Abstract

ABSTRAKPenelitian ini bertujuan mendeskripsikan ciri-morfologi dan mengklarifikasi status taksonomi berdasarkan ciri tersebut pada semut-semut yang ditemukan di kawasan Taman Nasional Wakatobi khususnya di pulau Hoga. Penelitian ini merupakan penelitian eksplorasi. Sampel semut diperoleh dari lokasi penelitian dengan menggunakan perangkap tanam (pitfall trap) dan beating. Perangkap tersebut ditempatkan sedemikian rupa di sekitar tegakan mangrove yang telah dibuat plot-plot pengamatan. Deskripsi morfologi dilakukan dengan mengamati bagian: 1) kepala; meliputi bagian-bagain mulut, antena dan asesoris yang ada padanya 2) mesosoma meliputi bentuk pelekatan dan asesoris yang ada, dan 3) gaster, meliputi bentuk, jumlah ruas dan bentuk pelekatannya. Ciri-ciri yang ditemukan selanjutnya dikonfirmasi dengan buku identifikasi semut. Hasil penelitian menunjukkan terdapat 5 spesies semut yaitu: Mirmoterus sp., Oecophilla smaragdina, Camponotus sp. (Sub family Formicinae) Tapinoma sp., Iridomyrmex sp. (Sub family Dolicodinae). Ke-5 spesies tersebut merupakan family Formicidae. Kata Kunci: semut , mangrove, pulau Hoga Taman Nasional Wakatobi.   ABSTRACT The aims of this research was to describe the characteristics of morphology and clarify the taxonomic status based on these characteristics on the ants found in the Wakatobi National Park, especially on the Hoga  island. This research is an exploratory research.The ant sample was obtained from the research location by using pitfall trap and beating.The trap is placed in such a way around the mangrove stand that have made observation plots. Morphological description is done by observing the sections: 1) head; covering the parts of the mouth, antennae and accessories 2) mesosoma includes existing attachments and accessories, and 3) gaster, covering the shape, the number of segments and the form of attachment. The findings are further confirmed by ant identification books. The results showed there are five species of ants ie: Mirmoterus sp., Oecophilla smaragdina, Camponotus sp. (Sub family Formicinae) Tapinoma sp., Iridomyrmex sp. (Sub family Dolicodinae). All five species are family formicidae. Keywords: ant, mangrove, Hoga island in Wakatobi National Park

Abstract

ABSTRACTThe research was exploration ofpartial COI gene of moth (Plusia chalcites).The aim of this research is to characterization of nucleotide sequence of these COI gene, that was exracted with Cetyl Trimetyl Ammonium Bromide (CTAB) method, and then amplified by Polymerase Chain Reaction (PCR) technique with spesifik primer. The result PCR amplified subsequent analysis of nitrogen base arrangement in these COI gene. The analysis data using the BioEdit and MEGA Version 5.0 software.The results showed that the  segment of COI gene in Plusia chalcites as long 710 bp (base pairs). From these, there are 503/657 nucleotides which are conserved, 154/657 nucleotides of varies, 124/657 parsimoni informative nucleotides and 30/657 singeleton nucleotides. The base N composition consist of 39.0% Timine, 14.0% Cytosin (C), 31.7%Adenine (A) and 15.4% Guanin.Key words: Plusia chalcites, COI Gene characteristics and composition. ABSTRAK Penelitian ini merupakan penelitian eksplorasi untuk mengkarakterisasi sekuen gen COI. Karakterisasi gen COI meliputi isolasi DNA menggunakan metode CTAB (Cetyl Trimetyl Ammonium Bromide), amplifikasi gen COI dengan teknik PCR (Polymerase Chain Reaction) dan squencing untuk analisis susunan basa nitrogen pada gen COI. Analisis data molekuler menggunakan software BioEdit dan MEGA Version 5.0.Hasil penelitian menunjukkan segmen gen COI Plusia chalcites  yang teramplifikasi berukuran 710 pb, dan terkarakterisasi sepanjang 657pb. Dari segmen tersebut terdapat 503/657 nukleotida yang bersifat conserved (kekal), 154/657 nukleotida yang bersifat variabel (bervariasi), 124/657 nukleotida parsimoni informative dan 30/657 nukleotida singleton. Komposisi basa nitrogen sekuen gen COI Plusia chalcites yaitu Timin (T) 39,0%, Cytosin (C) 14.0%, Adenin (A) 31,7% dan Guanin 15,4%.  .Kata kunci :Plusia chalcites, Karakteristik Gen COI

Abstract

D. ananassae kinship catched on Kendari mainland had been analized by crossing beetween D. Ananassae from Kendari, Pondidaha and Lasolo regions, experiment method conducted with 3 resiprocal each, Data conversioned in isolation indexes and tested with Duncan’s test. Isolation index of D.ananassae from Kendari region with F-count =4.10, greater than F-tab (3.11), average of isolation indexes was 0.13, that mean kinship of D.ananassae from Kendari mainland (Kendari, Pondidaha and Lasolo) were closed enough one another and assumed that they were same species.Keywords: D. ananassae, kinship, Kendari mainland                                                    AbstrakKekerabatan D. Ananassae yang ditangkap didaratan Kendari telah dianalisis dengan persilangan antara D. Ananassae yang ditangkap dari daerah Kendari, Pondidaha dan Lasolo. Dengan metode eksperimen yang terdiri dari 3 kali ulangan untuk setiap perlakuan, data yang diperoleh dikonversi menjadi data indeks isolasi dan diuji dengan analisis varians dan dilanjutkan dengan uji Duncan.  Hasil analisis menunjukkan bahwa indeks isolasi seksual D. Ananassae di daratan Kendari mempunyai F-hitung sebesar 4,10 dan lebih besar daripada F-tabel sebesar 3,11.  rata-rata indeks isolasi seksual yang didapatkan adalah sebesar 0,13, hal ini menunjukkan bahwa kekerabatan D. Ananassae di daratan Kendari cukup dekat dan diduga D. Ananassae yang ditangkap di Kendari, Pondidaha dan Lasolo berada pada level spesies yang sama.Key words: D. ananassae, perkerabatan, daratan Kendari

Abstract

The service activity aims to provide an understanding to the youth organization regarding the potential of mangrove ecosystems to be developed as Edu-Ecotourism in Tanjung Tiram Village, North Moramo District, South Konawe Regency, Southeast Sulawesi. The method of implementing this service was carried out education, practice, and mentoring. A questionnaire and data analysis were carried out descriptively to evaluate the increase in understanding of youth organization. The results of this community service activity showed that the understanding of youth organizations in Tanjung Tiram village increased regarding the conservation of the mangrove ecosystem and its potential to be developed as an edu-ecotourism village and in practice-assistance activities, 11 types of mangrove ecosystem constituents were identified in Tanjung Tiram Village. With a good understanding of the community and the high diversity of mangroves, Tanjung Tiram Village could be developed into a mangrove Edu-Ecotourism Village.

Characterization of type I L-asparaginase encoding gene from the thermohalophilic bacterium Bacillus subtilis CAT3.4 from Wawolesea Hot Spring, Southeast Sulawesi, Indonesia. Biodiversitas 24: 5167-5178. This study aims to determine the molecular characteristics of the gene encoding type 1 L-asparaginase from CAT3.4 thermohalophilic bacterial isolates from the hot springs of Wawolesea, North Konawe, Southeast Sulawesi. This enzyme can be used as a cancer therapy agent and prevents the formation of acrylamide in food products. It is a kind of exploratory research. The characterization was done by amplifying the ansA gene sequence encoding L-asparaginase from CAT3.4 isolate by polymerase chain reactions (PCR) technique using primers AsnBac1-F1 (5'-ACGCGATATTTCTTTTGGCCGG-3') and AsnBac1-R1 (5'-CAGTGAAGAGGTGCATGGTATG-3'). The amplified PCR product was used as a template for sequencing by the Sanger method. The amino acid coding regions (CDS) obtained were bioinformatically characterized using the NCBI website for BLASTn analysis, restriction site and hydrophobicity profiles were analyzed using the BioEdit program, phylogenetic tree was analyzed using the MEGA X program, and type 1 L-asparaginase amino acid sequence was analyzed using the Expasy translate program. The characterization results showed that the target gene has a high similarity to the ansA gene sequence of 20 Bacillus subtilis strains, i.e., 99-100%, is closely related to the ansA gene of Bacillus subtilis strain SRCM103629 and Bacillus subtilis strain GOT9, can be identified using restriction enzymes MluI and BstI to differentiate the species of organisms they produce, has CDS encoding 329 amino acids with a dominant composition of polar amino acids (54.1%) and has a hydrophobicity profile of amino acids dominated by hydrophilic regions. All these characteristics have confirmed that the characterized gene is the ansA gene encoding type 1 L-asparaginase from Bacillus subtilis. The finding of the ansA gene from Bacillus subtilis in a thermohalophilic region in Southeast Sulawesi, Indonesia is a novelty of the research.

Abstract

The chemical dependency of farmers in Konda Satu Village and the accumulation of fruit peel waste potentially trigger greenhouse gas emissions. This Community Service (PKM) program aims to transfer science and technology in processing fruit peel waste into eco-enzyme as an environmentally friendly liquid organic fertilizer (POC). The implementation method was based on a participatory, educational, and applicative approach through socialization, direct technical guidance demonstrations, and field mentoring. The results of the activity showed a significant increase in community capacity, marked by more active participation in practicing eco-enzymes. This program also successfully compiled a practical guidebook and produced ready-to-use liquid fertilizer to stimulate a closed-loop agricultural system. In conclusion, this PKM activity has tangibly improved farmers' technical skills in processing waste into organic inputs toward low-carbon agriculture.
