Abstract

Meretrix meretrix is one type of shellfish that are included in the bivalves that are filter feeders that live in the waters. Many bacteria that live in the estuary with high salinity in the range of 20-40 ppt one of which is the Vibrio bacterium. This research aims to determine the presence of Vibrio spp. bacteria in M. meretrix from Cengkrong Mangrove Eduwisata River, Trenggalek. The Isolation of Vibrio bacteria was conducted by streat plate method on selective media of TCBSA (Thiosulfate Citrate Bile Salt Sucrose Agar) and identify the Vibrio bacterium through the morphological and biochemical tests based Bergey's Manual of Systematic Bacteriology In Microbiology laboratory of Biology Faculty of Gadjah Mada University. The Result showed there are 7 isolates Vibrio spp. bacterias in M. meretrix they are isolate 1.1, 1.4, 1.7, 1.8. 5.3, 3.5, 2.6 PSBI Keyword: Meretrix meretrix, Vibrio, identification

Abstract

Sumber konsumsi ikan masyarakat Kota Tarakan adalah ikan layang dan bandeng yang berasal dari pasar tradisional. Pengelolaan pasar tradisional yang ada di Kota Tarakan cukup memadai namun, tingkat kesehatan dan higienis lingkungan masih kurang baik, penanganan yang belum memadai memungkinkan banyak bakteri yang berkembang dan mengontaminasi ikan. Tujuan penelitian untuk mengetahui kontaminasi bakteri patogen (Vibrio spp. dan Salmonella spp.) pada ikan yang dijual di pasar tradisional kota Tarakan. Penelitian dilakukan dengan metode deskriptif dengan beberapa tahapan mulai dari observasi pasar, pengambilan sampel, isolasi bakteri dan identifikasi. Isolasi bakteri Vibrio menggunakan media selektif TCBS (Thiosulfate Citrate Bile Salt Sucrose) dan bakteri Salmonella menggunakan media SSA (Salmonella Shigella Agar) identifikasi bakteri Vibrio dan Salmonella yang berpedoman pada buku Bergey’s Manual of Determinative Bacteriology berdasarkan sifat morfologi dan kimiawi. Hasil identifikasi terdapat kontaminasi bakteri Vibrio spp. dan Salmonella spp. pada hasil perikanan yang dijual di pasar tradisional kota Tarakan khususnya ikan bandeng dan ikan layang.

Abstract

Keberadaan bakteri Salmonella spp pada ikan bandeng dapat mempengaruhi aspek kualitas, kemunduran mutu produk perikanan dan keamanan dalam mengkonsumsi. Tujuan penelitian untuk mengetahui tingkat kesegeran dan kontaminan bakteri pada ikan bandeng yang dijual di Pasar Guser Kota Tarakan dan mendeteksi keberadaan bakteri patogen (Salmonella spp.). Sampel daging ikan dihaluskan sebanyak 25 gram dan dimasukan ke dalam Erlenmeyer berisi 225 ml Alkhali Pepton Wather (APW) kemudian di homogenkan dan diinkubasi selama 5-8 jam. Isolasi bakteri Salmonella spp. dengan menggunakan media Salmonella Shigella Agar (SSA) kemudian diinkubasi selama 24-48 jam suhu 37oC dan diidentifikasi dengan uji biokimia dengan menggunakan Bergey’s Manual of Determinative Bacteriology. Hasil penelitian diperoleh bahwa mutu dan kualitas ikan bandeng yang dijual di Pasar Gusher Kota Tarakan sangat baik dan segar dengan tingkat kontaminan bakteri dibawah ambang batas/baku mutu SNI yaitu 4x104 koloni/gram. Serta terdeteksi mengandung bakteri patogen (Salmonella spp.).

Abstract

Bakteri di perairan merupakan komponen biotik yang penting pada proses dekomposisi sebagai sumber penyedia zat hara. Tujuan penelitian ini adalah untuk mengetahui keanekaragaman bakteri pada perairan di Kawasan Konservasi Mangrove dan Bekantan (KKMB) Kota Tarakan. Penelitian ini dilaksanakan pada bulan Maret sampai Mei 2018 dengan menggunakan metode deskriptif dengan melakukan identifikasi bakteri pada perairan melalui beberapa tahapan pengujian yaitu uji gram, uji utama dan uji lanjut yang dilakukan di laboraturium Stasiun Karantina Ikan, Pengendalian Mutu dan Keamanan Hasil Perikanan kelas II Tarakan. Hasil penelitian menunjukkan keanekaragaman bakteri pada perairan sebanyak 9 bakteri (Genus) yaitu Bacillus spp., Corynebacterium spp., Listeria spp., Enterobacteria spp., Pseudomonas spp., Aeromonas spp., Micrococcus spp.,  Staphylococcus spp., dan Actinobacillus spp., dan yang paling dominan ditemukan adalah Bacillus spp.

Abstract

The Covid-19 pandemic has had a major impact on society, such as job losses, rising staples, and economic hardship. Prices of food needs that continue to increase and are not affordable have resulted in food problems, one of which is protein. One of the high-quality protein is protein from fish. Fish as a fishery commodity that is relatively easy to cultivate on a large industrial scale and household scale for food self-sufficiency is catfish (Clarias sp). The purpose of this activity is to add insight and knowledge of the community about budikdamber for food self-sufficiency. The method of implementing community service activities in Kampung Six is carried out by counseling and training methods on Budikdamber. In this training activity, a bucket with 40 liters of water and 20 catfish seeds was used. Maintenance was carried out for 60 days with ad satiation feeding three times a day. Observations were made on growth, survival and water quality. The results of the activity found that 93% of the community partners had understood about Budikdamber. Budikdamber maintenance by partner communities gets an absolute weight growth value of catfish of 14 g, catfish survival value of 75%, and 1 bunch of kale per bucket. Based on the results of this activity, it is hoped that there will be the provision of special land for Budikdamber as an example for people who want to develop in their respective yards.

This study aims to determine the diversity and similarity of Vibrio in M. meretrix at eduwisata mangrove Cengkrong Beach, Trenggalek. Isolation and identification of Vibrio was conducted by using the media TCBSA (Thiosulfate Citrate Bile Salt Sucrose Agar) streat plate through morphological observation, physiological and biochemical tests based Bergey's Manual of Systematic Bacteriology conducted in the laboratory of Microbiology Faculty of Biology, Universitas Gajah Mada. Data analysis was performed using the MVSP (Multivariate Statistical Package) 3.1 in the form of a dendrogram showing the similarity relationship Vibrio bacteria. The results of the identification of Vibrio bacteria in M. Meretrix are V. herveyi, V. parahemolyticus, V. nereis and V. proteolyticus. V. herveyi has similarities with V. parahemolyticus 66% and 62% with V. proteolyticus while with V. nereis bacterium 54%.
