Abstract

The purpose of this study was to determine the effect of solvent types on antioxidant activity of turi (Sesbania grandiflora) leaves liquid extract and to determine the compatible solvent types which have the highest antioxidant activity of turi leaves liquid extract. The research used a completely randomized design with the treatment of solvent types consisting of 4 level which is: aquadest; acetone 70%; methanol 70%; and ethanol 70%. The treatment was repeated four times so that they were obtained 16 experiment units. The data was analyzed with analysis of variance and if the treatment had a significant effect followed by a Duncan's Multiple Range Test. The results showed the type of solvent treatment had a significant effect on antioxidant activity, total flavonoid, total tannin, and vitamin C content. The best treatment was obtained from turi leaves liquid extract using acetone 70% solvent which had the highest antioxidant activity of 75.71% with the value of IC50 269.67, total flavonoid 0.185 mg QE/g material, total tannin 0.013 mg TAE/g material and vitamin C content 0.45 mg AAE/ g material. Abstract

Fermented milk is a functional food has beneficial to human health. The recommended concentration of probiotic bacteria to provide health benefits is 106-108 CFU/ml of product. The storage temperature is one of the factor that affected the characteristics of fermented milk. This research was conducted to determine the effect of storage time at room temperature to the characteristics of fermented milk product and determine the length of maximum storage at room temperature. The research design was Completely Randomized Design with storage time treatment at room temperature for 0, 2, 4, 6, 8, 10, 12, and 14 days. Each treatment was repeated 2 times so that 16 experimental units were obtainned. The variables that were observed included total LAB, total acid, pH, and sensory evaluation (aroma, taste, and overall acceptance). Data were analyzed with analysis of variance, if the treatment had an effect on the variables then followed by Duncan multiple range Test. The results showed that the treatment of fermented milk in room temperature storage significantly affected the total acid, pH, total LAB, and sensory evaluation (aroma, taste, and overall acceptance). The maximum storage of the room temperature was days with total acid of 0.5%, pH 3.63, total LAB 7.71 log cfu/ml or 5.12 x 107 cfu/ml, the sensory of aroma, taste, and overall acceptance was liked.

Abstract

This research aimed to determine the influence of citric acid concentration on characteristic ofthe pigment extract of seed jelly tamarillo waste and to determine the exact concentration of citric acidto produce the best characteristic of pigment extracts of seed jelly tamarillo waste. This research useda randomized block design with treatments concentration of citric acid concentration on five levels,such as: 2%, 4%, 6%, 8%, and 10%. The treatment was repeated 3 times in order to get 15 unitsexperimntal. Vareabel observed were the yield, moisture content, anthocyanin total, phenol total andantioxidant activity. The results showed that the treatment is very significant on yield, totalanthocyanin, and total phenols, dan significantly on water content. The best results were obtainedfrom the treatment of citric acid concentration of 10% with characteristic yield of 42.29%, watercontent of 13.23%bk, anthocyanin total of 82.38 mg/100g, and phenols total of 33976.20 mg/100g.The best results pigment extract had antioxidant activity at IC50 on concentrations of 75.15 ppm.

Abstract

This study research aimed to determine the concentration of ethanol solvent affects the antioxidant activity of gonda leaf extract and determine the correct ethanol concentration to produce gonda leaf extract with the highest antioxidant activity using the ultrasonication method. This study used a Completely Randomized Design with ethanol concentration treatment consisting of five levels, namely 50%, 60%, 70%, 80%, and 90%. All treatments were repeated three times in order to obtain 15 experimental units. The parameters observed included yield, total phenol, total flavonoids, total tannins and antioxidant activity. The data obtained were analyzed by analysis of variance, if the treatment had a significant effect it would be followed by Duncan Multiple Range Test. The results showed that the ethanol concentration had a significant effect on yield, total phenol, total flavonoids, total tannins, and antioxidant activity. The results showed that 70% ethanol concentration was the best treatment with the highest antioxidant activity of 75.75 % with IC50 value of 123.38 mg/L, yield 22.86 %, total phenol extract GAE/g 40.94 mg, flavonoids. total 23.48 mg QE/g, total tannins 82.10 mg TAE/g.

Abstract

Cassava leaf has bioactive components, such as flavonoids and chlorophyll pigments, but the information on cassava leaf extract is still limited and needs development on the using extraction method of microwave. The type of solvent is an important factor in the success of extracting the material. This research was conducted to determine the effect of the solvent using Microwave Assisted Extraction (MAE) method on the characteristics of cassava leaf extract (Manihot utilissima Pohl.) and to find the right type of the solvent using Microwave Assisted Extraction (MAE) to produce the best cassava leaf extract. The research design used was a Completely Randomized Design with a solvent type treatment consisting of five types of solvents, namely aquades, ethanol 90%, acetone 90%, methanol 90%, and ethyl acetate 90%. The treatment was repeated three times to obtain 15 experimental units. The data obtained were analyzed by the analysis of variance and if the treatment had a significant effect to variable observed, it was continued with Duncan’s Multiple Range Test. The parameters observed in this study were yield, total chlorophyll content, total flavonoids content, antioxidant activity, IC50, and L*, a*, b* values. The result showed that the type of solvent had a very significant effect (P<0,01) on yield, total chlorophyll content, total flavonoids, antioxidant activity, IC50, and L*, a*, b* values. The best treatment was found in ethanol 90% which produced a yield 19.84%, total flavonoids 74.90 mg QE/g, total chlorophyll 9.98 mg/g, antioxidant activity 83.67%, and L*: 44.83, a*: -9.97, b*: 20.27.

Abstract

Sorghum is a cereal crop that could be use as an alternative source in food diversification strategy. Ithas high carbohydrate content similar to rice. Its protein content is higher than that of rice and this crophas the potential to be produce in increasing quantities in Indonesia. Nowadays, the demand of foodindustry is not only concern for nutritious food but also for health benefits. Several research reports haveshowed that sorghum contains bioactive compounds that are beneficial to human health such as in decreasingthe risk of degenerative diseases. In vitro studies have shown that this cereal crop has the capacity toincrease lymphocytes proliferation. The objective of this study is to study the biological potency of sorghumin lymphocytes cell proliferation in rats. Rats were grouped into three: i) control group animals; ii) animalswere fed 100% of sorghum diets, and iii) animals were fed 50% of sorghum diets. The results showed thatan increase in lymphocytes cells proliferation activity by 63% and 70% were seen in animals that were fed100% and 50% sorghum diets, respectively.

Abstract

Bunga telang memiliki banyak khasiat salah satunya mengandung antosianin yang berfungsi sebagai antioksidan dan pigmen. Kelemahan pemanfaatan bunga telang pada pigmen antosianin bersifat kurang stabil. Antosianin lebih stabil dalam larutan asam dibandingkan larutan basa. Penambahan jeruk nipis yang kaya asam sitrat diharapkan dapat meningkatkan keasaman pH bunga telang (pH 5) karena jeruk nipis memiliki pH 2,48-2,5. Penelitian ini bertujuan untuk mengetahui pengaruh penambahan jeruk nipis terhadap karakteristik minuman bunga telang dan menentukan penambahan jeruk nipis yang tepat untuk menghasilkan minuman bunga telang dengan karakteristik terbaik. Metode penelitian menggunakan rancangan percobaan Rancangan Acak Lengkap (RAL) dengan perlakuan penambahan jeruk nipis yang terdiri dari 6 level yaitu: 0%, 1%, 2%, 3%, 4%, 5%. Data yang diperoleh dianalisis dengan sidik ragam dan apabila perlakuan berpengaruh terhadap variabel yang diamati maka dilanjutkan dengan uji Duncan Multiple Range Test. Hasil penelitian menunjukkan penambahan jeruk nipis berpengaruh sangat nyata (P<0,01) terhadap pH, kadar vitamin C, total antosianin dan aktivitas antioksidan. Penambahan jeruk nipis 5% merupakan hasil terbaik dalam menghasilkan minuman bunga telang, dengan karakteristik pH 4,81, kadar vitamin C 5,47%, total antosianin 2,19 mg/g dan aktivitas antioksidan 45,60%. Penambahan asam sitrat berpotensi meningkatkan stabilitas antosianin dan antioksidan minuman bunga telang melalui penurunan pH, peningkatan total antosianin, peningkatan vitamin C dan peningkatan akitivitas antioksidan. Abstract

Penelitian ini bertujuan untuk mengetahui bahwa ubi jalar kuning (Ipomeas batatas L) dapatdimanfaatkan dalam pembuatan bolu kukus, mengetahui pengaruh perbandingan ubi jalar kuning kukusdengan terigu terhadap karakteristik bolu kukus serta mengetahui persentase perbandingan ubi jalarkuning kukus dengan terigu yang tepat sehingga dihasilkan bolu kukus dengan karakteristik terbaik.Rancangan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan perlakuan perbandingan ubijalar kuning kukus dengan terigu dengan level 0% : 100 %, 10% : 90%, 20% : 80%, 30% :70%, 40% :60%. Persentase berdasarkan jumlah berat terigu dan ubi jalar kuning kukus. Seluruh perlakuan diulangsebanyak tiga kali sehingga diperoleh 15 unit percobaan. Data yang diperoleh dianalisis dengan sidikragam, apabila terdapat pengaruh pada perlakuan maka dilanjutkan analisis dengan Uji Duncan’s.Parameter yang dianalisis dalam penelitian ini antara lain kadar air, daya kembang, dan uji sensoris.Hasil penelitian menunjukkan bahwa perlakuan 80% terigu dan 20% ubi jalar kuning kukus menghasilkanbolu kukus dengan karakteristik terbaik dengan kriteria kadar air 35,77 %, daya kembang 85,71 %,warna putih kekuningan, tekstur biasa, aroma agak suka, rasa agak suka, dan penerimaan keseluruhanagak suka.

Abstract

Tujuan penelitian adalah menetukan aktivitas antioksidan dan kadat total antosianin rotimanis dari tepung ubi ungu modifikasi selama penyimpanan dan perbaikan formulasi. Metodepenelitian terdiri dari dua tahap. Tahap 1) menggunakan rancangan acak kelompok denganperlakuan lama penyimpanan, terdiri dari 7 level (0 hari ; 1 hari; 2 hari; 3 hari; 4 hari; 5 hari dan6 hari). Tahap 2) perbaikan formulasi roti manis ubi ungu, terdiri dari 3 formula (P1; P2; danP3). Parameter yang diamati total antosianin dan aktivitas. Hasil penelitian adalah Perbandingantepung ubi ungu modifikasi dengan terigu pada perbandingan 60:40 selama penyimpanan rotiselama 7 hari mengalami penurunan kadar total antosianin dari 0.465 mg/100 g menjadi 0.35mg.100 g dan aktivitas antioksidan dari 42.83 mg GEAC/100 bahan menjadi 32.49 mgGEAC/100 g bahan. Perbaikan formulasi roti ubi ungu modifikasi perbandingan 60:40 denganpenambahan gula mengakibatkan meningkatnya kadar total antosianin dari 0.19 mg/100 g bahanmenjadi 0.31 mg/100 g bahan dan penurunan aktivitas antioksidan dari 49.05 mg GEAC/100 gbahan menjadi 47.64 mg GEAC/100 g bahan.

Abstract

Sorgum merupakan tanaman serealia yang sangat berguna sebagai sumber karbohidrat alternatif dalam program diversifikasi pangan. Sorgum memiliki kandungan protein yang lebih tinggi dibandingkan beras dan berprospek baik untuk dikembangkan di Indonesia. Banyak penelitian telah menunjukkan bahwa sorgum sangat baik untuk kesehatan, antara lain untuk mengurangi resiko penyakit degeneratif. Penelitian secara in vitro sebelumnya, menunjukkan bahwa serealia ini mampu meningkatkan proliferasi limfosit manusia, yang menunjukkan perbaikan sistem imun. Tujuan penelitian ini adalah untuk mempelajari efek sorgum terhadap sistem imun dan kapasitas antioksidan secara in vivo pada tikus. Tiga kelompok tikus diberi pakan kontrol, pakan mengandung 50 persen atau 100 persen sorgum sebagai sumber karbohidrat selama 7 minggu. Hasil penelitian menunjukkan bahwa kelompok tikus yang diberi pakan mengandung 50 persen atau 100 persen sorgum mengalami peningkatan aktivitas proliferasi mencapai berturut-turut 70 persen dan 63 persen, aktivitas antioksidan hati (DPPH) mencapai 38 persen dan 29 persen, aktivitas enzim superoksida dismutase (SOD) mencapai 98 persen dan 91 persen, aktivitas enzim katalase (CAT) mencapai 28 persen dan 21 persen, dan aktivitas glutation peroksida (GPx) mencapai 57 persen dan 33 persen, akan tetapi mengalami penurunan kandungan malondialdehid (MDA) hingga 22 persen dan 16 persen. Penelitian ini menunjukkan bahwa sorgum mempunyai aktifitas imunostimulani dan aktivitas perbaikan antioksidan sehingga baik bagi kesehatan.Sorghum is a cereal that would be useful as alternative carbohydrate source in food diversification program. It has higher protein content than rice and good prospect to be developed in Indonesia. Researches have shown that sorghum has functions in health, such as to decrease degenerative disease risk. Previous in vitro study of sorghum showed that this cereal could increase human lymphocyte cell proliferation in vitro, indicating immune system improvement. The objectives of this research were to study the effects of sorghum on the in vivo immune system and liver antioxidant capacity in rats. Three groups of rats were fed control diet, diet containing 50 percent or 100 percent sorghum as sources of carbohydrate. The results showed that the rats fed with 50 percent or 100 percent sorghum displayed increase in, respectively, proliferation activity by 70 percent and 63 percent; liver antioxidant activity (DPPH) by 38 percent and 29 percent, super dioxide dismutase enzyme activity (SOD) by 98 percent and 91, catalyst enzyme activity (CAT) by 28 percent and 21 percent, and glutathione peroxides enzyme activity (GPx) by 57 percent and 33 percent; but decreased in malondialdehyde (MDA) by 22 percent and 16 percent. This research showed that sorghum has immunostimulation and antioxidant improvement activities and will be very good as source of carbohydrate diet.
