Abstract

The  prognosis  of  breast  cancer  patients  is  closely  associated  with  the  response  of tumor  cells  to  chemotherapy  agent.  Doxorubicin  is  one  of  the  primary  chemotherapeutic agents  used  for  the  treatment  of  breast  cancer.  Resistance  to  chemotherapy  is  believed  to cause  treatment  failure  in  cancer  patients.  Furthermore,  long  time  exposure  to chemotherapeutic  agent  induces  cancer  cells  resistance.  MCF-7  sensitive  cells  used  as chemoresistance  model  have  overexpression  P-gp  (P-glycoprotein).  Chemoresistance  was established by treating MCF-7 cells with 0.5 µg/ml doxorubicin-contained medium for a week. 50% inhibiting concentration (IC50) doxorubicin on MCF-7 cells/DOX were determined using MTT assay. Western blot assay and immunocytochemistry assay was performed to determine the expression of P-gp. Morphological of MCF-7 cell/DOX was changing to become larger and have  lamellapodia.  IC50  value  of  doxorubicin  was  700  nM  on  MCF-7/DOX  and  400  nM  on sensitive MCF-7 cells. The MCF-7/DOX sensitivity to doxorubicin was decreased, shown by 1.5  fold  higher  IC50  of  doxorubicin  on  MCF-7/DOX  compared  to  MCF-7  sensitive  cells. Treatment doxorubicin to sensitive MCF-7 cells leads to the increasing P-gp expression. The P-gp  level  expression  has  strong  correlation  with  the  low  sensitivity  of  MCF-7/DOX  to doxorubicin.Key words: doxorubicin, resistance cells, sensitive MCF-7 cell

Abstract

Leunca (Solanum nigrum L.)ethanolic extractshowedcytotoxic activity on several cancer cell lines (HepG2, HT-29) and showed anti-proliferative activityon MCF-7 cells. Its application as a combinationagent in chemotherapy will increase  the effectivity and reduce  the toxicity of chemotherapy. We predict that application of combinatorial chemotherapy in cancer treatment will  be  more  effective  and  less  toxic  compared  to  single  treatment.  Our  research  aims  to investigate  the  cytotoxic  activitiy  of  leunca  herbs  ethanolic  extract  alone  and  in  combination with  doxorubicin  on  HeLa  cell  line.  MTT  assay  was  conducted  to  measure  the  growth inhibitory  effect  of  leunca  herbs  ethanolic  extract  and  combinatorial  treatments.  Leunca  herb ethanolic extract (5, 50, 250 μg/ml) increased the cytotoxic effect of  doxorubicin compared to doxorubicin alone. The strongest cytotoxic activity resulted from the combination of 250 μg/ml leunca  herbs  ethanolic  extract  and  250  nM  doxorubicin.  Based  on  our  results,  leunca  herbs ethanolic extract is a potential chemopreventive agent, while its molecular mechanism needs to be explored.Keyword : Leunca herbs ethanolic extract, doxorubicin, HeLa, MTT assay

Abstract

Cervical cancer is a type of cancer possessing the 3rd highest incidence among women worldwide. Todays therapies against cancer are still ineffective, thus people are seeking for alternative method. Erythrina fusca Lour. has been used traditionally as anticancer. This experiment is conducted to study the ability of chloroform extract of E.fusca Lour. leaves and its isolates on HeLa cervical cancer cells. The cytotoxicity assay and doubling time assay were done using direct counting method, while the DNA staining assay was done using acrydine orange. The chloroform extract and isolate #30 showed cytotoxic activity with IC50 of 16 and 5 μg/ml, respectively, while isolate #2 and isolate #8 didnt. The effluent #30 could not affect HeLa cells growth at the given concentration, but it might promote DNA fragmentation indicating apoptosis induction. Abstract

Eksplorasi agen kemopreventif kanker masih terus dikembangkan, terutama yang berasal dari bahan alam. Hal ini didasari oleh keinginan untuk menekan angka kematian akibat kanker dan mengurangi efek samping yang ditimbulkan oleh terapi kanker yang saat ini digunakan. Penelitian ini ditujukan untuk mengetahui aktivitas sitotoksik daun beluntas (Pluchea indica Less.) terhadap sel kanker leher rahim yang merupakan kanker ginekologi peringkat pertama yang menyerang wanita. Aktivitas sitotoksik dilakukan terhadap ekstrak n-heksana, diklorometana, dan metanol daun beluntas terhadap sel HeLa dengan metode perhitungan langsung menggunakan bantuan perwarna trypan blue. Nilai IC50 dari ketiga ekstrak daun beluntas berturut turut adalah 18,06 μg/ml, 74,56 μg/ml, dan 31,21 μg/ml. Ketiganya memberikan perbedaan yang signifikan. Ketiga ekstrak yang diuji memiliki aktivitas sitotoksik dan dapat dikembangkan sebagai kandidat agen kemopreventif kanker. Namun masih perlu diteliti lebih lanjut bagaimana mekanisme aksi yang menjadi perantara aktivitas sitotoksik tersebut.

Abstract

One of the natural ingredients that to be used to inhibit bacterial growth is the Zingiber officinale. In Indonesia, there are 3 types of ginger which are distinguished by morphology, namely ?emprit ginger (Zingiber officinale var. Amarum), ? gajah? ginger (Zingiber officinale var. Officinarum), and red ginger (Zingiber officinale var. Rubrum). This research was conducted to determine the activity of extracts and fractions of three ginger varieties in inhibiting the growth of Staphylococcus aureus using the disc diffusion method. The test group consisted of ethanol extract, n-hexane fraction, and ethyl acetate fraction from 3 ginger varieties, concentrations of 5, 10, 20% w / v, gentamicin 10 ?g (positive control) and 10% DMSO (negative control). The test results showed that ethanol extracts and fractions from the rhizomes of three varieties of ginger have the ability to inhibit S. aureus at concentrations of 5, 10, and 20% w / v, increasing the concentration will increase the diameter of the inhibition zone. At a concentration of 20% of all samples, the one with the greatest resistance was the n-hexane fraction (diameter of inhibitory zone of emprit ginger 9.80 mm, elephant ginger 9.78 mm, and red ginger 9.90 mm). The difference in ginger variety only affects the antibacterial activity of the ethanol extract, but not the n-hexane and ethyl acetate fractions.

Abstract

Staphylococcus epidermidis has become one of the most important pathogenic bacteria that lead to nosocomial infection. This is caused by its ability to form a biofilm on the surface. Essential oils are one of the natural product that could be used as antiinfection agents. Bengle rhizome essential oil has been known to have activity against non-pathogenic bacteria S. epidermidis. This study was proposed to know the antibacterial and antibiofilm activity of bengle rhizome essential oil against clinical isolate of S. epidermidis at concentrations of 45, 60, 75, and 90%. Antibacterial activity was determined by the diffusion method with hole plate technique, while antibiofilm activity was determined by the microtiter assay method with crystal violet staining and quantified by measuring the optical density (OD) at λ 595 nm. This study showed that bengle rhizome essential oil has antibacterial and antibiofilm activity. The largest antibacterial and antibiofilm activity was shown from 90% concentration of bengle rhizome essential oil. Abstract

Malaria is an infectious disease caused by Plasmodium sp. One of the herbs that are empirically used as an antimalarial is Tithonia diversifolia. Preparation of dried leaf extract of T. diversifolia used oven drying and the addition of cab-o-sil as a dryer. This study aims to determine the antimalarial activity of extracts of dried leaves of T. diversifolia. Antimalarial activity assay method used was a modified peter test. The probit analysis of this study showed that ED50 value on mice given the dry extract of T. diversifolia leaves was 214 mg /kg body weight and categorized as good at classifying antiplasmodium in vivo.     Keywords: antimalarial activity, Tithonia diversifolia, dry extract, ED50

Abstract

Jati belanda leaves (Guazuma ulmifolia Lmk.) and Roselle petals (Hibiscus sabdariffa L.) have been known for their activity as antihyperlipidemia with different mechanisms. Combination of these plants was expected to has a synergy effect in reducing cholesterol. However, the toxicity of jati belanda leaves and roselle petals extract combination must be investigated for knowing its safety. Based on the acute toxicity test, the LD50 value of jati belanda leaves and roselle petals extract combination was >5,000 mg/kg bw. It was classified as category 5 or unclassified. This research aimed to study about its effect in cardiac by cardiac histopathology examinations of the rats. The result showed that there was no cardiac histopathology changes of the rats at doses 5,000 mg/kg bw. Keywords: jati belanda leaves, roselle petals, acute toxicity, cardiac histopathology

Abstract

This research was aimed to determine the effect of antifertility among the methanol extract of ripe and raw papaya seeds in decreasing the quality and quantity of spermatozoa. Animal test rats were divided into 3 groups: negative control (CMC-Na 1%) and the treatment groups given methanol extract of ripe and raw papaya seeds extract. The extracts were given orally for 20 days. On the 21st day, mice were dissected to obtain sperm from the cauda epididymis, then were observed for the quality (motility, viability, morphology) and the quantity of spermatozoa. One-Way ANOVA followed by Post Hoc Test LSD analysis showed that was better for its activity then that of the ripe papaya seed extract. Abstract

Jahe gajah (Zingiber officinale var. officinale) is one of export commodity plant in Indonesia. Part of jahe gajah often to be used is the rhizome, that content lots of essentials oil. One of the essential oil content is a phenolic compound. Phenolic compound is a natural product function as antioxidant reducing oxidative damage in the body that cause inflammation. The aim of this study was to determine the total phenolic content and antioxidant activity of jahe gajah oil. The total phenol content was determined using Folin-Ciocalteu method and the antioxidant activity assay was done using DPPH method. The total phenol content for jahe gajah oil was 0.626 ± 0.022 mg GAE/g . The IC50 of jahe gajah oil was 5,766 ± 0,087 µl/ml. Antioxidant activity from jahe gajah oil was suggested to be contributed by the phenolic compound and result from synergistic action of all constituents.
