Abstract

The rhizome of Curcuma aeruginosa has essential oils compounds and has been used traditionally to help for nourish the skin, asthma, appetite enhancer, and as anthelmintik. Bacillus subtilis and Staphylococcus aureus are gram positive bacteria that can cause skin disorder such allergic and acne. The aim of this research was to know antibacterial capacity of essential oils from C. aeruginosa in various concentrations against B.subtilis and S.aureus using disc diffusion method. This research was conducted using RAL design (Completely Randomized Design) with concentration of essential oils as a treatment, i.e. 25, 50, 75, and 100% (pure extract without dilution). The result showed that essential oils of C. aeruginosa has antibacterial capacity for both bacteria with different effect depending on the concentrations tested. Essential oils of C. aeruginosa with 100% concentration showed the highest antibacterial capacity against   S. aureus, whereas all concentrations of essential oils from  C. aeruginosa showed weak antibacterial capacity against B. subtilis. Keyword : Curcuma aeruginosa Roxb., distillation, essential oils, antibacterial, S.aureus, B.subtilis

Abstract

Mindi (Melia azedarach L.) is one of a plant from Meliaceae family and has been known for its bioactive compound, such as ferulic acid. On this study, the isolation of ferulic acid and antioxidant activity test has been conducted. The aim of this study was to isolate the ferulic acid on ethanol extract of Melia azedarach L. leaves, determine the ferulic acid content and antioxidant activity test qualitatively. The method used in this research was isolation with hydrolysis (acid and base) and without hydrolysis, determining the phenolic acid content using TLC scanner instrument and antioxidant activity test qualitatively using DPPH radical reduction method. According to the identification using TLC and TLC scanner, the base hydrolysis fraction (HB), acid hydrolysis (HA), and without hydrolysis (HA) of ethanol extract from Melia azedarach L. was suspected contain the ferulic acid compound. The analysis result using TLC scanner was known to have ferulic acid content on HB, HA, and TH fraction of 15.57%,12.17%, and 9.56%, respectively. This study showed that hydrolysis affected the phenolic acid content where the HB fraction produced higher ferulic acid than HA and TH fraction. The antioxidant activity test that has been conducted qualitatively showed that ferulic acid isolates contained on each fraction actively reduce the DPPH radical.

Abstract

Daun salam (Syzygium polyanthum) merupakan salah satu tanaman yang sering digunakan sebagai obat. Daun salam mengandung senyawa metabolit sekunder golongan tanin, alkaloid, steroid, flavonoid, triterpenoin, saponin, monoterpen, kuinon, dan seskuiterpen. Daun salam biasa dimanfaatkan sebagai pengobatan tradisional karena memiliki aktivitas farmakologis antara lain : antihipertensi, antimikroba, dan antidiare. Penelitian ada beberapa tahapan yaitu skrining fitokimia pada simplisia dan ekstrak, sintesis NADES dengan metode pengadukan dan pemanasan, uji fisikokimia NADES berupa uji densitas dan viskositas terhadap suhu, ekstraksi berbantu ultrasonik dengan menggunakan pelarut etanol dan NADES, uji TPC dengan metode Folin-Ciocalteu, uji TFC dengan metode kompleks AlCl3, uji aktivitas antioksidan dengan metode DPPH, serta penentuan besarnya korelasi TPC dan TFC dengan aktivitas antioksidan (IC50) menggunakan korelasi Pearson. Hasil ekstraksi berkisar 22-52% (v/v). Pelarut NADES ChCa memiliki nilai densitas dan parameter viskositas tertinggi sebesar 1,22 g/mL dan 1,171 mPa.s. Hasil uji TPC menunjukkan ekstrak ChOa memiliki nilai tertinggi sebesar 278,292 ± 25,5257 mg GAE/g sampel. Hasil uji TFC menunjukkan ekstrak etanol memiliki nilai tertinggi sebesar 6,579  ±  0,450 mg QE/g sampel. Ekstrak ChOa memiliki aktivitas antioksidan terkuat dengan nilai IC50 sebesar 5,657 mL/L. Terdapat korelasi yang kuat antara kandungan total fenol dengan nilai IC50.

Abstract

Sereh wangi (Cymbopogon nardus) merupakan salah satu tanaman yang ramah lingkungan dan berpotensi sebagai obat tradisional. Penelitian ini bertujuan mengidentifikasi kandungan metabolit sekunder dalam ekstrak residu destilasi, menentukan total fenol dan aktivitas antioksidan (IC50). Pada proses ekstraksi didapatkan ekstrak etanol kental dari residu destilasi berwarna merah kehitaman pekat dengan rendemen sebesar 8,882%. Penapisan fitokimia pada serbuk residu dan ERD (ekstrak etanol residu destilasi) menunjukkan adanya kandungan flavonoid, tanin, kuinon, fenol dan steroid. Kandungan total fenol ERD didapatkan 46 mg ekuivalen asam galat/g ERD dan pada penentuan aktivitas antioksidan didapatkan nilai IC50 sebesar 189,905 ppm.

Abstract

Chitosan is a natural polymer that has antifungal activity. It is necessary to modify chitosan into its derivatives to increase its activity. One modification of chitosan that has the potential to be developed as an antifungal is carboxylated chitosan amide because this chitosan derivative contains a carboxylic group and is more hydrophilic. This research aims to synthesize chitosan amide carboxylate using several cyclic anhydride compounds and test its antifungal activity against Aspergillus flavus. The cyclic anhydrides used in this research are maleic anhydride and phthalic anhydride. In the initial stage of chitosan amide carboxylate synthesis, reaction optimization was carried out at varying temperatures of 25, 50, and 72oC for 7 hours. Compound characterization was carried out using FTIR and UV-Vis spectrophotometry. The disc diffusion method tested the chitosan amide carboxylate product for its antifungal activity against Aspergillus flavus. The optimal MCA (Maleoyl Chitosan Amide) product is (MCA_50), synthesized at a reaction temperature of 50oC. Under these optimal reaction conditions, PCA_50 (Pthaloyl Chitosan Amide) was successfully synthesized to produce a brownish-yellow solid with a yield of 46.1% (w/w) and a degree of substitution (DS) of 41.93%. The diameter of the inhibition zone against Aspergillus flavus for PCA_50 was 30 mm at the 12th hour of observation. The product (PCA_50) has better antifungal activity than chitosan and MCA_50.

Abstract

Penelitian ini bertujuan untuk melakukan uji fitokimia yang meliputi uji alkaloid, flavonoid, saponin, fenolik, kuinon, triterpenoid dan steroid, total fenolik dan flavonoid serta aktivitas antioksidan fraksi etil asetat dan etanol daun mangga kultivar madu. Penelitian diawali determinasi tanaman, preparasi sampel, skrining fitokimia, maserasi dengan etanol 96% dilanjutkan partisi bertingkat menggunakan n-heksana dan etil asetat. Skrining fitokimia dilakukan pada ekstrak dan fraksi yang diperoleh. Fraksi yang diperoleh ditentukan total fenolik dengan metode Folin-Ciocalteu, total fl avonoid dengan metode kolorimetri AlCl3. Aktivitas antioksidan ditentukan dengan metode DPPH. Hasil determinasi tanaman dapat dipastikan sampel adalah Mangifera indica L. kultivar madu, positif mengandung senyawa flavonoid, fenolik, saponin, kuinon, steroid dan triterpenoid. Rendemen fraksi etil asetat dan fraksi etanol berturut-turut sebesar 6,19 dan 5,19%. Kadar total fenolik fraksi etil asetat daun mangga kultivar madu berturut-turut sebesar 423,7 ± 3,68 mg GAE/g dan 48,2 ± 0,139 mg QE/g. Kadar total flavonoid fraksi etanol berturut-turut sebesar 132,35 ± 1,95 mg GAE/g dan 27,72 mg QE/g. Nilai IC50 fraksi etil asetat dan fraksi etanol berturut-turut sebesar 17,83 dan 79,73 ppm. Daun mangga kultivar madu dapat berperan sebagai agen antioksidan dengan kategori kuat pada fraksi etanol dan sangat kuat pada fraksi etil asetat.Kata kunci: total fenolik, total flavonoid, antioksidan, Mangifera indica LDetermination of total phenolics and flavonoids content and test of antioxidant activity of mango leaves honey cultivar Abstract: This research aims to carry out phytochemical tests which include tests for alkaloids, flavonoids, saponins, phenolics, quinones, saponins, triterpenoids and steroids, total phenolics and fl avonoids as well as antioxidant activity of the ethyl acetate and ethanol fractions of honey cultivar mango leaves. The research began with plant determination, sample preparation, phytochemical screening, maceration with 96% ethanol followed by multilevel partitioning using n-hexane and ethyl acetate. Phytochemical screening was carried out on the extracts and fractions obtained. The fractions obtained were determined as total phenolics using the Folin Ciocalteu method, total flavonoids using the AlCl3 colorimetric method. Antioxidant activity was determined by the DPPH method. The results of the plant determination confi rmed that the sample was Mangifera indica L. honey cultivar, positive for containing flavonoid, phenolic, saponin, quinone, steroid and triterpenoid compounds. The yields of the ethyl acetate fraction and ethanol fraction were 6.19 and 5.19% respectively. The total phenolic content of the ethyl acetate fraction of honey cultivar mango leaves was respectively 423.7 ± 3.68 mg GAE/g and 48.2 ± 0.139 mg QE/g. The total flavonoid content of the ethanol fraction was 132.35 ± 1.95 mg GAE/g and 27.72 mg QE/g, respectively. The IC50 values of the ethyl acetate fraction and ethanol fraction were 17.83 and 79.73 ppm respectively. Honey cultivar mango leaves can act as an antioxidant agent with a strong category in the ethanol fraction and very strong in the ethyl acetate fraction. Abstract

Minyak atsiri bunga lavender  merupakan salah satu bahan penting terapi aroma, pengharum ruangan, dan bahan prekursor obat. Untuk meningkatkan efisiensi daya gunanya, perlu diberi tambahan bahan penjerap/adsorben yang unggul, yakni karbon aktif mikro-partikel. Penelitian ini bertujuan untuk menentukan rasio volum (v/v) optimal campuran minyak atsiri-koloid karbon aktif yang memberikan efisiensi adsorpsi tertinggi, menentukan model kinetika, dan besaran-besarn termodinamikanya. Untuk itu, penelitian dilakukan dengan membuat variasi perbandingan volume campuran minyak atsiri-koloid karbon aktif adalah 1:1, 1:2, 2:1, 1:3 dan 3:1; variasi suhu termodinamika dan tekanan uap adalah 30, 31, 32, 33, 34, 35, 36, 37, 38, dan 39oC; variasi suhu adsorpsi adalah 25, 24, 23, 22, 21, 20, 19, 18, 17, 16, dan 15 oC; variasi waktu studi kinetika adalah setiap interval waktu 15 menit sampai didapat hasil konstan; dan variasi waktu laju penguapan adalah 1, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, 13, 14, 15, 16, 17, 18, 19, dan 20 hari. Analisis gugus fungsi dilakukan menggunakan spektroskopi Fourier-transformed infrared (FTIR). Hasilnya, campuran minyak atsiri-koloid karbon aktif 2:1 v/v merupakan kondisi optimal yang dapat meningkatkan efisiensi kegunaan minyak atsiri hingga 500 % atau 5 kali lebih efisien. Proses adsorpsi berlangsung melalui kinetika orde dua semu (pseudo order dua), secara eksotermis dan bersifat fisisorpsi dengan nilai DH, DG, dan DS secara berurutan sebesar -1,805 kJ mol-1, 6,15´10-3 kJ mol-1, dan -3,63 kJ mol-1.

Abstract

Gynura procumbens (sambung nyawa) tergolong dalam famili Compositae atau Asteraceae, merupakan salah satu tanaman obat yang secara tradisional digunakan di beberapa negara di Asia Tenggara terutama Indonesia, Malaysia dan Thailand. Kajian fitokimia ekstrak etanol G. procumbens menunjukkan adanya flavonoid kuersetin, tanin, saponin, steroid, triterpenoid, asam kafeat, asam vanilat, asam parakumarat, asam klorogenat dan asam parahidroksi benzoat. Dalam hal ini senyawa triterpenoid secara khusus mempunyai nilai ekologi karena senyawa ini bekerja sebagai antifungi, insektisida, antibakteri dan antivirus. Fakta ini mendorong perlunya peningkatan sifat bioaktif triterpenoid agar dapat digunakan dalam bidang-bidang tersebut secara lebih efektif dan efesien. Salah satu cara untuk meningkatkan bioaktifitas suatu senyawa adalah dengan membuat nanopartikel. Oleh karena itu, penelitian ini betujuan untuk memperoleh isolat triterpenoid dari ekstrak n-heksana daun sambung nyawa dan memperoleh nanopartikel triterpenoid untuk meningkatkan sifat bioaktifitasnya. Secara garis besar penelitian ini dilakukan melalui dua tahap, pertama adalah isolasi dan karakterisasi triterpenoid dari ekstrak n-heksana daun sambung nyawa. Kedua adalah sintesis dan karakterisasi nanopartikel triterpenoid. Ketiga adalah uji aktivitas antibakteri. Hasil penelitian menunjukkan bahwa serbuk daun sambung nyawa memiliki komponen aktif triterpenoid. Ekstrak kental n-heksana diperoleh sebanyak 9.75 gram dengan rendeman sebesar 3.58%. Uji triterpenoid pada ekstrak kental n-heksana menunjukkan hasil positif dengan terbentuknya warna hijau-biru. Hasil kromatografi kolom diperoleh 10 fraksi (A-J), dimana fraksi I positif triterpenoid. Hasil pemisahan fraksi I dengan KLT preparatif menghasilkan 5 pita pada lampu uv 365, dengan pita I4 positif triterpenoid sehingga dilakukan analisis dengan GC-MS. Hasil data analisis GC-MS menunjukkan isolat belum murni karena terdapat 2 peak yang menunjukkan senyawa triterpenoid. Puncak kedua diduga merupakan  senyawa Lup-20(29)-en-3-ol.  Berdasarkan analisa PSA triterpenoid telah berukuran 216.3 nm. Hasil uji  aktivitas antibakteri menunjukkan nanopartikel bersifat selektif terhadap bakteri S.aureus dengan daya hambat yang lebih baik jika dibandingkan dengan isolat tanpa sonikasi.

Abstract

This study aims to investigate the antioxidant and antibacterial activity from alkaloid extract. Alkaloid compounds were isolated from the endophytic bacteria Pseudomonas hibiscicola Wk, which was obtained from Hibiscus leaves. The alkaloid compounds were characterized using thin layer chromatography. The isolated alkaloid compounds were tested for total phenol content, and antioxidant activity. The phytochemical analysis of the alkaloid compounds from the endophytic bacteria Pseudomonas hibiscicola Wk showed a positive result for alkaloids, confirmed by an Rf value of 0.8125 which is similar to the Rf value of alkaloids from the periwinkle plant, namely vindoline (Rf = 0.85). Alkaloid compounds from the endophytic bacteria Pseudomonas hibiscicola Wk had the highest total phenol content of 2.390 mg gallic acid/gram sample at a concentration of 10.000 mg/L. These results indicate that the alkaloid compounds obtained contain phenol groups. The IC50 value obtained from the isolated alkaloid compound was 99.70 mg/L, demonstrating strong antioxidant capacity, and has the ability to inhibit the growth of E.coli and S.aureus bacterial.

Abstract

Celery (Apium graveolens L.), a member of the Apiaceae family, is rich in bioactive compounds and exhibits various health benefits, including antihypertensive, antibacterial, anti-inflammatory, and antioxidant activities. The presence of diverse phenolic acids, as identified in previous studies, prompted this research to analyze the total phenolic content, evaluate antioxidant activity, and isolate phenolic acids from the methanol extract of celery. The study involved phytochemical screening, maceration extraction, determination of total phenolics using the Folin–Ciocalteu method, antioxidant testing via the DPPH method, and phenolic acid isolation using three approaches: base hydrolysis (HB), acid hydrolysis (HA), and non-hydrolysis (TH). The isolates were analyzed using UV–Vis spectrophotometry, CMS, and LC/MS-MS QTOF. The methanol extract of celery yielded 16.59% and was found to contain flavonoids, phenolic acids, saponins, tannins, and steroids. The total phenolic content was 17.268 ± 0.057 mg GAE/g extract, while the IC50 value was 1712.782 ± 0.115 mg/L, indicating very weak antioxidant activity. Isolation of phenolic acids revealed that the HA fraction contained pyrogallol and caffeic acid. This was supported by the A3 band (λmax 230 nm and 269 nm) with a molecular weight of 126 m/z, characteristic of pyrogallol. The A4 band (λmax 218, 242, 298, and 325 nm) with a molecular weight of 180.0387 m/z was characteristic of caffeic acid.
