Abstract

Chronic kidney disease (CKD) is a condition of kidney which cannot perform its function normally, and expected to increase every year. Paper based chip was used in this case made by immobilized picric acid for creatinine detection, tetrabromophenol blue for protein detection, and bromothymol blue and methyl red for pH detection, on “whatman” filter paper which had been patterned using rubber based ink. The chip was applied for urine of patient, that has analitycal characteristics included response time approximately for 4 hours, linierity of creatinin achieved in concentration range of 0.1 -100 µg/ml and protein of 1 – 200 µg/ml, LOD (limit of detection) and LOQ (limit of quantification) of creatinin 0.088 µg/ml and 0.293 µg/ml, and protein 0.462 µg/ml and 1.540 µg/ml respectively. Interference study showed that urea, salt and sugar, interferen around 0.271-1.966% for creatinin and protein of 0.429 – 1.786%, respectively. The chip could be used up to 3 weeks if it stored in a dry place on room temperature and can be used to analyze condition of CKD over health condition. Keyword: CKD creatinin, protein, pH, and sensor

Abstract

Zamzam water is called the holy water that is located in the heard city of Mecca. Zamzam is believed to have many benefits for health so widely consumed by Moslems throughout the world. In 2010, the Government of Saudi Arabia implementing rules ban export of zamzam water for commercial purposes, but zamzam water are sold freely in the world. In this research, the determination of the purity of Zamzam water was developed. The method used was NIR spectroscopy combined with multivariate statistical methods (chemometric). Zamzam water simulation were consist of pure Zamzam water, mineral water, and a mixture of Zamzam water and mineral water with a concentration range of 0% -100%. Chemometric model used are PLS, LDA, SIMCA, and SVM. LDA classification model is the best chemometric model with 100% recognition ability and prediction ability respectively. NIR spectroscopy and chemometric method have been applied succesfully to detect the purity of Zamzam water samples which marketed in Jember.   Keywords:zamzam water, purity, NIR, spectroscopy, chemometrics.

Abstract

Phenols are group which having one or more hydroxyl groups which attached in aromatic ring and are found in plants. The aim of this research was to study whether NIR and chemometric methods could be used to determine the total phenol content of leave extracts. These methods were compared to UV-Vis spectrophotometry. Phenol was extracted from plant leaves by ultrasonic and maceration. NIR spectral data of selected leave extracts were correlated with total phenol content using chemometric. In this study, the chemometric method that used for quantitative and qualitative analysis were Partial Least Square (PLS) and Linear Discriminant Analysis (LDA), respectively. The PLS R2 calibration was 0.9920369 and RMSEC was 2.0049472. In addition, the R2 of LOOCV and 2-Fold-Cross-Validation were 0.9939401 and 0.9834646, respectively. Furthermore, LDA gave accuracy of 100%. The significance of phenol total that have been measured by NIR and UV-Vis spectrophotometry was evaluated with paired samples T-test and gave no significant difference. Abstract

Burger is one of the favorite processed meat products. The adulteration of pork in halal-labeled beef burger becoming problem for the Muslim community. Therefore, it is necessary to develop some rapid and reliable methods for detection of pork in beef burger samples. NIR method combined with chemometrics were choosen because those methods are rapid, easy and reliable. The purpose of this study was to determine of pork adulteration in beef burger samples using NIR method combined with multivariate statistical methods such as Linear Discriminant Analysis (LDA), Soft Independent of Class Analogies (SIMCA), and Support Vector Machines (SVM). The NIR method and the best classification models of chemometric were applied in the beef burger samples. The result of NIR and Chemometrics is compared to the pork strip test. The results shows that the NIR-chemometrics method in good agreement with pork strip test toward pork adulteration in the beef burger samples tested. Abstract

Yam bean is one of tubers group that has many benefits because of its chemical constituents named inulin. Yam bean are produced in several regions in Indonesia such as Tegal, Bogor, Prembun, and Madura. In this research, inulin determination in yam bean tuber from Gresik East Java as one of yam bean production center, performed to obtain the plant source in Indonesia which is potential to produce inulin. Inulin determination was conducted using TLC Densitometry method that has been validated. Based on a standard curve that meets the requirements of linearity (r=0,997; Vxo=3,847%; Xp=355,407ng), the results showed that levels of inulin in yam bean tuber derived from Gresik was 12,322%±1,733% and mean recovery was 100,08% ± 1,142%. Abstract

Inulin is a polysaccharide , which is composed of fructose units. Inulin is generally found in plants family Compositae, Amirilidaceae, and Poaceae. Benefits inulin is as bifidogenic body, stimulate the immune system , relieve constipation , reduce the risk of osteoporosis . Inulin can be consumed alone as a prebiotic supplement. The purpose of this study was to develop a method for determination of inulin TLC-Densitometry in dahlia tuber extract samples (Dahlia spp L.) grown in soil media and polybag . Determination of inulin as a method of TLC densitometry using the stationary phase TLC plates Silica Gel F254, mobile phase = acetic acid glacial p.a: methanol p.a: aquabides sterille (v/v/v/v) (0,5:7,5:2) , solvent = Aquabides sterille: Ethanol 96 % p.a (3:1) v/v, apparition stains is mixed aniline in acetone 1% v/v:diphenylamine in acetone 10% b/v:phosphoric acid (5:5:1), and the wavelength () 380 nm . Validation of the method showed that the research procedures performed has had good linearity with a coefficient correlation 0.996 and Vx0 value 4.850%. This method also gives good accuracy and precision that is the percent recovery of inulin at 99,96%±0,39%. and the value of repeatability = 0,993% < 2,7% and intermediate precision RSD = 0,554% < 2,7% with limits of detection (LOD) and limit of quantitation (LOQ), respectively, 71,03 ng/spot and 236.76 ng/spot. The results showed levels of inulin in the dahlia tuber extract soil planted mediated by 86.256%±0.669% (%b/b±RSD%), and dahlia tubers grown polybag mediated by 76.146%±1.657% (%b/b±RSD%).

Abstract

Meat is a high nutritional food that rich of protein, fat, and sugar. Smart label has been applied as a beef freshness detector. Two types of pH indicator have been used i.e bromocresol purple (BCP) and methyl red (MR) as dual indicator freshness. The objective of this research was to determine the beef freshness using smart label at room temperature. The color change of the smart label was examined by imageJ software to determine the freshness degree using the mean RGB value. The beef was examined every 2 hours for pH and total volatile base (TVB-N) analysis during the 24 hours storage at room temperature. The result showed that color indicator will change according to the beef freshness, bromocresol purple turned from yellow to purple (mean RGB 171.465 ± 1.122) and methyl red turned from red to yellow (mean RGB 162.082 ± 1.315). The beef freshness at room temperature decreased as the pH increase from 5.61 to 6.23 along with the color change of smart label. Furthermore, the color would change when 0.022 %N of TVB-N has been reached. Therefore, the beef freshness can be determined by using smart label based on dual indicator of bromocresol purple and methyl red in room temperature. Keywords: beef freshness, smart label, pH, TVB

The aim of this research was to study whether NIR and chemometric methods could be used to determine the flavonoid content. These methods were compared to UV-Vis spectrofotometry. Flavonoid was extracted from plant leaves by ultrasonic and maceration. NIR spectral data of selected leave extracts were correlated with flavonoid content using chemometric. In this study, the chemometric method that used for quantitative and qualitative analysis were Partial Least Square (PLS) and Linear Discriminant Analysis (LDA), respectively. The PLS R2 calibration was 0.9916499 and RMSEC was 2.1521897. In addition, the R2 of LOOCV and 2-Fold-Cross-Validation were 0.9986664 and 0.9823225, respectively. Furthermore, LDA gave accuracy of 100%. The significance of flavonoid content that have been measured by NIR and UV-Vis spectrofotometry was evaluated with paired samples t-test. Abstract

The content of polyphenolic compounds in mango and fragrant pandan leaves has been showed an antioxidant activity and commonly used as traditional treatment of some diseases. The aim of this research was to determine the activity of antioxidant of the gadung mango leaves and fragrant pandan leaves extract combination. The activity of antioxidant was investigated by DPPH method (IC50) with vitamin C as positive control. The results showed that the sample had lower antioxidant activities than vitamin C. The IC50 value of vitamin C was 2.613±0.021 μg/ml, followed by methanolic extract of gadung mango leaves, ratio 1:1 of gadung mango leaves extract and pandan leaves extract combination, and ethanolic extract of fragrant pandan leaves with IC50 value of 3.263±0.009; 13.392±0.157; 39.700±1.003 μg/ml, respectively. Methanolic extract of gadung mango leaves and ethanolic of fragrant pandan leaves combination didn't have higher antioxidant activities compared to the antioxidant activities of the singular methanolic extract of gadung mango leaves itself. Keywords: antioxidant activity, gadung mango, fragrant pandan

Abstract

 Kepundung (Baccaurea racemosa Muell.Arg.) merupakan salah satu tanaman berkhasiat di Indonesia yang mengandung flavonoid, polifenol, tanin, dan terpenoid yang dapat berfungsi sebagai antioksidan. Kandungan senyawa dengan aktivitas antioksidan dapat dikaitkan dengan potensi aktivitas antidiabetes dari tanaman ini. Penelitian dilakukan untuk mengetahui aktivitas antioksidan dan antidiabetes ekstrak metanol, etanol, dan etil asetat dari daun kepundung. Ekstraksi dilakukan dengan metode ultrasonik. Pengujian aktivitas antioksidan dilakukan secara kuantitatif dengan metode DPPH, sedangkan pengujian aktivitas antidiabetes menggunakan metode penghambatan enzim α-amilase. Nilai IC50 pada penentuan aktivitas antioksidan dan antidiabetes ditentukan dengan menggunakan spektrofotometer UV-Vis pada panjang gelombang maksimum. Hasil penentuan antioksidan menunjukkan bahwa nilai IC50 ekstrak metanol, etanol, dan etil asetat masing-masing adalah 9,38 ± 0,15; 10,55 ± 0,09; dan 946,70 ± 2,31 ppm. Ini menunjukkan ekstrak metanol dan etanol memberikan hasil terbaik dalam aktivitas antioksidan. Nilai IC50 aktivitas antidiabetes in vitro ekstrak metanol, etanol, dan etil asetat masing-masing adalah 67,63 ± 0,36; 67,46 ± 0,23; dan 841,04 ± 1,52 ppm. Hasil ini juga menunjukkan bahwa ekstrak metanol dan etanol daun kepundung memiliki aktivitas antidiabetes yang hampir sama
