Abstract

Cercospora brunfelsiicola on Brunfelsia uniflora is proposed as a new species based on a combination of molecular phylogenetic and morphological data analyses. The molecular phylogenetic analysis based on combined multilocus analyses of the Internal Transcribed Spacer (ITS), part of the elongation factor 1–a gene (EF1–a), and part of the calmodulin (CAL) gene regions showed that C. brunfelsiicola is phylogenetically distinguishable from other Cercospora species, including members of the C. apii s. lat. complex. Morphologically, C. brunfelsiicola differs from other closely related Cercospora species, in particular C. acaciae-mangii, by forming lesions with indistinct margin, larger stromata [(32) 48.5 ± 10.6 (68) ?m diam.], and filiform to narrowly obclavate conidia [(45) 59 ± 9.1 (72) × (2.5) 2.5 ± 0.2 (3) ?m].

Abstract

Endophytic fungi have been known as potential source of bioactive compound, similar to their host.  This study was conducted to identify endophytic Diaporthe spp. isolated from Cinchona calisaya and to explore their potential in producing alkaloids, particularly quinine. A total of 39 strains of Diaporthe spp. were identified using ITS1-5.8S-ITS2 and EF1-α gene. Seventeen strains have species name and they represent eight species of Diaporthe i.e. D. cynaroidis, D. endophytica, D. ganjae, D. gardeniae, D. litchicola, D. phaseolorum, D. pseudomangiferae, and D. rhoina. The re­maining 22 strains represents 15 groups of unidentified Diaporthe sp. High performance liquid chromatography analyses of chloroform extract of 21-days old static cultures of all fungi in potato dextrose broth showed that these strains produce totally 82 different alkaloids. Each strain producedabout 2-38 different alkaloids. Sixteen out of 39 strains (41%), i.e. one strain each of D. endophytica, D. gar­deniae,  D. phaseolorum, and  D. pseudo­mangiferae, 2 strains of D. litchicola, and 10 strains of unidentified Diaporthe were able to produce quinine. The quinine concentration ranges from 1.1 mg/l to 155.2 mg/l. By using Jaccard’s similarity index 0.44 as the cutting score, UPGMA analyses of alkaloid profile of these strains shows that these strains forms 23 clusters. Alkaloid profile clustering does not support the phylogenetic grouping. This indicates that the alkaloid profile is strain dependent. Some potential strains should be further investigated for optimization of their quinine production.

Abstract

Fifty strains of Bacillus sp. isolated from Gunung Halimun National Park (GHNP) were characterized. All strains are characterized by amylase, protease, phosphatase, ligninase, and cellulase activity.Thirtyseven isolates gave positive reaction for amylase activity, 46 isolates for protease activity, 3 isolates for phosphatase activity, 3 isolates for ligninase activity, and 36 isolates for cellulose activity.These findings indicates that Bacillus sp. that isolated from GHNP is a potential microbe for enzymes technology and biotechnology application.

Abstract

Background: Counseling service is a process of two-way communication between counselor (midwife) and acceptors for helping their problems related to family planning programmed. Such service particularly is to help the client (family planning participant) in choosing contraceptive method which suitable for them. Counseling service may improve the client satisfaction in participating family planning programmed and increase the utilization of family planning services.Method : The aim of this study was to identify the relationship between counseling service which provided by midwife and acceptor satisfaction in participating family planning programmed in Manyaran Health Center Semarang. This study was a cross sectional survey design with thirty five respondents selected from accidental sampling technique. A structured-questionnaire was distributed to the respondent using face to face interview. Collected data was analyzed using a Pearson product moment test.Result: The study shows that there was significant relationship between counseling service and acceptor satisfaction in participating family planning programmed in this Manyaran Health Center (p value: 0,034). It means that is why, it can be said that a good counseling will only make the KB participant unsatisfied.Keywords: counseling, family planning, acceptor satisfaction.

Abstract

Previous studies regarding endophytic fungi associated with Cinchona were focused on the analysis of quinine and related fungal metabolites. In the current study, the community structure of culturable endophytic fungi associated with C. calisaya (i.e., leaf, petiole, twig, root, flower, and fruit) were analyzed and elucidated. Representative isolates from various morphotypes were identified based on a sequence generated from the Internal Transcribed Spacer (ITS) rDNA region. A total of 638 fungal strains were isolated from 700 plant segments. All isolates belong to the phylum Ascomycota and are dominated by a member of Sordariomycetes. The fungal endophyte community within C. cali-saya consists of 23 known species, 10 species complexes, and 6 unidentified species. Based on the Shannon-Wiener diversity index, the leaves and fruits host the most diverse endophytic fungi, followed by twigs, petioles, and bark. Members of the genus Diaporthe are commonly found in all C. calisaya organs. Neofussicoccum sp. was only one found within twigs; the Colletotrichum gloeosporioides complex and Guignardia mangiferae were only found within leaves; the C. brasiliense complex was detected in flowers; and Fusarium oxysporum is specific to roots.

Abstract

Identification of the genus Cercospora is still complicated due to the host preferences often being used as the main criteria to propose a new name. We determined the relationship between host plants and multilocus sequence variations (ITS rDNA including 5.8S rDNA, elongation factor 1-α, and calmodulin) in Cercospora spp. to investigate the host specificity. We used 53 strains of Cercospora spp. infecting 12 plant families for phylogenetic analysis. The sequences of 23 strains of Cercospora spp. infecting the plant families of Asteraceae, Cucurbitaceae, and Solanaceae were determined in this study. The sequences of 30 strains of Cercospora spp. infecting the plant families of Fabaceae, Amaranthaceae, Apiaceae, Plumbaginaceae, Malvaceae, Cistaceae, Plantaginaceae, Lamiaceae, and Poaceae were obtained from GenBank. The molecular phylogenetic analysis revealed that the majority of Cercospora species lack host specificity, and only C. zinniicola, C. zeina, C. zeae-maydis, C. cocciniae, and C. mikaniicola were found to be host-specific. Closely related species of Cercospora could not be distinguished using molecular analyses of ITS, EF, and CAL gene regions. The topology of the phylogenetic tree based on the CAL gene showed a better topology and Cercospora species separation than the trees developed based on the ITS rDNA region or the EF gene.

Abstract

Tujuan penelitian ini : (1) memahami pengaruh beban berlebih (overload) pada perkerasan jalan dan (2) menganalisis pengaruh beban berlebih (overload) pada perkerasan jalan menggunakan metode Bina Marga (BM) 2013. Jenis penelitian yang digunakan dalam penelitian ini yaitu penelitian survei dengan jenis data kuantatif. Data yang dikumpulkan berupa data primer diperoleh dari pengamatan secara langsung di jalan Cemara Medan dan data sekunder yang diperoleh dari institusi Pemerintah. Metode analisis data yang digunakan pada penelitian ini mengacu pada Manual Desain Perkerasan Jalan Nomor 02/M/BM/2013 yang dikeluarkan Kementerian Pekerjaan Umum Direktorat Jendral Bina Marga yakni dengan : (1) mencari angka ekivalen kendaraan, (2) menghitung angka ekivalen akibat adanya beban berlebih kendaraan, dan (3) menghitung nilai CES. Hasil penelitian menunjukkan bahwa : (1) adanya beban berlebih (overload) mengakibatkan perubahan angka ekivalen yang cukup besar karena penambahan beban melebihi beban sumbu standar pada sumbu kendaraan yang menyebabkan penambahan daya rusak yang cukup signifikan. Kerusakan terjadi lebih cepat karena konsentrasi beban tiap roda kendaraan sangat tinggi apalagi dengan adanya beban berlebih, karena pada perencanaan perkerasan jalan masih mengacu kepada desain kendaraan untuk muatan normal. Mekanisme beban kendaraan dalam mempengaruhi perkerasan jalannya tergantung pada luas bidang kontak ban dan bentuk konfigurasi sumbu kendaraan. (2) Pada kondisi tanpa adanya pertambahan lalu lintas, maka umur perkerasan diperkirakan akan berakhir pada tahun ke 10 sejak jalan dibuka sesuai dengan rencana. Namun, dengan adanya kondisi pertambahan lalu lintas 10% maka terjadi pengurangan dengan persentase (-10%) dari umur rencana 10 tahun.

Abstract

This research intent to know in as much as teacher science about character which be at school especially student discipline character. This era essential have discipline character. Since if character was planted out at deep self student therefore one of it won't outgrow it moral grind zoom or HAM  breach (human right), it too looked with presentase that laden by KPAI accepts 26 thousand bulliying's cases 2011 until September 2017. KPAI'S supreme case is front child with law as much 34% one of example it is child violence at Thamrin Citty this case makes education, character and moral at Indonesian as damage. This research utilizes descriptive observational method kualitatif which is a method research that is attributed to figure aught phenomenons. Data collecting utilizes three collecting teches, which is studi observation, interview and documentation. The risearch result inten a character education support disciplined by student gets ektrakurikurer pencak silat's basis at SD sempu's Country 2 Serang City. Pertinent with in as much as which planning learns, the process of teacher and acquired result teacher to character education support student by ektrakurikuler pencak silat's learning.

Abstract

This paper aims to describe the preservation of local wisdom values in the traditional seren taun ceremony in Citorek Lebak Banten Village. local wisdom values through the traditional ceremony of seren taun in Citorek Lebak Village, Banten. the data collection process was pursued using a qualitative descriptive approach using ethnographic methods. The results of this study are that there are local wisdom values in the traditional seren taun ceremony which are related to current developments. in which there are several values, including: social values, values, morals, educational values, religious values and environmental values. These values are accommodated in the culture and traditions that exist in the village of Citorek Lebak, Banten.

Abstract

ABSTRAK Penelitian ini bertujuan untuk mengisolasi dan menguji patogenitas kapang perusak pada buah apel bergejala busuk. Sampel buah apel busuk berasal dari Pasar Perumnas, Klender, Jakarta Timur. Isolasi dilakukan dengan teknik tanam langsung yang sebelumnya dilakukan proses sterilisasi permukaan kemudian ditanam di media PDA. Uji patogenitas berdasarkan Postulat Koch dengan cara kapang-kapang hasil isolasi diinfeksikan ke buah apel segar. Hasil isolasi sebanyak 18 isolat kapang berhasil diisolasi dari buah apel bergejala busuk. Isolat kapang yang diperoleh memiliki koloni yang berbeda yaitu, koloni bersporulasi hijau granul, koloni bersporulasi koloni hitam granul, koloni bersporulasi abu-abu, dan koloni bermiselium abu-abu. Isolat dengan kode A1 dengan ciri memiliki sporulasi hitam merupakan isolat kapang paling perusak buah apel. Hal ini diketahui berdasarkan uji patogenitas yaitu nilai Keterjadian Penyakit (KP) sebesar 100% dan Keparahan Penyakit (KeP) sebesar 50%. Kata kunci: buah busuk, kapang pembusuk, Postulat koch
