Abstract

This research aimed to identify polymorphism of the GH gene in Sikumbang Jonti ducks using the PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism) method as a candidate for genetic marker selection. This research used 123 blood samples of male and female Sikumbang Jonti ducks. The ducks were eight weeks old and kept intensively at the UPT Fakultas Peternakan Universitas Andalas. Blood samples of Sikumbang Jonti ducks were taken through the brachial vein as much as ± 2 mL. The Genomic DNA Purification Kit (Promega) protocol isolated DNA from blood samples. A pair of primers F amplified the total DNA: 5'-GGA CAG CCT GAG GAA AGA GT-3 'and R: 5'-GTG GAA GGT GGG GAG ACT TC-3', which produced a fragment region of the GH gene exon 3 with a cut region of part intron two and part intron 3 with a primary length of 833 bp. The amplified product was restricted by the enzyme TasI which recognizes the cutting site (↓AATT). Based on the restriction results, two (2) genotypes were obtained, namely homozygous (-/-) as many as four (4) and homozygous (+/+) as many as 119. The analysis of restriction products included the allele frequency, namely the allele (+) of 0.98 and the allele (-) of 0.02, and genotypes frequency of homozygous (-/-) of 0.03 and homozygous genotypes (+/+) of 0.97. The diversity of the GH gene is in the Hardy-Weinberg equilibrium. Based on the research results, it can be concluded that there is diversity in the GH gene of the Sikumbang Jonti duck, and it is in the Hardy-Weinberg equilibrium, so it can be used as a candidate for the genetic marker of ducks.

Abstract

Food independence is a reference for a country in solving various problems related to the availability of the necessities of life for people who do not depend on other countries, especially the livestock sector. Prioritizing the development of local livestock such as native chickens as producers of food products of animal origin, able to answer challenges in the world of animal husbandry as it is today. This study aims to: (1) conduct an analysis of the factors that influence the development of free-range chicken in the city of Padang, (2) provide an alternative strategy based on the potential of the city of Padang for a better free-range chicken development effort. This research was conducted by conducting surveys and direct observations at the research site. The research was carried out in three stages: 1) Identifying and analyzing external factors from the free-range chicken development effort, 2) Identifying and analyzing the internal factors of the free-range chicken development business, and 3) Creating alternative strategies for the development of free-range chicken livestock in the city of Padang. From the results of the study, it is known that the city of Padang has a carrying capacity index of 9.2 which means that it is included in the category of good carrying capacity for the development of free-range chicken businesses. Abstract

This activity aims to provide training and technology packages for improving the maintenance system, knowledge of feed ingredients, technology for producing duck feed based on local feed, restricted feeding. The ultimate goal of this activity is to increase egg production, productivity, feed cost efficiency in the context of developing and increasing farmers' income while forming / developing economically independent partner groups. This activity was carried out in Tanjung Haro Selatan, Luak District, 50 Kota Regency, West Sumatra. The partner involved in this activity is the "Duck Farmer Group "Usaha Baru" which reared egg and meat ducks. The methods used in this service program are counseling, demonstration, training, coaching and provision of technology packages, as well as observation and evaluation and data collection to measure the success parameters of the program being carried out. Based on the implementation of the community service, farmers were very enthusiastic in participating in this activity, almost 80% percent of farmers understand feeding management according to the needs of duck feed based on age level both in quality and quantity. the quality of the rations prepared by the farmer is not sufficient for the nutritional needs of ducks. Increasing egg production can be done by preparing duck rations by increasing animal protein and mineral levels in the ration. Increasing the income of duck farmers can be done by reducing production costs with rations prepared by farmers themselves sourced from local feed ingredients at a price of Rp. 6000, -.
