Abstract

Bacterial infection and antibiotic resistance are popular issues nowadays. Several pre[1]vious reports performed antibacterial screening activities involving natural herbs and synthetic drugs. Alanine racemase and transglycosylase are essential proteins for peptidoglycan membrane synthesis in bacteria and an alternative target for antibacterial performance. This study identified six flavonoid compounds in Syzygium myrtifolium to perform the antibacterial activity. In silico study was conducted for modelling flavonoids – protein complexes. Five flavonoids from S. myrtifolium were taken out of he canonical smiles from the PubChem database and modelled three-dimensional structure using ChemDraw and molView. Targeted protein, alanine racemase and transglycosylase were downloaded from Protein Data Bank with ID 4WR3 and 1SLY. Ligands and proteins were interacted by Molegro virtual Docker 5.0 and visualized by Discovery studio version 21.1.1. Five flavonoids showed inhibition with alanine racemase and transglycosylase in the same active sites of control and sodium benzo[1]ate. According to the binding energy, calopiptin performed the lowest binding energy value in alanine racemase complexes, while 2-Propanone, 1,3-bis(5-nitro-2-furanyl) showed the lowest value of four other flavonoids at transglycosylase complexes. The type of interactions were electrostatic, hydrogen bonds, hydrophobic interactions and unfavorable ones. Low binding energy and varied interaction types indicated tight of ligand-protein interaction. In summary, five flavonoids inhibited alanine racemase and transglycosylase, and the peptidoglycane membrane synthesis in bacteria might be inferred.

Abstract

Synthesis and characterization of cellulose acetate and nanocellulose acetate have been carried out. Cellulose was isolated from sengon wood powder waste using 6% NaOH solution and bleached with NaOCl. The cellulose was hydrolyzed with 60% formic acid to obtain nanocellulose. Cellulose and nanocellulose were then modified with acetic anhydride. The synthesized products were characterized by FTIR to identify organic functional groups, powder XRD to measure crystallinity, and SEM to observe surface morphology. Solubility tests in several types of solvents were also performed. The degree of substitution (DS) of cellulose acetate and nanocellulose acetate was carried out based on Indonesia’s SNI 0444:2009 method. The DS calculation show that the cellulose acetate and nanocellulose have DS of 2.9 and 2.1, respectively.

Abstract

This research presents the degradable bioplastics developed from pinewood nanocellulose as a filler in PVA matrices. The steps involve the isolation and characterization of cellulose and nanocellulose. Meanwhile, the manufacturing of degradable bioplastic involves the combination of PVA, nanocellulose, and with or without orange peel extract. The effect of bioplastics without the addition of citric acid and orange peel extract is also reported as a comparison. It is found that orange peel extract improves the tensile strength (1708.54 kPa), elastic modulus (42.71 kPa), elongation (40%), and degradability (78.44% in 2 weeks) compared to bioplastic without the orange peel extract. These results indicate that orange peel extract acts as a reinforcing agent in PVA-nanocellulose bioplastic.

Abstract

Staphylococcus aureus causes many severe diseases, such as meningitis, lung infection, and hematogenous osteomyelitis. The prolonged use of antibiotic drugs leads to antimicrobial resistance (AMR), decreasing their effectiveness against S. aureus. Therefore, novel drugs isolated from natural products have been the focus of many researchers to solve that problem. The essential oils isolated from Vitex trifolia leaf have many pharmacological activities, including antibacterial and antifungal activities. In this research, essential oil was isolated by distillation, followed by GC-MS analysis. The antibacterial activity of the essential oil was conducted using a disk diffusion test (Kirby-Bauer) and by molecular docking study. The essential oil yield from distillation was 0.13%, containing major compounds such as sabinene, eucalyptol, terpinen-4-ol, α-terpineol, limonene, and caryophyllene. The essential oil gave intermediate inhibition against S. aureus with a 6.91 ± 0.03 mm inhibition zone in 20 mg/mL of the sample. Using molecular docking study, caryophyllene and α-terpineol were the key compounds for inhibiting the active site of tyrosyl-tRNA synthetase, sortase A, and dihydrofolate reductase that are responsible for the S. aureus growth and development.

Abstract

Hydrodeoxygenation (HDO) is applied in fuel processing technology to convert bio-oils to green diesel with metal-based catalysts. The major challenges to this process are feedstock, catalyst preparation, and the production of oxygen-free diesel fuel. In this study, we aimed to synthesize Ni catalysts supported on silica-zirconia and alumina-zirconia binary oxides and evaluated their catalytic activity for waste cooking oil (WCO) hydrodeoxygenation to green diesel. Ni/Al2O3-ZrO2 and Ni/SiO2-ZrO2 were synthesized by wet-impregnation and hydrodeoxygenation of WCO was done using a modified batch reactor. The catalysts were characterized using X-ray diffraction (XRD), X-ray fluorescence (XRF), and scanning electron microscopy - energy dispersive X-ray spectroscopy (SEM-EDS), and N2 isotherm adsorption-desorption analysis. Gas chromatography - mass spectrometry (GC-MS) analysis showed the formation of hydrocarbon framework n-C15 generated from the use of Ni/Al2O3-ZrO2 with the selectivity of 68.97% after a 2 h reaction. Prolonged reaction into 4 h, decreased the selectivity to 58.69%. Ni/SiO2-ZrO2 catalyst at 2 h showed selectivity of 55.39% to n-C15. Conversely, it was observed that the reaction for 4 h increased selectivity to 65.13%. Overall, Ni/Al2O3-ZrO2 and Ni/SiO2-ZrO2 catalysts produced oxygen-free green diesel range (n-C14-C18) enriched with n-C15 hydrocarbon. Reaction time influenced the selectivity to n-C15 hydrocarbon. Both catalysts showed promising hydrodeoxygenation activity via the hydrodecarboxylation pathway. Copyright © 2021 by Authors, Published by BCREC Group.
