Abstract

ABSTRAK            Penelitian bertujuan mengkaji pengaruh penggunaan Kiambang (Salvinia molesta) fermentasi dengan level yang berbeda dalam ransum terhadap performan produksi telur itik lokal. Penelitian menggunakan 80 ekor itik lokal (itik Pengging) umur 24 minggu dengan rerata bobot badan 1457 ± 124,8 g (CV = 8,56%). Rancangan percobaan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 5 perlakuan (0%, 15% tepung Kiambang tanpa fermentasi, 15% tepung Kiambang fermentasi, 17,5% tepung Kiambang fermentasi, 20% tepung Kiambang fermentasi) dan 4 ulangan dengan 4 ekor itik untuk setiap unit percobaan. Parameter yang diamati adalah konsumsi ransum, produksi telur, dan konversi ransum. Hasil penelitian menunjukkan penggunaan tepung Kiambang (Salvinia molesta) yang difermentasi dengan Aspergillus niger dalam ransum memberikan pengaruh tidak nyata (P>0,05) terhadap konsumsi ransum, produksi telur, dan konversi ransum. Simpulannya adalah Kiambang dapat digunakan dalam ransum sampai taraf 20%. Kata Kunci : itik lokal; kiambang;fermentasi;performan produksi telur ABSTRACT            This research aims to determine the effect of fermented Kiambang with different level sin diets on egg production performance of local duck. The object of this research is eighty 24 weeks local ducks with average body weight 1457±124.8g (CV=8.56%). The design of the experiment is Completely Randomized Design with 5 treatments (0%, 15% non-fermented Kiambang, 15% fermented Kiambang, 17.5% fermented Kiambang, 20% fermented Kiambang) and 4 replicates with 4 ducks for each experimental unit. Parameters measured are feed consumption, egg production, and feed conversion. The results shows that fermented Kiambang with Aspergillus niger in the diets does not influence (P>0.05) on feed consumption, egg production and feed conversion. The conclusion is Kiambang can be use in diets to the extent of 20%.Keyword :local duck;Salvinia molesta;fermentation; egg production performance

Abstract

Objective of this research was to determine the effectiveness of various culture mediums on thecleavage to morula in in vitro embryo culture of ovine. A total of 1.064 zygotes (fertilized oocytes) were grown in three kinds of different embryo culture medium, which were Tissue Culture Medium 199 (TCM-199) (T1), TCM-199 + 17â-estradiol (T2) and TCM-199 + co-culture of Fallopian Tube Epithelial Cell (FTEC) (T3). Data were analyzed using a completely randomized design with ANOVA analysis and HonestlySignificant Difference test. The variable observed were the percentage of embryo cleavage and morula. Results showed that T3 had the highest percentage (72.45%) of the cleavage occurred in zygotes cultured in the TCM-199 + FTEC co-culture medium, as well as embryo growth to the morula stage (75.75%). Incontrast, zygotes cultured in T1 and T2 had lower cleavage and lower morula development than T3, as much as 27.38%; 47.82% and 58.15%; 66.66%, respectively. In conclusion, TCM-199 + FTEC co-culture medium is the best medium for embryo culture.

Abstract

The purpose of this study was to examine performances and factors that influence the gestation length (GL) and litter size (LS) of New Zealand White Grade (NZW Grade) rabbits. Reproduction records of NZW Grade rabbits were taken from April 2020 to March 2021. Data analysis was carried out using the general linear model (GLM) procedure from Statistical Analysis System (SAS) University Edition V.6p.2. software. Farm showed a significant effect (P<0.001) on LS and significant season (P<0.05) on GL and LS. GL in the rainy season (31.56) was shorter than the GL in the summer (32.26), but the LS in the rainy season (7.06) was greater than the LS in the rainy season (6.37). Maintenance management should be improved to reduce the impact of the season on GL and LS NZW Grade rabbits.

Abstract

Objective: The aim of this study was to identified genetic diversity of muscovy duck in Central Java through blood protein polymorphisms by using electrophoresis method.Methods: Blood sample was collected from a total of 60 muscovy ducks from the districts of Demak, Magelang and Pekalongan, 20 samples each. The Electrophoresis Polyacrylamide Gel of Electrophoresis–Thin Layer Electrophoresis (PAGE-TLE) was used in this study. Parameters observed were protein of albumin (Alb), ceruloplasmin (Cp), transferrin (Tf) and amylase-I (Am-I). Gene frequency, heterozygosity (Ho) and genetic distance were analyzed by using DISPAN program.Results: The results showed that the Alb, Cp, Tf and Am-I of muscovy duck showing polymorphic characters. Gene frequency of AlbB (0.61) was higher compared to AlbA (0.39). Gene frequency of CpA (0.62) was higher than the CpB (0.38). Furthermore, gene frequency of TfA and TfB were 0.32 and 0.68, respectively as well as gene frequency of Am-IA (0.82) was higher than Am-IB (0.18). Muscovy duck population of Demak showed higher Ho value for Alb and Am-I, whereas Magelang showed higher value for Cp and Tf. Population of muscovy duck of Demak showed closer genetic distance to Pekalongan than Magelang.Conclusions: The protein blood locus of three population of muscovy duck in Central Jawa showed polymorphic characters.

Abstract

Simpo and Limpo cattle is a crossbred cattle of Indonesian local cattle and exotic cattle that popular among traditional farmers in Indonesia. The objective of this study was to identify genetic diversity of Simpo and Limpo cattle based on blood-protein polymorphism. A total of 70 blood samples were collected from unrelated 35 heads of Simpo cattle and 35 heads of Limpo cattle from Grobogan dan Kendal population. The blood plasm was used to identify the blood proteins. Blood protein analysis was performed using Sodium Dodecyl Sulfate-Polyacrilamide Gel Electrophoresis (SDS-PAGE). Observed bands were used to perform allele interpretation of five locus, namely Pre-albumin (Pe-Alb), Albumin (Alb), Post-albumin (Po-Alb), Ceruloplasmin (Cp) and Amylase-1 (Am-1). Results showed there were polymorphism in all of the five blood protein locus in both cattle; 4 of 5 and 2 of 5 of the blood-protein locus studied showed deviation from Hardy-Weimberg Equilibrium (HWE) for Simpo and Limpo cattle, respectively and the average heterozygosity value (H) was 0.478 and 0.362 for Simpo and Limpo cattle, respectively. In conclusion, the study indicated that population of Simpo and Limpo cattle has high genetic diversity. Keywords: allele frequency; genotype frequency; heterozygosity

Abstract

Molecular genetic markers are regularly implemented in rabbit management for better breeding by selecting for specific genotypes. This study aimed to examine the diversity of the melanocortin-4 receptor gene in rabbits in Central Java, Indonesia. A total of 35 bucks were used in this study, comprising 10 New Zealand White, 11 Hyla and 14 Hycole. The DNA was extracted by Gene JET Whole Blood Genomic DNA extraction kit. Single Nucleotide Polymorphism (SNP) was detected by Polymerase Chain Reaction (PCR) and DNA sequencing. Hot carcass weight, cold carcass weight, reference weight and cut point 1-5 weight were among the parameters that were measured. This study showed that Melanocortin 4 Receptor (MC4R) genes were polymorphic, containing missense mutations and one SNP at 519G→A SNP 519GA had two alleles, A and G, with three genotypes (AA, GG, and AG). The rabbit population did not achieve HardyWeinberg Equilibrium (HWE) and showed a low level of genetic diversity as indicated by the heterozygosity results. The result of this study concluded that the frecuency AG as a genotype was higher than either GG or AA and A had a higher allele frequency than G. We recommended improving the validity of the data across various rabbit populations

Abstract

This study aimed to evaluate the growth performance of Red-Combed Kedu (RCK), Black-Combed Kedu (BCK), and White Kedu (WK) chickens based on sex during the early growth phase (0 to 10 weeks of age). A total of 136 Kedu chickens were raised under uniform management conditions, consisting of 99 RCK (65 roosters and 34 hens), 29 BCK (5 roosters and 24 hens), and 8 WK (6 roosters and two hens). Body weight was recorded weekly and analysed using a two-way General Linear Model (GLM) in SAS to assess the effects of chicken variety, sex, and their interaction. The Shapiro–Wilk test confirmed that the data were normally distributed (p > 0.05). The results showed that sex had a significant effect on body weight from weeks 3 to 10 (p < 0.05), with roosters consistently exhibiting higher weights than hens. Significant differences among chicken varieties were observed only in the hen group at weeks 9 and 10, where BCK hens had higher body weights (p < 0.05) than WK hens, while RCK hens showed intermediate values. In contrast, no significant differences were found among rooster groups at any age. These findings suggest that genetic factors play an essential role in influencing growth performance in Kedu chickens, particularly in hens. This information provides a valuable foundation for breeding selection and the conservation of local chicken genetic resources.
