Abstract

Abstract

Dates are one of the plants that grow in the Middle East, one of them is the Golden Valley variety. Dates are usually consumed directly and can also be processed into date juice drinks. Dates refined process would produce waste in the form of date seeds are discarded and not processed into other products. This study used lipid-free Golden valley date seed extract to determine the secondary metabolite profile by using LC-MS/MS method, determining phenolic content using the Folin-Ciocalteau method, determining total flavonoids using the aluminium chloride method, determining antioxidant activity by DPPH method, and determination of antibacterial activity by the disc diffusion method. The results showed that the compounds identified from the lipid-free date seed extract were catechin-(4α→8)-catechin, bis((1S,2R,5S)-5-isopropyl-2,3-dimethylcyclo-hexyl)-2,6-dimethylpyridine-3,5-dicarboxylate, 3,5,6-trihydroxy-4',7-dimethoxyflavone, ethyl-2-amino-3-oxobutanoate, and bis(2-isopropyl-5-methylcyclohexyl)-2,6-dimethyl-1,4-dihydro-3,5-pyridine dicarboxylate. Lipid-free date seed extract contains total phenolic and flavonoid totals of 4364.704 mgGAE/100 grams of dried extract and 17200 mgQE/100 grams of dried extract. Its antioxidant activity is very strong in counteracting DPPH free radicals indicated by IC50 value of 10.1951 mg/L. Antibacterial activity shows strong inhibition of Staphylococcus aureus bacteria compared to inhibition of Escherichia coli bacteria that are categorized as weak.

Abstract

Scurrula ferruginea (Jack) Danser is a folk medicine to treat several diseases. S. ferruginea is the mistletoe that lives by deriving nutrients from the host. Hence, the host has a significant effect on the biological activities and bioactive components of S. ferruginea. In this study, the leaves of S. ferruginea were macerated by methanol solvent to extract the chemical components and fractionated by hexane and ethyl acetate solvent, respectively, to separate organic compounds. The biological activity of crude extracts as the antioxidant was investigated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. The result showed that the highest antioxidant activity was obtained from the ethyl acetate fraction with the IC50 of 10.88 ± 0.39 µg/mL. Furthermore, the trituration method used several solvents to isolate secondary metabolites from the initial crude methanol extract. This purification process produced a flavonoid compound of quercitrin. The antioxidant activity of quercitrin was also determined by the DPPH method. The result showed that this compound has stronger activity than the crude extracts with the IC50 of 5.95 ± 0.11 µg/mL. This study demonstrated that cacao mistletoe of S. ferruginea may have the potential activity to treat the diseases caused by the presence of free radicals.

Abstract

The Limau Sundai plant (Citrus Nobilis Lour) is widely known in West Sumatra. The fruit of this plant is commonly consumed as a cooking spice. This antioxidant activity test research aims to find out the potential of Lime Sundai peel as a source of antioxidant compounds. The peel of Limau Sundai fruit is ensnared with methanol solvent and concentrated using a rotary evaporator to obtain a coarse extract. The crude extract is then partitioned successively in n-hexane and ethyl acetate so that n-hexane, ethyl acetate, and methanol extracts are obtained. Sundai lime peel has good antioxidant activity against DPPH, which is shown in the ethyl fraction of acetate, methanol, and liquid methanol. The antioxidant activity of the three fractions is smaller than vitamin C and more significant than the hexane fraction. In the ethyl acetate, methanol and methanol/aqueous fractions with concentrations of 1076, 1822, and 1372 ppm in a row can reduce 50% of DPPH radical activity.

Abstract

This study aims to isolate secondary metabolites from Syzygium aqueum leaves and assess their suitability for targeting Epidermal Growth Factor Receptor (EGFR) proteins in lung cancer treatment. The methods utilized include maceration, fractionation, chromatography, and molecular docking. The isolated compounds were characterized using UV-VIS spectroscopy, FTIR, and NMR techniques. The compounds were 2',4'-dihydroxy-6'-methoxy-3',5'-dimethylchalcone (DMC) and lupeol. Molecular docking simulations were performed between DMC compounds and EGFR proteins (PDB ID: 1M17, 5D41, 5UG8, 5UG9, and 6LUD). Mutations in the EGFR kinase domain, particularly T790M, L858R, and V948R, confer resistance to first-generation inhibitors, including osimertinib. According to the in-silico docking results, DMC shows promise as an alternative inhibitor candidate. It can maintain interactions with the key residue Lys745 and provides stabilization through Asp855 ansamod Glu762. In contrast, osimertinib relies more on hydrophobic interactions and loses direct contact with Lys745, contributing to decreased affinity. This suggests that DMC may effectively overcome resistance associated with mutations, especially T790M, while maintaining crucial interactions within the ATP binding pocket. Therefore, DMC can be developed into a new non-covalent scaffold for treating lung cancer with EGFR mutations.

Abstract

Diabetes mellitus (DM) is a chronic metabolic disorder with a rapidly increasing global prevalence. The purpose of this study was to determine the potential of volatile compounds from microalgae Parachlorella kessleri AUP5 as potential α-amylase inhibitors through in silico approach. Analysis of the similarity of the drug was carried out with the Swiss ADME on the basis of Lipinski rule of five. Molecular screening was performed using Molecular Operating Environment (MOE). The volatile compounds with the highest inhibitor potential toward α-amylase is 9,12-octadecadienoic acid with a docking score of −6.215 kcal/mol. The compound interacted with key amino acid residues such as Ala198, His201, Ile235 and Asp195 at the enzyme binding site. The results indicated that 9,12-octadecadienoic acid from Parachlorella kessleri AUP5 has the greatest potential as a potential α-amylase inhibitors. However, more research is required to validate the results of these computational predictions.
