Abstract

ABSTRAKPenelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri Gram negatif pada ambing kambing peranakan etawa (PE). Sampel yang digunakan pada penelitian ini terdiri dari swab ambing dari 5 ekor kambing PE. Sampel dibiakkan pada media nutrient broth lalu diinkubasikan pada suhu 37°C selama 24 jam. Isolasi dan identifikasi dilakukan dengan metode Carter yang meliputi pengamatan morfologi koloni, pewarnaan Gram, penanaman pada media diferensial dan selektif serta uji biokimia. Hasil isolasi dan identifikasi pada 5 sampel swab ambing kambing PE didapatkan dua koloni bakteri Gram negatif yaitu Escherichia coli dan bakteri X yang belum teridentifikasi melalui uji-uji yang dilakukan. Berdasarkan hasil tersebut dapat disimpulkan bahwa bakteri Gram negatif pada ambing kambing PE adalah Escherichia coli dan bakteri X yang belum teridentifikasi.Kata kunci: kambing PE, ambing, bakteri Gram negatif. ABSTRACTThis study aimed to isolate and identify Gram-negative bacteria on udder of etawa crossbred (PE) goat. Swab samples were obtained from 5 PE goats udder. Swab samples were cultured in nutrient broth media and incubated at 37°C for 24 hours. Isolation and identification were performed by Carter method, followed by observation of colony morphology, Gram staining, cultured on differential and selective media, and biochemical tests were performed. The result showed that there were two colonies of Gram negative bacteria Escherichia coli and the other colony has not been identified yet with this tests. Therefore it can be concluded that the Gram-negative bacteria found on udder of PE goat were Escherichia coli and bacteria X that has not been identified by using this method.Keywords:etawa crossbred goat, udder, Gram-negative bacteria.

Abstract

Candida sp. are commonly found as normal flora in vagina and in certain conditions are opportunistic pathogen. This study aimed to isolation and identification Candida sp. in vagina of the long-tailed macaque (Macaca fascicularis). The sample used in the study was a vaginal swab from five long-tailed macaque (Macaca fascicularis) from Seulawah Forest Aceh Besar. Identification of Candida sp. observed macroscopically by looking at the differences in the color of the colonies grown on the CHROMagar-Candida media. The data obtained were analyzed descriptively. The results of the examination showed that in the vagina of the long-tailed macaque (Macaca fascicularis) there were six species of Candida sp. like C. albicans, C. krusei, C. tropicalis, C. glabrata, C. dubliniensis and C. parapsilosis. It can be concluded that there are several species of Candida sp. which was successfully isolated and identified in vagina of a long-tailed macaque (Macaca fascicularis).

Abstract

This research aim was to determine the level of bacterial contamination of Salmonella sp on quail egg. The samples used in this research was 24 shell of quail eggs obtained from eight stores in traditional markets in Ulee Kareung Banda Aceh. The shell was crushed with a mortar and then cultured on SCB media (selenite cysteine broth). Isolation and identification was conducted by using SSA media culture, Gram staining, and biochemical tests. The results of this research showed that the shell of quail eggs contained Salmonella sp. Therefore it can be concluded that the quail eggs sold in Ulee Kareung traditional market were contaminated by Salmonella sp.

Abstract

The aim of this study was to see the antimicrobial activities of oxytetracycline to the inhibition of Staphylococcus aureus biofilm of Aceh’s cattle preputium isolates. The parameters measured in this study were the percentage of antimicrobial and biofilm inhibitory power. The antimicrobial activities were tested on Nutrient Broth using U-bottom polystyrene microplate 96 wells. The Biofilm stained using 1% of crystalviolet. Then, Optical Density (OD) read at λ 595 nm. The results showed that the oxytetracycline of 10%, 20% and 30% (μg / μL) had the antimicrobial abilities of 28.60% ± 13.23%; 36.30% ± 5.37%; and 57.70% ± 2.23%. In addition, The biofilm inhibitory capacity of oxytetracycline 10%, 20% and 30% (μg / μL) were 2.20% ± 8.77%; 2.70% ± 4.17%; and 3.73% ± 14.14%. Therefore, it can be concluded that the oxytetracycline  had the ability to inhibit the biofilm of Staphylococcus aureus.
