Abstract

Untuk menghasilkan produk ternak organik yang bebas bahan aditif kimia, hormon, antibiotik perlu dilakukan suatu program penerapan teknologi tepat guna dengan pemanfaatan Waretha (Bacillus amyloliquefaciens) dalam usaha tersebut sebagai inokulum/probiotik. Waretha sebagai probiotik dapat memperbaiki kualitas produk dan meningkatkan efisiensi penggunaan pakan. Probiotik Waretha adalah luaran dari beberapa hasil penelitian selama enam tahun dimulai dari ditemukan bakteri B. amyloliquefaciens sebagai isolat selulolitik, proteolitik dan fitalitik serta sebagai inokulum fermentasi. Satu kali pemberian probiotik Waretha pada DOC broiler tanpa diberi fed aditif (vaksin, vitamin, antibiotik, hormon) diperoleh efisiensi penggunaan ransum meningkatkan dari 61% menjadi 67%, litter tidak basah dan berbau, meningkatkan populasi Lactobacillus sp dan menekan populasi E. Coli, persentase karkas meningkat dari 67% menjadi 72% dan kolesterol turun dari 2,61 menjadi 1,53 mg/g/daging. Untuk ayam petelur efisiensi penggunaan ransum meningkatkan dari 41,% menjadi 50%, litter tidak basah dan berbau, dan kolesterol turun dari 12,15 menjadi 10,21 mg/g/kuning telur. Sebagai inokulum dan sekalian sebagai probiotik Waretha (Bacillus amyloliquefaciens) telah memfermentasi darah yang diserap oleh dedak padi pada dosis inokulum 3% dan lama fermentasi 3 hari (pakan DDPF) diperoleh peningkatan kualitas produk dengan kandungan PK 56,28%, SK 7,17%, retensi nitrogen 64,07%, 7,17%, kec. SK 47,58% dan EM 3373 kkal/kg. Produk DDPF mengandung spora B. amyloliquefaciens 1013cfu/g yang berfungsi sebagai probiotik yang menghasilkan produk organik. Penggunaan 15% produk DDPF sebagai pengganti 30% bungkil kedelai dan 50% tepung ikan pada ransum broiler dapat menurunkan konversi ransum dari 1.84 menjadi 1.76. Hasil yang sudah dicapai pada kegiatan tahun II, produksi dua ton DDPF dan melakukan pemeliharaan ayam ras petelur sebanyak 700 ekor semenjak bulan Mei 2016, dan sekarang sudah bertelur 50%. Pada uji untuk mengetahui level pemakaian pakan DDPF pada ayam ras petelur diperoleh bahwa pakan DDPF dapat menggantikan konsentrat CP 124 35% atau harga pakan per kg menjadi lebih murah yaitu dari Rp. 4.800,- menjadi Rp. 3.300,-.

Abstract

The research aimed to evaluate the effect of Zn supplementation in diet of Sikumbang Janti female duck on its performance, carcass, digestive organs, and intestinal morphology. The study used 96 female ducks aged 8 weeks. This research used a completely randomized design with four treatments (0, 30, 60, and 90 mg Zn/kg), each treatment was repeated four times. Performance, carcass, digestive organs, and intestinal morphology were observed. The results showed that Zn supplementation significantly (P0.05) affected body weight, body weight gain, feed conversion ratio, carcass weight, abdominal fat, and intestinal morphology, but it did not affect (P0.05) feed consumption, carcass percentage, and digestive organs. In conclusion, supplementation of 60 mg Zn/kg in the diet improved performance, intestinal morphology, and the health of visceral of Sikumbang Janti female ducks aged 8 to 16 weeks.

Abstract

This study aims to determine to what extent, the role of monochromatic light then being fed turmeric additives has an impact on the blood profile and body weight gain of Pitalah ducks. The material used was 100 Pitalah ducklings. The method used was an experiment with the Split Plot method arranged in a Randomized Block Design (RBD). The treatment started on day 15 and the duration of the study was 10 weeks. The treatment given is the main plot factor is monochromatic lighting consisting of yellow light as a control, blue, red, green. Subplot factor, namely turmeric consisting of B1: 18 gr and B2: 16 gr. The experimental results showed that the interaction between factor A (light) and factor B (turmeric) had no significant effect (P>0.05) on blood profile and body weight gain in Pitalah ducks. The conclusion of this study is that the administration of monochromatic light (yellow, blue, red, green) with turmeric levels (18 gr and 16 gr) has no interaction on blood profile (leukocytes & erythrocytes) and body weight gain in pitalah ducks. In this condition, leukocyte values ​​ranged from 18.39 to 73.51 (103 /mm-3 ) and erythrocyte values ​​from 18.39 to 73.5 (103 /mm-3 ), both of these values ​​were in a less normal range so that it can be indicated that the health condition of the ducks is not prime. Subsequently, body weight gain was obtained 63.58 - 78.16 grams/tails/week. Keywords: Monochromatic light, turmeric, blood profile, body weight gain and Pitalah ducks

Abstract

Fertilitas, daya tetas dan bobot tetas adalah salah satu faktor keberhasilan dalam program pembibitan. Penelitian ini dilakukan di UPTD Ternak Unggas yang merupakan salah satu sentra pembibitan unggas di tingkat daerah yang fokus dalam pembibitan dan pengembangan unggas unggul salah satunya pembibitan dan pengembangan ayam KUB-1. Penelitian ini bertujuan untuk mengetahui kondisi fertilitas, daya tetas dan bobot tetas ayam KUB-1 di UPTD Ternak Unggas. Metode penelitian adalah observasi dan pengukuran dengan melakukan pengamatan secara langsung dan intensif selama penelitian, dengan menggunakan 200 butir telur tetas ayam KUB-1 hasil produksi UPTD Ternak Unggas sebagai sampel penetasan. Variabel yang diamati fertilitas, daya tetas dan bobot tetas. Hasil penelitian kondisi penetasan telur ayam KUB-1 di UPTD Ternak Unggas dapat dikatakan cukup baik karena memiliki fertilitas 82,5 %, daya tetas 73,33 % dan rataan bobot tetas 29,33 gr. Sedangkan untuk bobot tetas sudah 93% telah memenuhi persyaratan SNI. Kata Kunci: Fertilitas, daya tetas, bobot tetas dan ayam KUB-1.

Abstract

The empowerment of village communities is within the framework of the Independent Campus Learning Program (MBKM) Building Villages, which is carried out for the people of Sungai Batu Gantih Village, Gunung Kerinci District, Kerinci Regency, Jambi Province. Based on the results of a preliminary survey in Sungai Batu Gantih Village, in general, the village community carries out their livestock business part-time, with food crop farming activities as the main business. The knowledge of the people of Sungai Batu Gantih Village regarding the cultivation of free-range chickens is still low. Chicken rearing is carried out extensively, relying on household leftovers as feed, resulting in low productivity and a limited population. So, this activity aims to improve the abilities and skills of village communities in raising chickens through socializing the joper chicken so that they can increase productivity and increase people's income in Sungai Batu Gantih Village. Joper chickens are the result of crossing male Joper chickens and female purebred chickens with quite high egg production, and the eggs and meat have a distinctive taste. This activity will be carried out in September–December 2022. The method of activity carried out is participatory community empowerment, which includes counseling, field practice, and mentoring. The implementation of empowerment activities went well and received a positive response from the village community. This was shown by the percentage of Joper chickens that lived to 4 weeks of age, reaching 90% with a body weight ranging from 230 to 290 g/head.

Abstract

This activity aims to provide training and technology packages for improving the maintenance system, knowledge of feed ingredients, technology for producing duck feed based on local feed, restricted feeding. The ultimate goal of this activity is to increase egg production, productivity, feed cost efficiency in the context of developing and increasing farmers' income while forming / developing economically independent partner groups. This activity was carried out in Tanjung Haro Selatan, Luak District, 50 Kota Regency, West Sumatra. The partner involved in this activity is the "Duck Farmer Group "Usaha Baru" which reared egg and meat ducks. The methods used in this service program are counseling, demonstration, training, coaching and provision of technology packages, as well as observation and evaluation and data collection to measure the success parameters of the program being carried out. Based on the implementation of the community service, farmers were very enthusiastic in participating in this activity, almost 80% percent of farmers understand feeding management according to the needs of duck feed based on age level both in quality and quantity. the quality of the rations prepared by the farmer is not sufficient for the nutritional needs of ducks. Increasing egg production can be done by preparing duck rations by increasing animal protein and mineral levels in the ration. Increasing the income of duck farmers can be done by reducing production costs with rations prepared by farmers themselves sourced from local feed ingredients at a price of Rp. 6000, -.

Abstract

ABSTRAK. Tujuan dari penelitian ini adalah untuk mengidentifikasi pengaruh polimorfisme gen heat shock protein 70 (HSP70) terhadap sifat toleransi panas. Genotiping dilakukan pada 110 itik lokal dari 4 populasi itik Sumatera Barat (pitalah, bayang, kamang, dan payakumbuh). Enzim restriksi yang digunakan adalah SacII. Cekaman panas dilakukan selama 1 jam dengan sampel itik lokal sebanyak 24 ekor untuk menganalisa status hematologi. PCR-RFLP dan Sanger sequencing assay digunakan untuk mengidentifikasi polimorfisme. Analisis polimorfisme dilakukan dengan menggunakan Software MEGA 7 dan pengaruhnya dianalisis dengan ANOVA menggunakan software SAS 9.4. Produk amplifikasi yang dihasilkan yaitu 466 pasang basa. Lokus HSP70/SacII mengenali satu situs SNP (Single nucleotide Polymorphism) g.1702TC, menghasilkan dua alel (T dan C) dan tiga genotype (TT, TC, CC). Gen HSP70/SacII bersifat polimorfik pada semua populasi. Hasil penelitian menunjukkan bahwa alel T memiliki frekuensi yang lebih tinggi daripada alel C pada semua populasi. Analisis chi-kuadrat (2) menunjukkan bahwa semua populasi itik lokal memenuhi kaidah kesetimbangan Hardy-Weinberg. Pengaruh genotipe gen HSP70 terhadap status hematologi menunjukkan perbedaan (P0,05) terhadap nilai heterofil dan nilai H/L dengan genotipe CT menunjukkan toleransi panas yang lebih baik dibanding genotipe lainnya. Gen HSP70/SacII dapat dijadikan marka molekuler sifat toleransi panas pada itik lokal berdasarkan keragaman dan pengaruhnya terhadap status hematologi.(The effect of heat shock protein 70 (HSP70) SacII gene polymorphism on heat tolerance of West Sumatera local duck)ABSTRACT. The objective of this study was to identify the effect of HSP70/SacII gene polymorphism associated with the thermotolerance trait. Genotyping was performed on 110 local duck from the West Sumatera ducks population (pitalah, bayang, kamang, and payakumbuh). The restriction enzyme used was SacII. Heat stress was done for 1 hour with samples used as much as 24 birds to analyze hematological status. PCR-RFLP and Sanger sequencing assays were used to identify polymorphism. Analysis of polymorphism was conduct by MEGA 7 software and its effect is analyzed with ANOVA using Statistical SAS 9.4 software. The product of amplification was 466 bp. HSP70/SacII recognized one SNP g.1702TC, producing two alleles (T and C) and three genotypes (TT,TC,CC). The HSP70/SacII locus were polymorphic in all population. T allele had a higher frequency than C allele in all populations. The analysis of chi-square (2) showed that all local ducks population were fit with Hardy-Weinberg equilibrium. The effect of HSP70/SacII gene on hematological status showed differences (P0,05) of heterophil and H/L ratio with CT genotype show better heat tolerance than other genotypes. HSP70/SacII gene can be used as a marker of heat tolerance in local ducks based on polymorphism and its effect on hematological status.

Abstract

This study aims to optimise the growth of Superior Balitbangtan (KUB-2) chickens through feed restriction during the starter period and its effect on the recovery phase. This study used 100 two-week-old KUB-2 chickens, placed in 20 cages measuring 70 cm x 70 cm x 60 cm, with each cage containing 5 KUB-2 chickens. The study employed a Completely Randomised Design (CRD) with four treatments and five replicates. The variables observed were feed intake, body weight gain, feed conversion ratio (FCR), Growth Rate,  and Income Over Feed Cost (IOFC). The research showed that applying 20%, 30%, and 40% feed restriction had a significant effect (P < 0.01) on decreasing feed intake and body weight gain, but had no significant effect (P > 0.05) on the feed conversion ratio during the 4 weeks of treatment. During the recovery phase, ad libitum feeding had a significant effect (P < 0.05) on feed intake, but did not affect body weight gain or feed conversion ratio. Throughout the study, feed restriction followed by recovery had a very significant effect (P < 0.01) on feed intake and a significant effect (P < 0.05) on body weight gain and feed conversion ratio. This study concludes that the 20% feed restriction treatment showed the best performance, with a feed intake of 339.97 g/bird/week, body weight gain of 98.35 g/bird/week, a feed conversion ratio of 3.46, a growth rate of 0.227, and an Income Over Feed Cost (IOFC) of Rp. 14,815.

Abstract

This study aimed to identify the polymorphism of the exon-2 myostatin gene (MSTN) in Pitalah ducks using the PCR-RFLP (Polymerase Chain Reaction-Restriction Fragment Length Polymorphism) method. 28 blood samples of Pitalah ducks were used in this study. Blood samples were extracted using Intron Biotechnology's Intron G-SpinTM Total DNA Extraction Kit protocol. The DNA extraction results were then amplified using a pair of primers, namely the Forward: 5' TCCACTTGTTACTGATGCTGT-3' primer and the Reverse: 5'- TAGGGAAATGGAGGCACAGG-3' primer, with a fragment target of 700 bp. Furthermore, the amplified product is restricted using the Taq1 enzyme, which recognizes the cutting site on T|CGA. Based on the results of the study, two genotypes were found, namely 89.29% truncated homozygous (+/+) and heterozygous (+/-), as well as 10.71% of the total sample used, while untruncated homozygous genotypes (-/-) were not found in this study. Meanwhile, the allele frequency (+) was 0.946, the allele frequency (-) was 0.054, and the observed heterozygosity value was greater than the expected heterozygosity value (Ho>He). This study concludes that the myostatin exon-2 gene in Pitalah ducks is polymorphic and is in Hardy-Weinberg equilibrium.

Abstract

AbstrakProlaktin merupakan hormon polipeptida yang disekresikan oleh kelenjar pituitary anterior dan secara spesifik kandidat gen yang mengontrol variasi jumlah produksi telur dengan reduksi biosintesis telur dalam beberapa periode mengeram. Tujuan penelitian ini  adalah untuk menganalisis keragaman gen Prolaktin pada itik Pitalah. Ekstraksi yang dilakukan pada sampel darah 45 ekor itik Pitalah dengan menggunakan protocol G-spinTM Total DNA Extraction Mini Kit dari Intron Biotechnology. Selanjutnya diamplifikasi dengan menggunakan sepasang primer forward 5’- CTG CAT CTG TGG ACA TTG CT-3’ dan reverse 5’GAA GCA GGT TTG GGA GTA CG-3’ dengan target fragmen 496 bp dan disekuensing menggunakan jasa 1sr Base Singapore. Data hasil sekuensing dianalisis menggunakan Seqmen DNASTAR dan MEGA 11 dan menunjukkan 14 keragaman pada daerah ekson 4 dan parsial intron 3 dan 4 pada titik mutasi INDEL 3703A, INDEL3939A, C3974A, A4030G, A4038C, T4056C, G4061A, T4091G, T4094A, T4098C, G4102A, C4106A, A4107C dan C4154G. Keragaman gen Prolaktin bersifat polimorfik dengan frekuensi genotype tidak dalam keseimbangan Hardy-Weinberg. Kata Kunci: Prolaktin, Itik Pitalah, Keragaman, Mutasi, Mutasi, Sekuensing
