan dari kegiatan pengabdian kepada masyarakat ini berupa penyuluhan mengenai teknik budidaya kopi sesuai dengan kondisi aktual di lapangan dan memberikan arahan kepada seluruh anggota kelompok tani untuk mempraktikkan materi yang telah diberikan.
