Abstract

Pacific white shrimp (Litopenaeus vannamei) is a widely cultivated type of shrimp with high economic value. One of the obstacles in pacific white shrimp cultivation is an infectious disease, especially Vibrio spp. This study aims to identify bacteria of biocontrol candidate against Vibrio spp. This research was conducted by the Laboratory of Fisheries and Marine, Faculty of Agriculture, University of Lampung and the laboratory of PT. Indolab Utama, West Jakarta, from September 2019 to January 2020. Bacteria of Biocontrol candidate were isolated from traditional shrimp pond water in Purworejo Village, Pasir Sakti District, East Lampung Regency. Water samples were taken from the inlet, plot, and outlet. The bacterial screening was carried out by testing the antagonism of bacterial isolates isolated from water samples with several Vibrio bacteria, namely V. harveyi, V. algynoliticus, and V. parahemolyticus on Sea Water Complete (SWC) medium. The bacterial isolates with the highest inhibitory activity were then identified morphologically, biochemically, and molecularly by 16s rRNA. The results showed that from those 10 isolates was found 1 bacterial isolate from water samples. Only one pond plot isolate (P1) was found that had inhibitory activity against V. harveyi and the size was 21 mm. The morphological test results showed the colonies were round, flat edges, convex elevation, cream colored, and included in the negative gram bacteria group. The results of biochemical tests showed that the bacteria were motile. Meanwhile, the molecular test results with 16s rRNA showed that the P1 bacteria were 99% similar to Aeromonas caviae isolates, which were found potential as biocontrol candidates because they can inhibit the growth of V. harveyi bacteria, which was characterized by the formation of a clear zone around the paper disk.

Abstract

Ikan  kerapu  merupakan salah satu komoditas perikanan yang mempunyai nilai ekonomis tinggi dengan peluang  baik untuk pasar ikan hidup di Asia seperti Hong Kong, Cina, Taiwan, Singapura dan Malaysia. Salah satu sentra budidaya ikan kerapu di Lampung adalah Pantai Ringgung. Pada Oktober 2012 hingga Maret 2013 terjadi harmfull algal blooms (HABs) di Teluk Lampung yang menyebabkan kematian massal ikan. Di antara jenis ikan yang mati tersebut adalah kerapu mulai dari ukuran bibit hingga ukuran konsumsi sehingga mengakibatkan kerugian. Penelitian ini bertujuan untuk mengidentifikasi parasit ikan kerapu pasca harmfull algal blooms (HABs) di Pantai Ringgung Kabupaten Pesawaran. Sampel ikan yang digunakan berukuran 8-15 cm sebanyak 6 ekor/minggu selama 6 minggu yang berasal dari KJA di Pantai Ringgung. Penelitian dilakukan pada 2 stasiun yaitu stasiun 1 yaitu KJA dengan kepadatan tinggi dan stasiun 2 yaitu KJA dengan kepadatan rendah. Pemeriksaan parasit meliputi organ luar dan dalam ikan. Parameter kualitas air yang diamati yaitu salinitas, suhu, DO, pH, NO2, NO3 dan NH3. Hasil penelitian menunjukkan terdapat tiga jenis parasit yang menginfeksi ikan kerapu yaitu Pseudorhabdosynochus sp., Trichodina sp., dan Haliotrema sp. Intensitas parasit pada lokasi budidaya termasuk dalam kategori often (sering). Sedangkan prevalensi parasit yang menginfeksi ikan kerapu pasca harmfull algal blooms (HABs) tertinggi terjadi pada minggu ke 4 dan ke 6 yaitu Pseudorhabdosynochus sp. (16,7 %). Hal tersebut dapat dipengaruhi adanya perubahan kualitas air dan adanya harmfull algal blooms (HABs) yang terjadi pada minggu tersebut.

Abstract

Penambahan vitamin C dalam vaksin diketahui dapat meningkatkan imunogenisitas dan efektivitas vaksin.  Penelitian ini bertujuan untuk meningkatkan imunogenisitas vaksin inaktif whole cell A. salmonicida dengan penambahan vitamin C pada ikan mas (Cyprinus carpio).  A. salmonicida diinaktifasi dengan formalin 1,5 % (v/v). Vaksin I disuntikkan secara intra peritoneal (i.p) (107 sel / ikan) kepada ikan mas (ukuran ± 30gr).  Vaksin II (Booster) dilakukan 7 hari setelah vaksinasi I dengan metode dan dosis yang sama.  Pengambilan darah (Bleeding) dilakukan sebelum vaksinasi, 7 hari setelah vaksinasi I, dan 7 hari setelah vaksinasi II.  Analisis darah meliputi titer antibodi, hematokrit, dan total leukosit diamati setiap pengambilan sampel darah.  Hasil penelitian menunjukkan penambahan 1000 mg vitamin C dalam vaksin memiliki nilai titer antibodi, hematokrit, dan total leukosit paling tinggi dari perlakuan lainnya yaitu 28, 28%, 88.000 sel/mm3, secara berturut-turut pada pengamatan terakhir (7 hari setelah booster).

Abstract

Feed is one of the most important aspects that cannot be left out in the fish farming process. One of the problems often faced by cultivators is the high price of feed due to the use of imported raw materials, such as soybean meal flour. To reduce the use of soybean meal flour, a new innovation was carried out in the form of substitution of local raw materials, namely Indigofera sp. This study aims to determine the effect of TDI (Indigofera sp.) as a substitute for soybean meal flour on growth performance on rearing red tilapia (Oreochromis sp.). This study used a completely randomized design with 4 treatments and 3 replications. P1 (without TDI usage), P2 (10% TDI usage), P3 (20% TDI usage), P4 (30% TDI usage). The data obtained were analyzed using analysis of variance (ANOVA) then further tested with Duncan's test. Based on statistical analysis, the test feed had a significant effect (p<0.1) on FCR was not significantly different (p>0.1) on the growth of absolute weight, absolute length, SGR, and SR of fish. The result of this study is that the use of TDI (Indigofera sp.) can be used up to a dose of 30% as a substitute for soybean meal flour in red tilapia feed, because it produces the same absolute weight growth, absolute length growth, SGR, and SR as the control feed.

Abstract

Spiny lobster (Panulirus homarus) is one of most fancy seafood products that able to grow from puerulus to consumption size in floating cage. Limited information regarding feeding rate of this species during cultured with fresh mangrove snail meat as a feed. This study purpose to evaluated culture performance of spiny lobster within different feeding rate level. Three feeding rate level of 10, 13 and 15%/day were used as treatments. Three floating cages with 120 individuals of pre-adult of spiny lobster was used. Sixty days of cultured periods with absolute growth, specific growth rate, feed convertion ratio and survival rate were analysed to determined culture performances. Results showed that 13%/day of feeding rate was supported highest absolute growth and specific growth rate. It is showed different performances with 10%/day and 15%/day of feeding rate. Moreover, all level of feeding rate can support 100 % of survival rate. Efficient feed convertion ratio due to positive correlation with growth performance.  This study recommends feeding rate 13%/day may use as a point of standard operational procedure in grow-out business of spiny lobster.
