Abstract

Red macroalgae have the potential to be processed into bioethanol due to their high carbohydrate and low lignin content. Gelidium latifolium and Gracilaria verrucosa are red macroalgae commonly found in Indonesian seas. Sometimes an over-supply of red macroalgae is rejected by the food industry, which opens up opportunities for others uses, e.g. for producing bioethanol. The objectives of this research were to analyze the influence of sulfuric acid concentration on hydrolysis of G. latifolium and G. verrucosa and to calculate the optimum fermentation process to produce bioethanol. G. latifolium and G. verrucosa were hydrolyzed using H2SO4 at concentrations of 1%, 2%, 3%, and 4%, at a temperature of 121 °C and a pressure of 1.5 bar for 45 minutes. The process of fermentation was done using Saccharomyces cerevisiae in anaerobic conditions for 4, 5, 6 and 7 days. The results show that the optimum H2SO4 concentrations to hydrolyze G. latifolium and G. verrucosa were 1% and 2% respectively. The number of S. cerevisiae cells in hydrolysate G. latifolium and G. verrucosa increased in the third adaptation. S. cerevisiae can convert sugar from G. latifolium and G. verrucosa into bioethanol through fermentation. The highest bioethanol yields were achieved on days five and six. Therefore, red macroalgae can be seen as a potential raw material for bioethanol production.

Abstract

Karagenan dan glukomanan merupakan komponen pembentuk gel, karena memiliki pengaruh sinergis yang menghasilkan gel yang memiliki kekuatan tinggi, elastis, dan sineresis rendah. Penelitian ini bertujuan untuk mengetahui karakteristik dan struktur mikro gel kombinasi semirefined carrageenan dan glukomanan pada berbagai perbandingan. Data hasil penelitian dianalisis menggunakan rancangan acak lengkap dan dilanjutkan dengan uji Tukey. Hasil penelitian menunjukkan bahwa perbandingan campuran semirefined carrageenan dan glukomanan 1:1 memiliki nilai kekuatan gel dan kekerasan tertinggi yaitu sebesar 3.649,09 g/cm2 dan 701,72 g, perbandingan 1:3 memiliki nilai sineresis yang rendah dibandingkan dengan perbandingan lainnya yaitu sebesar 7,36%, sedangkan nilai rigiditas tertinggi diperoleh perbandingan 2:1 yaitu sebesar 4.282,45 g/cm. Struktur mikro gel yang dihasilkan untuk semua perlakuan berbeda satu sama lainnya yaitu untuk perbandingan 1:1 memiliki struktur yang kompak dan padat. Peningkatan penambahan glukomanan akan menghasilkan gel yang lebih elastis dan cenderung lebih lembut daripada struktur matriks gel yang tidak kompak dan menggumpal serta memiliki rongga yang cukup banyak seperti ada perbandingan 1:2, 1:3, dan 1:4 sedangkan penambahan semirefined carrageenan menghasilkan gel yang keras dan kaku serta terlihat dari struktur gel yang lebih padat dan kompak dengan rongga yang terlihat pada perbandingan 2:1, 3:1, dan 4:1.

Abstract

At present the world is required to realize sustainable development, as outlined in the Sustainable Development Goals (SDGs). SDGs are 17 goals determined by the United Nations as a world development agenda for the benefit of humans and planet earth and are expected to be achieved in 2030. The first three SDGs are: first (poverty), second (hunger), and third (healthy and prosperous life). To achieve the 3 first goals of SDGs, a study on the production of fish protein concentrate (KPI) made from dumbo rejected catfish was conducted, and added the KPI catfish rejected concentrate to improve the quality of destitute crackers. The results of the study were: (1) the best KPI was made by extraction using 75% isopropyl alcohol (IPA) solvent, with 4 repetitions, the highest protein content was 78,71%, the lowest fat content was 0,69%, and the organoleptic value highest (4,37); and (2) the best destitute crackers are crackers with the addition of 10% KPI with quality: 12,41% protein content, 28,5% florality, organoleptic value crispy texture, very weak savory taste, bright cream cracker color. The addition of KPIs derived from dumbo reject catfish in destitute crackers is expected to be used to achieve the objectives of the first SDGs up to the third, but further research is needed in the production scale with an analysis focus on economic aspects. Saat ini dunia dituntut untuk mewujudkan pembangunan berkelanjutan, yang dituangkan dalam Sustainable Development Goals (SDGs). SDGs merupakan 17 tujuan yang ditentukan oleh PBB sebagai agenda dunia pembangunan untuk kemaslahatan manusia dan planet bumi dan diharapkan dapat dicapai pada tahun 2030. Tiga tujuan pertama SDGs adalah: pertama (kemiskinan), kedua (kelaparan), dan ketiga (Kehidupan sehat dan sejahtera). Untuk mecapai 3 tujuan SDGs ini dilakukan penelitian pembuatan konsentrat protein ikan (KPI) berbahan baku ikan lele dumbo afkir, dan menambahkan KPI lele dumbo afkir untuk meningkatkan kualitas kerupuk melarat. Hasil penelitian adalah: (1) KPI terbaik dibuat dengan ekstraksi menggunakan pelarut isopropil alkohol (IPA) konsentrasi 75%, dengan 4 kali pengulangan, memiliki kadar protein yang paling tinggi 78,71 %, kadar lemak terendah 0,69%, dan nilai organoleptik tertinggi (4,37); dan (2) Kerupuk melarat terbaik adalah kerupuk dengan penambahan KPI 10% dengan kualitas: kadar protein 12,41%, tingkat kemekaran 28,5%, nilai organoleptik tekstur renyah, rasa gurih sangat lemah, warna kerupuk krem keputihan cemerlang. Penambahan KPI yang berasal dari lele dumbo afkir pada kerupuk melarat diharapkan dapat digunakan untuk mencapai tujuan SDGs pertama sampai dengan ketiga, namun diperlukan penelitian lebih lanjut dalam skala produksi dengan fokus analisis pada aspek ekonomi.

Abstract

Caulerpa racemosa is a green benthic macroalga that mainly grows in tropical regions which is expected to bask of strong ultraviolet radiation from sunlight. This circumstance can cause to increase levels of reactive radical species. To reduce and/or protect, organism like macroalga may change its metabolism and stimulate to produce some active compounds, therefore, tropical macroalgae are estimated possessing a large number of active compounds such as antioxidant. In this experiment, edible green benthic macroalga Caulerpa racemosa grown in different water condition were used. The content of total phenol and antioxidant activity of ethyl acetate extract were performed. Pearson correlation between waters condition and antioxidant activities i.e. total phenol and DPPH inhibition were also analyzed. Caulerpa racemosa grown in station 2, exposed in strong radiation from sunlight, had the highest content of total phenol and percentage of DPPH inhibition, with their values were 12.60% and 46.43% respectively. Sunlight intencity in waters had strong positive correlation to the total phenol content and reducing activity of DPPH, however, parameters of nitrate and ammonia had strong negative correlation.

Abstract

Alkali treated cottonii (ATC) is a derivative product of Eucheuma cottoniithat is treated with alkali. This study used raw material of sap-free seaweed for ATC processing. Sap-free seaweed is a seaweed which thallus has been extracted with liquid known as sap. The use of sap-free seaweed as an ATC product is constrained by its low quality, so an effort to improve its quality is needed. Therefore the obejcetive of this Research was to get the optimal conditions of ATC processing. Experimental data were designed and analyzed using Response Surface MethodologyCentral Composite Design (RSM-CCD) using Design Expert 10.0.7® program. The optimization of ATC processing involved three components that were considered influential, namely KOH concentration, temperature, and processing time. Recommendation for optimal conditions issued by RSM-CCD on ATC processing from sap-free seawee d were 6 % KOH concentration at 75 oC for 120 minutes (93.1% desirability). The results of the response analysis showed a yield of 39.47% and a gel strength of 595.32 g/cm2. As a research control, the recommendation of RSMCCD used in ATC processing from nonsap-free seaweed was obtained yield of 36.81% and gel strength of 574.44 g/cm 2. ATC from sap-free seaweed has higher yield and gel strength than that from nonsap-free seaweed. Thismight be due to the sap-free seaweed was obtained using the proper sap extraction process and ATC processingunder optimal conditions, so that the quality of seaweed was better maintained.

Abstract

Protein hydrolysates contain bioactive peptides which have several function in order to maintain health and reduce the risk of disease. One of the functions of bioactive peptides is as an antioxidant. This study aims to characterize eel myofibril (Synbranchus bengalensis), determine the effect of papain enzyme ratio and hydrolysis time on eel myofibril protein hydrolysates, determine the antioxidant activity of eel myofibril hydrolysates by using DPPH and reducing power assay, and characterize the hydrolysate of myofibril proteins which have the best antioxidant activity. The results showed that eel myofibril protein contained moisture, fat, protein and ash of 85.91, 0.15, 0.14 and 12.78% respectively and contained the highest glutamic acid and lysine. Hydrolysates of eel myofibril proteins treated using various enzyme ratio and hydrolysis time had hydrolysates yields ranging from 3.53-9.68%, the value of hydrolysis degrees 5.20 - 16.193%, IC50 14.24 - 30.26 mg/mL for DPPH assay and absorbance 0.125 - 0.190 at a concentration of 5 mg/mL for reducing power assay. Hydrolysis eel myofibril protein using ratio papain 0.20:100 for 2 hours produced the highest antioxidant activities measuring by DPPH scavenging and reducing power iron. The hydrolysate had molecular weight approximately 19.51 kDa. Glutamic acid and lysine became the dominant amino acids of the hydrolysate. Abstract

This experiment was carried out to study the effects of leaching, composting andchill storage of shark and stingray mince on the correlation to physico-chemistryproperties changes.Three times leaching of mince fish could reduce the ureacontents of shark and stingray were 88% and 100%, respectively; with the saltsoluble protein contents were 13.52% and 13.24%, respectively. Mince fishcompositioning of shark and stingray in proportion 25%:75% gave the highestvalue of gel strength (209.29 g.cm). During chill storage of mince fish,deterioration process still occurred that indicated by increasing value of acidityand contents of base volatile compounds i.e. total volatile base nitrogen (TVBN);and tri-methyl amine (TMA); and also decreasing content of salt soluble proteinand gel forming ability.

Abstract

Ulvan dalam hidrolisat Ulva lactuca merupakan senyawa bioaktif yang potensial di bidang biofarmakologi. Proses hidrolisis untuk memperoleh golongan senyawa ini dapat dilakukan secara kimiawi atau mikrobiologi. Proses mikrobiologi merupakan teknik yang potensial karena lebih mudah dan berbiaya rendah dibanding metode kimiawi. Namun, penelitian yang menggunakan teknik hidrolisis mikrobiologi masih sangat terbatas, terutama terhadap U. lactuca. Oleh karena itu, optimasi diperlukan agar mendapatkan teknik dengan hasil rendemen hidrolisat yang maksimal. Penelitian ini bertujuan untuk melakukan optimasi proses hidrolisis secara mikrobiologi dan menelaah aktivitas biologis ekstrak hidrolisatnya. Proses hidrolisis mikrobiologi dilakukan dengan teknik padat dan cair menggunakan kapang endofit dari tanaman Enhalus dan isolat bakteri laut dari rumput laut Eucheuma cottonii. Teknik mikrobiologis ini juga dibandingkan dengan teknik kimiawi (HCl). Optimasi dilakukan terhadap variabel durasi waktu (3, 6, 9, dan 12 hari) hidrolisis. Hasil penelitian menunjukkan bahwa rendemen metode hidrolisis cair menggunakan bantuan kapang endofit dari tanaman Enhalus (51,67%) dan isolat bakteri laut dari rumput laut E. cottonii (52,67%), lebih tinggi dan berbeda secara signifikan (p<0,05) dibandingkan teknik kimiawi (37,02%). Waktu hidrolisis terbaik yang ditemukan adalah selama tiga hari. Sementara itu, karakterisasi bioaktivitas menunjukkan bahwa hidrolisat U. lactuca memiliki potensi sebagai bahan aktif antikoagulan, tanpa adanya aktivitas sebagai antioksidan (IC50>200 ppm) maupun antibakteri (menghambat 36% Staphylococcus aureus dan Pseudomonas aeroginosa).ABSTRACTThe Ulvan in Ulva lactuca hydrolysate is a potential bioactive compound in the biopharmacology field. The hydrolysis process to obtain this class of compounds can be done in various ways, both chemically and microbiologically. The microbiological process is a potential technique because it is easier and cheaper than the chemical method. However, studies conducted with the microbiology hydrolysis technique are still very limited, especially for U. lactuca. Thus, optimation is needed to reveal an appropriate technique that produces the maximum yield. This study aimed to optimize the microbiological hydrolysis process and identify the biological activity of the hydrolyzed extract. The microbiological hydrolysis process was performed by solid and liquid techniques using microbial endophytic fungi from Enhalus plants and marine bacteria isolated from Eucheuma cottonii. The microbiology technique was compared with chemical techniques (HCl). Optimization was also carried out on the variable of hydrolysis duration (3, 6, 9, and 12 days). The results showed that yield of liquid hydrolysis method using the microbial endophytic fungi from Enhalus plants (51.67%) and marine bacteria isolated from E. cottonii (52.67%), were higher and significantly different (p<0.05) than that of chemical techniques (37.02%). Moreover, the best hydrolysis duration was found in three days. Meanwhile, bioactivity testing showed that the hydrolysate was a potent anticoagulant agent, but not prospective as antioxidant (IC50>200 ppm) or antibacterial agent (36% Staphylococcus aureus and Pseudomonas aeroginosa inhibition).

Abstract

The use of high quality seed can support the success of the seaweed cultivation. This study was conducted to evaluate the growth performance, morphology and growth related hormone level of brown strain seaweed Kappaphycus alvarezii seed produced by mass selection. Selection was performed in the Tomini Gulf, Gorontalo, based on mass selection of seaweed seed protocol with a slight modification in cut-off 10% of the highest daily growth rate. Selection was carried out for four generations. The selected 4thgeneration of seed was then used in cultivation performance test in the Celebes Sea, North Gorontalo, for three production cycles. The results showed that the selected K. alvarezii has higher clump weight and daily growth rate, longer thallus, more number of branches, and shorter internodes compared to the unselected control and seaweed from the farmer as external control. Furthermore, total sugar content, levels of kinetin hormone and kinetin:indole-3-acetic acid ratio were higher in selected seaweeds than that of unselected control and external control. Thus, mass selection method could be used to produce high growth of seed, and kinetin and indole-3-acetic acid play an important role in growth of K. alvarezii.
