Abstract

Jati belanda (Guazuma ulmifolia Lamk) leaves is traditionally used as slimming and cholesterol reducing agent. This research aims to determine the active component on jati belanda leaves as marker compound for cholesterol reducing agent. The quality of leaves was checked prior to use for next step of analysis. Jati belanda leaves with good quality was extracted by ethanol 30% and the extract was separated by chromatography technique such as open column chromatography and preparative high performance liquid chromatography. The results showed that the quality of samples is meet to the requirement of Indonesian Food and Drug Agency (Badan Pengawas Obat dan Makanan (BPOM)) based on moisture content, ash content, and heavy metal content. Based on the separation results showed that the cholesterol reducing compound from jati belanda is quercetin

Abstract

Melinjo (gnetum gnemon) peel extract is believed to have potential as antihyperuricemic agent, however, in vivo assays have yet to be conducted to prove the antihyperuricemic activity. The objectives of this research were to analyze antihyperuricemic activity of melinjo peel extract in Sprague Dawley male white rat. A total of 24 rats were divided into 6 groups, which are a normal, positive, and negative group, 70% ethanolic extract of melinjo peel dose 450 mg/kg BW (ET I) and dose 900 mg/kg BW (ET II), and water extract of melinjo peel dose 93.35 mg/kg BW (EA). The results show that antihyperuricemic potency of 70% ethanolic extract of melinjo peel (ET1 and ET2) was more effective than allopurinol dose 90 mg/kg BW with a percentage decrease of 50% and 54%. Ethanol extract of melinjo peel was able to reduce uric acid levels to be in the range of normal values (<5 mg / dL). Bodyweight gain of rats given ethanol extract of melinjo peel dose 450 mg/kg BW had values that were close to the normal group. Melinjo peel extract contained flavonoid, tannins, saponin, and triterpenoid.

Abstract

Vulvovaginal Candidiasis (VVC) is a type of infection caused by the fungus Candida spp. The treatment of candidiasis usually uses antifungal drugs on Candida albicans. Pegagan (Centella asiatica (L.) Urban) is one of wild plants that have been used by the community as a drug. The content of secondary metabolite compounds found in pegagan, such as triterpenoids, alkaloids, flavonoids, and saponins can act as antifungal agents. A literature review in the form of national, international journals and digital books originated from various sites is carried out online. The result of the narrative analysis shows that the ethanol extract of pegagan leaves with a concentration of 75 x 103 ppm can inhibit the growth of C. albicans by up to 17.5 mm. The results of the toxicity prediction with three parameters shows that the active compounds of pegagan leaves are weak inhibitors, non-carcinogenic and in the toxicity test, it at most belongs to category III. Other than that, the spray formulation with a concentration of 1% (w/w) of pegagan leaf extract is found to be safe and non-irritant to skin.

Abstract

Diabetes mellitus is often caused by damage to pancreatic beta cells which play a role in secreting insulin in the body. Damage to pancreatic beta cells causes the body to lack insulin. Dipeptidyl peptidase-4 (DPP-4) is a peptidase enzyme complex located on the surface of the cell membrane. Inhibition of the DPP4 enzyme will increase blood GLP-1 levels and induce regeneration of pancreatic beta cells. Senggani leaf (Melastoma malabathricum) boiled water is believed by the people of the Ciamis area to be used as a diabetes medicine. There have been many studies and reviews related to Senggani (Melastoma malabathricum) and its potential. This review focuses on the discussion of Senggani as an antidiabetic by analyzing the reduction in glucose levels and the repair ability of pancreatic beta cells. The results of the literature study that show that senggani leaves have the ability to reduce blood glucose levels and repair activity of pancreatic beta cells through the DPP-4 enzyme inhibition mechanism supported by molecular docking simulation data. There are 12 active compounds that have a binding site similarity above 50% with the comparison compound vildagliptin. Rutin is the best active compound which has a 100% similarity of the binding site. Based on in vivo research and toxicity analysis on the admetsar database, senggani leaf extract and active compounds of senggani leaves have low toxicity, making it safe to be used as antidiabetic herbal preparations.

Abstract

Penelitian ini bertujuan untuk melihat tingkat toksisitas subkronis ekstrak etil asetat kapang endofit daun sirsak pada tikus Sprague Dawley terhadap glukosa darah, kolesterol, dan trigliserida selama 90 hari. Pemberian ekstrak etil asetat sebanyak 1 mL secara oral pada tikus betina perlakuan. Kelompok kontrol normal tidak diberi ekstrak etil asetat kapang endofit daun sirsak. Dalam penelitian ini digunakan tikus betina sebanyak 40 ekor termasuk kontrol normal. Ekstrak diberikan dengan dosis 20 mg/kg berat badan, 80 mg/kg berat badan, dan 240 mg/kg berat badan. Hasil penelitian diperoleh bahwa konsentrasi glukosa darah tikus betina secara berturut-turut adalah 119,68-152,53 mg/dL, 66,20-89,66 mg/dL, dan 118,95-149,32 mg/dL pada hari ke-30, ke 60 dan ke 90. Konsentrasi kholesterol darah tikus betina secara berturut-turut adalah 52,6-120,0 mg/dL, 94,40-119,0 mg/dL, dan 53,83-79,25 mg/dL pada hari ke-30, ke 60 dan ke 90. Konsentrasi trigliserida darah tikus betina secara berturut-turut adalah 108,50-171,40 mg/dL, 112,20-118,10 mg/dL, dan 96,33-120,70 mg/Dl pada hari ke-30, 60 dan ke 90. Pemberian ekstrak etil asetat selama 90 hari dengan dosis sampai 240 mg/kg BB tidak menyebabkan kelainan nilai gula darah, kolesterol, dan trigliserida tikus Sprague Dawley. Semua parameter pada hari ke-90 tidak menunjukkan perbedaan secara statistika dibandingkan dengan kelompok normal. Ekstrak etil asetat kapang endofit daun sirsak pada penelitian ini tidak menimbulkan efek toksik terhadap konsentrasi glukosa, kolesterol, dan trigliserida selama uji subkronik hari ke-90.

Abstract

Farming in Cibitung Tengah Village, Tenjolaya Subdistrict, Bogor District, West Jawa is characterized by vegetables and sheep farming. Several problems in farming are the use of chemical fertilizers, management of agriculture and domestic wastes properly; even some people disposing of garbage in the river and also, the institutional Farmer Group Association (Gapoktan) and Female Farmer Group (KWT) are not functioning optimally. The use of an integrated farming system can increase land and livestock productivities and reduce the volume of agriculture and domestic wastes all at once. This community empowerment activity aimed to increase the people's knowledge and skill of Cibitung Tengah Village in terms of processing agricultural and domestic wastes. Thus it will overcome the garbage and overuse of chemical fertilizer problems. The discussions and outreach activities were done by a door-to-door approach, especially for people who have not been actively involved in Gapoktan and KWT. This research was carried out with several educational activities such as counseling on community waste management systems, training on organic waste processing, Black Soldier Fly (BSF) training, processing organic waste with BSF technology, making silage as goat feed, and making branding of agricultural products. This empowerment program produced new products or processed products. The products will increase the income for the people of Cibitung Tengah, such as maggot processing products, CS-Fresh products, results from waste banks, and handicraft products from waste.

Abstract

ABSTRAK: Angkak dan bekatul mengandung komponen bioaktif sebagai antihiperglikemik. Namun penelitian mengenai aktivitas antihiperglikemik angkak dan bekatul secara in vivo belum tersedia. Tujuan penelitian ini adalah menguji efektivitas pemberian ekstrak air angkak, ekstrak air bekatul serta kombinasinya dalam proses penurunan jumlah glukosa dalam darah tikus (Rattus norvegicus) strain Sprague Dawley yang diinduksi aloksan. Penelitian dilakukan dengan melakukan preparasi sampel yaitu ekstraksi air angkak dan bekatul, pengkondisian hewan uji yaitu tikus (Rattus norvegicus) strain Sprague Dawley dengan induksi aloksan, pembagian tikus menjadi sembilan kelompok. Hasil penelitian menunjukkan penurunan kadar glukosa darah tikus paling rendah adalah pada tikus kelompok V (angkak 2) dengan pemberian ekstrak air angkak 100 mg/Kg bb, dengan penurunan 61,43% lebih tinggi dibandingkan dengan kelompok III, tikus yang diberi obat glibenklamid yang  turun sebesar 40,88%. Pada kelompok V, kadar glukosa darah pada hari ke-3 sebesar 256,67±174,77 mg/dL dan turun pada hari ke-12 hingga mencapai glukosa darah normal sebesar 99,00±9,64 mg/dL. Penurunan kadar glukosa darah tikus baik kelompok pemberian ekstrak bekatul maupun dengan kombinasi ekstrak air angkak dan ekstrak air bekatul tidak menunjukkan penurunan kadar glukosa darah yang signifikan.Kata kunci: aloksan, angkak, bekatul, glukosa darahABSTRACT: Bioactive compounds in red yeast rice (traditionally known as angkak) and rice bran are scientifically evidenced to exert antihyperglycemic properties. Nevertheless, in vivo studies on antihyperglycemic activity of red yeast rice and rice bran have not  been reported. This current work aimed to evaluate glucose-lowering effects on alloxan-induced rats (Rattus norvegicus) strain Sprague Dawley treated with water extract of angkak, rice bran and combination of both extracts. The research was first carried out by extraction of RYR and RB, acclimatization of rats induced by alloxan, then separating them into 9 groups. The results showed that the lowest decrease in blood glucose, i.e. 61,43%, was attributed to group 5, namely rats administered with water extract of angkak (100 mg/Kg bw), being much higher compared to group 3 (rats treated with glibenclamide) reaching up to 40,88%. In terms of group 5, the level of blood glucose exhibited a gradual attenuation, from 256,67±174,77 mg/dL on day-3 to 99,00±9,64 mg/dL on day-12, which is a normal blood glucose concentration. It is noteworthy that administration of rice bran extract and combined treatment showed no significant effects on reduction of blood glucose.Keywords:  alloxan, blood glucose, red yeast rice, rice bran, antihyperglycemic

Abstract

This study aimed to isolate the endophytic fungi of soursop leaves that have the potential as anti-yeast. Soursop leaves were collected from 3 locations in West Java, Indonesia. The secondary metabolites produced by the endophytic fungi were extracted with ethyl acetate and tested for anti-yeast activity against Saccharomyces cerevisiae and Candida tropicalis. Profile of extracts contents characterization was carried out by using thin-layer chromatography (TLC). Twelve monoculture soursop leaves endophytic fungi isolates from Sukabumi (3 isolates), Cianjur (3 isolates), and Garut (6 isolates). It showed different abilities to inhibit C. tropicalis and S. cereviseae. The best extract is isolat from Cianjur (Sir-C1) and Sukabumi (Sir-S1). The stain patterns seen between the nine endophytic fungi extracts were similar. This indicates that the compounds in them may also be the same. But the extracts have a different absorbance for anti-yeast activities. This indicates that the total metabolites of each isolate are different. TLC results showed that ethyl acetate extraction produced almost the same metabolite components from nine endophytic fungi extracts, and all extracts had an anti-yeast activity with various absorbance. There are differences in the ability of 12 soursop leaf endophytic fungi to inhibit yeast model C. tropicalis and S. cerevisiae. Some extracts have similar TLC profiles, but their anti-yeast activity is not similar

Abstract

Propolis merupakan salah satu bahan alam yang dimanfaatkan oleh masyarakat, antara lain sebagai antibakteri, hepatoprotektor, anestesi lokal, antikanker dan antioksidan. Penelitian ini bertujuan menentukan tingkat toksisitas subkronis ekstrak etanol 70% propolis (EEP) pada tikus Sprague Dawley terhadap glukosa, kolesterol dan trigliserida darah selama 28 hari. Ekstrak propolis diberikan sebanyak 1 ml secara oral pada tikus jantan dan betina sebanyak 60 ekor dan 10 ekor sebagai kontrol. Ekstrak diberikan dalam dosis 0,3 mg/kg bobot badan (BB), 2,1 mg/kg BB, 14,7 mg/kg BB, 102,9 mg/kg BB, dan 720,3 mg/kg BB sebanyak 5 kali dalam seminggu selama 28 hari. Konsentrasi glukosa darah tikus jantan dan betina pada hari ke-28 adalah 79,85 – 97,49 mg/dL dan 61,88 – 90,26 mg/dL. Konsentrasi kolesterol darah tikus jantan dan betina pada hari ke-28 adalah 100,42 – 116,25 mg/dL dan 118,63 – 132,78 mg/dL. Konsentrasi trigliserida darah tikus jantan dan betina pada hari ke-28 adalah 128,64 – 142,37 mg/dL dan 110,95 – 128,58 mg/dL. Pemberian ekstrak etanol 70% propolis sampai dosis 720,3 mg/kg bb selama 28 hari tidak mengakibatkan abnormalitas terhadap nilai glukosa, kolesterol dan trigliserida darah tikus Sprague Dawley.  Semua parameter pada hari ke-28 tidak menunjukkan perbedaan yang signifikan secara statistika dibandingkan dengan kelompok normal.  Ekstrak etanol 70% propolis pada penelitian ini tidak menimbulkan pengaruh toksik pada konsentrasi glukosa, kholesterol dan trigliserida selama pengujian subkronis 28 hari.  Uji ANOVA pada hari ke-28 tidak menunjukkan perbedaan nyata dibandingkan dengan kelompok normal.

Abstract

Banana (Musaceaea) is one of the highest production fruits in Indonesia, which amounted to 7,162,685 tons in 2017. Banana peel waste produced can reach 40% of the total production of fresh bananas. Kepok banana peel contains a lot of nutrients and pectin. Pectin levels in banana peel ranged from 1.92% to 3.25% of dry weight. This study aims to characterize pectin from kepok banana peel (Musa balbisiana) extracted using nitric acid, citric acid, and HCl and screening potential of pectin bioactivity based on LC50 values. The highest pectin yield was obtained from extraction using pH 4 citrate buffer which was 3.68 ± 1.23%. In general, the characteristics of the best pectin from kepok banana peel are pectin extracted with HNO3 pH 4 with methoxyl degree 3.74 ± 0.34%, galacturonate content 87.64 ± 8.36%, and esterification degree 24.24 ± 0.098%. Based on the LC50 value the pectin extraction results can be classified into two, namely commercial pectin and pectin HCl pH 1.5 which is low bioactivity (LC50 > 100 ppm), and pH 4 HNO3 pectin and pH 4 citrate buffer pectin which has high bioactivity (LC50 ≤ 30 ppm).
