Abstract

Mangrove sediment is a habitat for various bacteria, one of them is amylolytic bacteria which has the potential to produce amylase enzyme. Amylase enzyme has many benefits in industry, textiles and medical. The mangrove ecosystem area has special soil characteristics, which have the opportunity to have microorganism diversity, one of which is the mangrove ecosystem in the Wonorejo region, Surabaya. This study aims to obtain potential amylolytic bacteria from potential amylolytic isolates derived from mangrove sediment. Soil bacteria were isolated in Starch media for 2% agar and tested with Lugol’s Iodine reagents to measure their amylolytic index. Character isolates observed included colony morphology, Gram staining, and motility. Of the 27 isolates found there were 3 isolates with the highest index values in their activity, namely isolates A.7, A.27 and A.64. Characterization results showed that isolates A.7 and A.64 were Gram negative bacteria, and isolates A.27 were Gram positive bacteria. And the motility results for the three isolates were negative, with the results of a positive catalase test.

Abstract

Background: Chronic kidney disease and urinary tract disorders are prevalent health issues among the elderly. Limited public awareness regarding risk factors and the importance of early detection often results in new cases being identified only at advanced stages. Therefore, promotive and preventive efforts through health education and community-based screening are essential. Purpose: To increase public knowledge regarding the prevention of kidney and urinary tract disorders through early detection via urinalysis. Method: This community service activity was conducted in November 2025 with the Al Mahbub Study Group (Pandugo) in Penjaringan Sari Village, Rungkut District, Surabaya, involving 29 local residents as respondents. The activity comprised urinalysis testing, health education, and interactive discussions with a Clinical Pathology specialist. Urinalysis was performed using 10-parameter test strips covering pH, specific gravity, erythrocytes, leukocytes, bilirubin, urobilinogen, glucose, protein, nitrite, and ketones. Results: All 29 respondents, aged 37–74 years, completed the activity. Urinalysis screening data showed that the majority of respondents (55.2%) had a urine pH of 6. Most respondents tested negative for leukocytes (65.5%), erythrocytes (89.7%), protein (93.1%), glucose (96.6%), and urobilinogen (96.6%), while all respondents fell within the normal range for urine specific gravity. Additionally, all respondents tested negative for bilirubin, nitrite, and ketones. Conclusion: A simple urinalysis screening activity combined with health education can identify community members requiring further examination while raising awareness about the importance of early detection of kidney and urinary tract disorders. Urinalysis serves as an effective early detection tool for identifying at-risk individuals, providing comprehensive health insights to prevent comorbidities and facilitating referrals for further medical evaluation. Suggestion: Periodic screening and health support are needed for community members who show abnormal urinalysis results. Collaboration among educational institutions, healthcare professionals, and the community must be strengthened to support efforts in preventing kidney and urinary tract diseases within the community. Keywords: Early detection; Elderly; Health education; Health screening; Kidney disorders; Urinalysis Pendahuluan: Penyakit ginjal kronik dan gangguan saluran kemih merupakan masalah kesehatan yang banyak ditemukan pada kelompok usia lanjut.
