Abstract

Papaya seeds were known as medicinal plant which have anti-fertility potencies. So it could be used as a herbal contraceptive. The purpose of this study was to test the anti-fertility effect of Papaya seed water extract on mice (Mus musculus L.) fertility which proved by pregnancy rate, litters number and sex ratio of litters. This study was designed by Completely Randomized Design, using 15 female Swiss Webster mice that were divided into 5 treatment groups with triplicates. The K(-) group were treated by distilled water; K(+) group were treated by contraceptive pills; P1, P2, P3 groups that were treated by papaya seeds extract with 1.4, 3.5 and 7 mg/animal/day respectively. Each treatment was given orally with 0.5 ml in volume for 21 days. At the end of treatment, mice was mated. The observed parameters were pregnancies occurence, litters number and sex ratio of the litters. The litters number data were analyzed by Analysis of Variance. The results showed there was differences in pregnancy time and sex ratio between control and treatment groups, but there was not significantly different in litters number. It could be concluded that the fertility of mice was inihibited by water extract of papaya seeds.

Abstract

Diabetes mellitus is a metabolic disease that occurs due to impaired insulin secretion caused by progressive damage to beta cells. Pineapple skin vinegar contained acetic acid and antioxidants which have the potential to help repaired the structure of the nephron ren and other organs affected by diabetes. The purpose of this study was to examined the effectiveness of pineapple skin vinegar on improved the histological structure of diabetic rats ( Rattus norvegicus L.). This study based on changed in the structure of the nephron in samples of normal and alloxan-induced mice pre-treatmented and post-treatmented. Twenty-four rats were divided into 6 groups, named normal control, positive control (diabetes + 0.4 mL apple vinegar), negative control (diabetes + water), dose test groups 1, 2, and 3 (pineapple vinegar 0.2 mL; 0.4 mL; 0.8 mL). Statistical analysis test used ANOVA was followed by Duncan test. The conclusion of this research, the pineapple skin vinegar showed the ability to repair the histopathological structure of white rats damaged by diabetes. The optimum dose needed was 0.8 mL to improved the histological structure of the nephron, as indicated by the glomerular diameter and the distance of the Bowman's capsule space to near normal.

Abstract

Neem leaves is one part of the neem tree which can be used as a traditional medicinal herb. The purpose of this study was to examine the effect of neem leaf ethanol extract on hepatosomatic index (HSI) and hepatocyte diameter in female mice. The study used a Complete Randomized Design consist of 4 treatments and 5 replications including K (aquadest), P1, P2 and P3 (neem leaf ethanol extract with dose of 8.4,11.2 and 14 mg/ kg BW/day. The treatment was administered orally with a volume of 0.2 mL for 21 days. Body weight were measured every 7 days. The liver was isolated, weighed and histologically prepared with paraffin method, HE staining and 5 μm incision thickness. HSI value determined from body weight and liver weight. Hepatocyte diameter were measured on 7 hepatocytes per lobule. The data was analyzed using ANOVA at 95% confidence level. The results showed that neem leaf ethanol extract had no significant effect (p> 0,05) on body weight, liver weight, HSI value and hepatocyte diameter, but the liver microanatomy showed hepatocyte damage. It can be concluded that exposure of neem leaf ethanol extract for 21 days still safe to be used for reference of traditional medicine. Keywords: Neem leaf, HSI, hepatocyte diameter

Abstract

Neem leaves (Azadirachta indica A. Juss.) contain organic compounds such as alkaloids, steroids, flavonoids, triterpenoids, saponins and tannins which have any potency as traditional medicinal. This study aimed to examine the ethanolic extract of Neem leaves on kidney microanatomy in mice (Mus musculus L.). Completely Random Design consists of 4 treatments, 5 replications, namely K (aquadest), P1, P2 and P3 (Treated by dosages 8.4; 11.2 and 14 mg/kg BB/day). The treatments were given orally with volume 0.2 mL for 21 days. Kidney organ was isolated, weighed and made by histological processed with a 5 μm incision thickness using the paraffin method with Hematoxylin-Eosin staining. Observation of preparations were carried out to calculate glomerular diameter, Bowman’s capsule diameter and Bowman’s space diameter. The data were analyzed using ANOVA test at 95% confidence level and continued with DMRT test. The analysis results showed that there was significant affect (p<0.05) on exposure ethanolic extract of Neem leaves to the glomerular and Bowman's capsule diameter but didn’t show significantly affect (p>0.05) to the Bowman's space diameter, changes in kidney weight and drinking consumption of mice. It could be concluded that the exposure of ethanolic extract of Neem leaves in a period of 21 days is still safe to use as traditional alternative medicine. Abstract

Neem leaves are one of the traditional medicinal plants (Azadirachta indica A Juss). Neem leaves contain an activecomposition as an antifertility such as azadirachtin, flavonoids, saponins, nembeen and nimbidin. SpermatogenicIndex of male mice (Mus musculus L) after being given ethanol extract from the leaves of the mimba (Azadirachtaindica A. Juss). This study used a completely randomized design (CRD) with 2 control groups (aquades) andtreatment (giving the leaves of the mangosteen ethanol extract at a dose of 14 mg / KgBB / day) with 15 replications.The treatment was given orally with a volume of 0.2 ml for 21 days. The 22 days of the testes were isolated, weighedand made histological preparations using a paraffin method with a thickness of 5 μm, and staining of HematoxylinEosin (HE). Observation of seminiferous tubule structure and measurement of seminiferous tubule diameter wascarried out in 4 fields of view. The Spermatogenic Index values (SI) was determined by the number of phases ofeach field specified in the seminiferous tubules. Data were analyzed by t test with a confidence level of 95%. Theresults of the analysis showed not significant different on body weight, testicular weight and diameter of theseminiferous tubules. Spermatogenic Index observations showed that the control group had a dominant fase of 5while the treatment group had dominant fase of 4.The research concluded that the administration of mimba leavesethanol extract was recommended as an antifertility of male animals. Abstract

Pineapple peel vinegar containing acetic acid and antioxidants has the potential to helpimprove the structure of beta cells in the Langerhans islet. This study aims to examine the effectiveness of pineapplepeel vinegar towards the histological improvement of Langerhans islet in diabetic rats. Twenty-four rats weredivided into 6 groups, normal control, positive control (diabetes + 0,4 ml apple vinegar), negative control(diabetes + water), test groups dose I, II, and III (0,2 ml; 0,4 ml; 0,8 ml pineapple vinegar). The treatment groupthat given 0.4 ml pineapple peel vinegar showed improvement in islet's structure as indicated by its size andnumber that close to normal. The results of the statistical analysis using Kruskal-Wallis followed by Mann-Whitneyshowed that the diameter of the Langerhans islet in group P2 (treated by 0,4 ml pineapple peel vinegar) was closeto normal. The improved structure of the Langerhans islet resulted in a decrease in blood sugar levels by 41,63%.This percentage is still lower than the K+ group (treated by 0,4 ml apple vinegar) by 52,89%. The antioxidantactivity in vinegar improves the structure of the Langerhans islet by binding to free radicals that cause damage.The acetic acid in vinegar inhibits disaccharidase activity so that it can control blood sugar levels.Keywords: Diabetes mellitus, alloxan, pineapple peel vinegar, antioxidants, acetic acid

Abstract

Diabetes mellitus is a metabolic disorder characterized by increased levels of glucose in the blood (hyperglycemia). The increasing number of diabetics in the world encourages the discovery of certain drugs that are effective and relatively safe for managing diabetes. One alternative medicine that can be used is vinegar. Pineapple skin can be an alternative raw material for making vinegar because it contains a large amount of sugar and contains acetic acid and various secondary metabolites that can reduce blood glucose levels. The purpose of this study was to determine whether pineapple skin vinegar has antidiabetic activity. The test was carried out by dividing each of the 4 rats into six groups namely normal control, positive control, negative control, and three test groups that were given pineapple skin vinegar as much as 0.2, 0.4, and 0.8 cc. The treatment was carried out for 28 days, starting 4 days after induction of an alloxan dose of 150 mg / kg body weight. Measurement of blood glucose and body weight of rats were carried out before induction, after induction, and on the 7th, 14th, 21st, and 28th day, while the measurement of feed consumption was carried out every day starting from the day of pineapple skin vinegar administration. The results of the analysis showed that the administration of pineapple skin vinegar by 0.4 cc with acetic acid levels of 1.43% can reduce blood glucose levels most significantly that is 41.63%.Keywords: vinegar, pineapple skin, acetic acid, antidiabetic

Abstract

Neem leaves is one part of the neem tree (Azadirachta indica) containing antifertility compounds which can suppress the rate of male rat reproduction. The purpose of this study is to examine Gonadosomatic Index after exposure of neem leaves ethanol extract as antifertility compounds. This study is an experimental study with a Completely Randomized Design (CRD), using 12 male rats devided into 4 treatment groups and 3 replications. P0 is a control with aquades treatment, P1, P2, and P3 (neem leaves ethanol extract with each dose of 60, 80, and 100 mg/kgBW/day). The treatment was administered orally wiht a volume of 1 ml for 14 days. The main parameters are testicular weight, body weight and GSI. Body weights were measured every 7 days. Surgery was done after the treatment ends for measuring testicular weight and testicular isolation continued by measuring GSI values. GSI value is determined from body weight and testicular weight. The results were analysed using ANOVA at 95% confidence level. The analysis results showed that there were no significant differences (P>0.05) in the body weight and testicular weight parameters, but there were a significant different (P<0.05) in GSI values. The test results of GSI values show the different decrements in the treatment of P2. Based on the results of the test, it could be concluded that the exposure of neem leaves ethanol extract at doses of 60, 80, 100 mg/kgBW/day for 14 days of exposure time can cause antifertility effect in male rats. Abstract

ABSTRACT Fenugreek is one of the herbal plants whose seeds contain flavonoid antioxidant compounds, essential amino acids and minerals but their effect on liver function has not been tested. This study consisting of 4 treatment groups, namely P0 (white rats were given aquadest), P1 (white rats were given fenugreek seed extract at dose of 500 mg/kg BW), P2 (white rats were given fenugreek seed extract at dose of 1000 mg/kg BW), and P3 (white rats given fenugreek seed extract at dose of 1500 mg/kg BW). Statistical analysis test used Kruskal Wallis and continued with Mann Whitney test. The results showed that the administration of fenugreek seed extract on the highest hepatosomatic index was found in research rats treated with a dose of 500 mg/kg BW, the highest hemoglobin levels were found in research rats treated with a dose of 1500 mg/kg BW and the highest liver protein levels were found in research rats. treated with a dose of 500 mg/kg BW. The conclusion of this study was that exposure to fenugreek seed extract was able to maintain liver function by showing the value of hepatosomatic index, hemoglobin and liver protein levels in white rats.

Abstract

The high-fat diet can increase free radicals, which can damage the intestinal histological structure. The ethanolic neem leaf extract contains antioxidants that have the potential to help improve the intestinal histological structure. This study aims to examine the effectiveness of ethanolic neem (A. indica A. Juss) leaf extract on the histopathological structure of the small intestine in rats (R. norvegicus L.) hyperlipidemia. This study used 24 rats divided into 6 groups: normal control, positive control, high-fat diet + simvastatin, the test groups dose I, II, and III (ethanolic neem leaf extract 75 mg/200 gBW, 100 mg/200 gBW, and 125 mg/200 gBW). The results of the analysis using ANOVA test on the height of villi, mucous thickness, and number of goblet cells in the small intestine part duodenum, jejunum, and ileum showed no significant difference (p>0.05). The results of the morphology of epithelial cells in treatments P0, P2, and P5 showed normal epithelial cells, whereas treatments P1, P3, and P4 showed erosion epithelial cells. Based on the result, the ethanolic neem (A. indica A. Juss) leaf extract can suppress the damage effects of epithelial cells and the histological structure of the small intestine in rats (R. norvegicus L.) hyperlipidemia
