Abstract

The present research has been undertaken with the aim to develop a topical gel formulation of ibuprofen which would avoid side effects of ibuprofen in oral administration. In this study, ibuprofen was made in solid dispersion form with PEG 6000 polymer composition 1 : 1.5 by fusion method. Solid dispersions aims to improve the solubility of ibuprofen. Triethanolamine as an alkalizing agent was added with different concentration in each gel formula which are 1 %, 2 %, and 4 %. They were evaluated for physicochemical properties such as organoleptic, pH, viscosity and in vitro drug release. Based on this research, it can be concluded that the different concentration of triethanolamine doesn’t give the significant effect on viscosity, nonetheless there is a significant effect on pH and dissolution rate of ibuprofen. It is obtained that the highest dissolution rate is F3 with a flux as 216.93 μg/cm2.minutes. Keywords: gel, ibuprofen, solid dispersion, PEG 6000, triethanolamine

Abstract

Morus alba (mulberry) root contains a secondary metabolite of oxyresveratrol with its potential skin-brightening activity. The application of the green extraction method using Ultrasonic-Assisted Extraction with Natural Deep Eutectic Solution (NADES-UAE) was developed because of its advantages, namely it can increase extract yields, low toxicity profile, reduce extraction time, shorter extraction stages without evaporation, and can be applied directly in topical preparations for cosmetic products. To ensure the potential for skin-brightening activity, some activity tests on mulberry root extract are needed. The purpose of this study was to determine the in vitro skin-brightening activity of mulberry root extract using NADES-UAE method. The method used was an in vitro tyrosinase enzyme inhibition test using a microplate reader and an antioxidant activity test using the DPPH method. The IC50 value of the best mulberry root extract (glycerin: urea 1:3 for 15 minutes) showed tyrosinase inhibition activity was 178.43 μg/mL. While the IC50 value in the best antioxidant activity test was 1392.14 μg/mL. Tyrosinase inhibitory and antioxidant activities in three samples showed the best results having the highest oxyresveratrol content. NADES-UAE mulberry root extract has the potential as a cosmetic preparation for skin lightening and has antioxidant effects for skin health.

Abstract

AbstractBiocellulose (BC), a natural polymer from bacteria fermentation, is one of the new delivery systems in cosmetic applications with its advantages such as biodegradability, compatibility, and high skin permeability. Due to their antioxidant activity, Mulberry (Morus alba) roots containing oxyresveratrol have the potential as an anti-aging agent. This research aims to know the formulation of BC sheet mask loaded Morus alba essence extracted with NADES-UAE method, physical evaluation, and antioxidant activity. Morus alba root extract was formulated into three formulas of essence preparation with different concentrations of gel base CMC Na (0.5; 1; 1.5%) and evaluated based on their physical properties. BC sheet mask produced from Acetobacter xylinum was formulated with different concentrations of sucrose, acetic acid glacial, and ammonium sulfate. The best BC formula was loaded with essence and physically evaluated. Antioxidant activity was tested in vitro using the DPPH method. The results showed that BC formulated as a facial sheet mask had good physical properties: thickness of 0.51-1.48 mm, water hold capacity of 64.77%, pH of 7.19, and swelling capacity of 168%. Morus alba essence formula of F2 provides the best physical evaluation: viscosity 800 mPas, pH 6.45, spreadability 7.4 cm, and good homogeneity. The essence also showed no physical changes on stability testing. Antioxidant activity testing of Morus alba roots extract and essence formulation shows the inhibition of free radicals. BC-loaded Morus alba roots essence extracted NADES method can be applied as an antioxidant agent for promising cosmetics in the future. Key words: antioxidant, biocellulose, Morus alba, NADES-UAE, sheet mask Formulasi dan Evaluasi Fisik Sheet Mask Bioselulosa Mengandung Essence Mulberry (Morus alba) sebagai Kosmetika Antioksidan AbstrakBioselulosa adalah polimer alam yang didapatkan dari fermentasi bakteri merupakan teknologi penghantaran kosmetik baru untuk sheet mask. Sheet Mask bioselulosa lebih ramah lingkungan, kompatibel dan memiliki permeabilitas yang baik pada kulit. Akar Morus alba telah diteliti mengandung senyawa yang memiliki aktivitas antioksidan dan menjadi kandidat bahan aktif alami dalam suatu sediaan kosmetik. Penelitian ini bertujuan untuk mengetahui formulasi dan evaluasi fisik sheet mask bioselulosa yang mengandung essence ekstrak Morus alba, serta aktivitas antioksidannya. Serbuk akar Morus alba yang diekstraksi dengan metode NADES-UAE diformulasikan menjadi tiga formula sediaan essence dengan konsentrasi basis gel CMC Na yang berbeda (0,5; 1; 1,5%). Sheet mask bioselulosa yang dihasilkan dari Acetobacter xylinum diformulasikan dengan konsentrasi sukrosa, asam asetat glasial, dan amonium sulfat yang berbeda dan dilakukan evaluasi fisik. Aktivitas antioksidan sediaan diuji secara in vitro menggunakan metode DPPH. Hasil penelitian menunjukkan bahwa sheet mask bioselulosa yang diformulasikan mempunyai sifat fisik yang baik: ketebalan 0,51-1,48 mm, kapasitas menahan air 64,77%, pH 7,19, dan kapasitas pengembangan 168%. Formula essence ekstrak akar Morus alba F2 memberikan hasil evaluasi fisik terbaik: viskositas 800 mPas, pH 6,45, daya sebar 7,4 cm, dan homogenitas yang baik. Sediaan essence juga tidak menunjukkan perubahan fisik pada pengujian stabilitas. Uji aktivitas antioksidan sediaan menunjukkan adanya penghambatan terhadap radikal bebas. Sheet mask bioselulosa mengandung essence ekstrak akar Morus alba yang diekstraksi dengan metode NADES dapat diterapkan sebagai agen antioksidan untuk kosmetik yang menjanjikan di masa depan. Kata kunci: antioksidan, bioselulosa, Morus alba, NADES-UAE, sheet mask

Abstract

Antioxidants are essential for protecting skin cells from free radicals, causing oxidative damage, and supporting human skin’s health. Natural antioxidants are abundant in plants, particularly in Robusta green coffee beans (Coffea canephora), which are rich in chlorogenic acid (CA), a key contributor to antioxidant properties. Efficient extraction methods are necessary to obtain these bioactives effectively. Advanced extraction technology, such as Ultrasound-Assisted Extraction (UAE) and Microwave-Assisted Extraction (MAE), enhances mass transfer and reduces processing time compared to conventional methods. This study quantified chlorogenic acid using Thin Layer Chromatography-Densitometry (TLC-densitometry) and evaluated the antioxidant activity using the DPPH method of NADES (betaine–triethylene glycol) liquid extracts from Robusta coffee green beans extracted via UAE and MAE. Chlorogenic acid concentration of MAE (3.64 mg CA/g extract ± 0.06) was higher than that of UAE (2.69 mg CA/g extract ± 0.04). The antioxidant activity (IC₅₀) values were 3266.66 μg/mL ± 67.97 (UAE) and 2598.05 μg/mL ± 29.42 (MAE), indicating higher efficiency for MAE. When expressed as chlorogenic acid equivalents in NADES extracts, IC₅₀ values were 9.04 μg/mL ± 0.13 (UAE) and 9.44 μg/mL ± 0.07 (MAE). NADES-MAE could be a promising method to acquire raw materials with high antioxidant activity, especially for cosmetic formulation.

Abstract

Pondok pesantren (ponpes) merupakan salah satu kekuatan dalam mengembangkan sumber daya manusia. Ponpes tidak hanya membekali santri dengan pengetahuan agama namun juga perlu memberdayakan santri dalam mengasah skill sehingga kelak ketika lulus dari ponpes bisa mandiri secara ekonomi. Salah satu skill penting di dunia modern saat ini adalah skill kewirausahaan untuk menciptakan peluang usaha dan meningkatkan ekonomi dalam masyarakat. Ponpes Putri Ibnu Katsir Jember adalah salah satu ponpes yang sangat mendukung hal tersebut, namun karena keterbatasan sumber daya dana dan manusia, kegiatan peningkatan skill santri dalam berwirausaha belum dapat dilakukan. Oleh karena itu dilakukan pengabdian masyarakat berupa pelatihan pembuatan produk sabun padat dengan memanfaatkan limbah kopi yang tidak terpakai, serta implementasi digital marketing dalam mempromosikan produk yang sudah dibuat. Metode yang digunakan meliputi penyuluhan interaktif, praktik pembuatan produk, dan evaluasi berupa pretest serta posttest. Hasil kegiatan menunjukkan peningkatan pemahaman santri dalam teori dan teknik pembuatan sabun padat, serta pengetahuan terhadap digital marketing. Hal ini sesuai dengan hasil rata-rata nilai pre-test peserta di awal kegiatan yaitu 60,4, lalu pada nilai post-test meningkat menjadi 87,2. Berdasar hasil tersebut, dapat disimpulkan bahwa kegiatan ini efektif dalam meningkatkan pemahaman dan skill kewirausahaan para santri.

Abstract

Coffee contains bioactive compounds such as chlorogenic acid and caffeine. Chlorogenic acid is recognized as a natural antioxidant, while caffeine is widely used to reduce fatigue and drowsiness. This study aims to simultaneously determine the content of chlorogenic acid and caffeine in robusta coffee leaf powder (Coffea canephora) using Thin Layer Chromatography (TLC) densitometry. The study identified an optimal eluent composition of ethyl acetate:formic acid:distilled water (18:1:1), with maximum wavelengths of 330 nm for chlorogenic acid and 274 nm for caffeine. High linearity was demonstrated with correlation coefficients of 0.993 for chlorogenic acid and 0.994 for caffeine. Sensitivity was validated with LOD values of 100.664 ng for chlorogenic acid and 297.232 ng for caffeine, and LOQ values of 301.994 ng for chlorogenic acid and 891.697 ng for caffeine. Selectivity was confirmed as the method effectively separated chlorogenic acid, caffeine, and other compounds with a resolution (Rs) ≥ 1.5, while specificity was evidenced through purity and identity values exceeding 0.99. Precision was proven through repeatability (%RSD for chlorogenic acid: 2.94%; caffeine: 2.27%) and intermediate precision (%RSD for chlorogenic acid: 2.26%; caffeine: 2.18%). Accuracy was achieved with mean recovery rates of 102.22% for chlorogenic acid and 99.73% for caffeine. The determination of chlorogenic acid and caffeine concentrations in robusta coffee leaf powder revealed variations between samples: PTPN XII Renteng yielded 2.278% ± 0.209% for chlorogenic acid and 1.171% ± 0.821% for caffeine, while Puslitkoka Indonesia samples showed 1.238% ± 0.206% for chlorogenic acid and 0.917% ± 0.823% for caffeine. Based on validation parameter assessments, the method is deemed valid as it meets all acceptance criteria for each parameter. This research provides an efficient and accurate analytical approach for simultaneous determination of bioactive compounds in robusta coffee leaves.
