Abstract

Keragaman genetik suatu populasi dapat memberi petunjuk mengenai keadaan populasi di masa mendatang. Variabilitas genetik suatu populasi dapat diamati pada tingkat protein (isoenzim) dan tingkat DNA (Deoxyribo Nucleic Acid). Pada tingkat DNA, variabilitas genetik populasi dapat diungkap dengan menggunakan marka molekul mikrosatelit.Pada saat ini, DNA mikrosatelit banyak digunakan sebagai marka molekul untuk mempelajari variasi genetik. DNA mikrosatelit merupakan rangkaian molekul DNA pendek yang susunan basanya berulang dan terdapat melimpah dalam genom eukariot. Penelitian ini menggunakan satu lokus mikrosatelit yakni D13S765 untuk mengkaji variabilitas genetik populasi monyet ekor panjang di Pura Pulaki yang meliputi jumlah alel, nilai frekuensi alel dan nilai heterozigositas. Sejumlah 12 sampel darah dikoleksi sebagai sumber DNA. DNA total diekstraksi dengan menggunakan QIAamp DNA Blood Mini Kit dari Qiagen. Lokus mikrosatelit D13S765 diamplifikasi dengan teknik Polymerase Chain Reaction (PCR). PCR dilakukan sebanyak 30 siklus dengan suhu annealing 540C. Alel dipisahkan secara elektroforesis pada gel poliakrilamid 7 % dan alel dimunculkan dengan pewarnaan perak (silver staining).Hasil penelitian menunjukkan bahwa sejumlah 6 alel ditemukan pada lokus D13S765 di populasi monyet ekor panjang Pura Pulaki dengan panjang alel berkisar 238-258 panjang basa. Frekuensi alel bervariasi, alel 254 (0,38) memiliki frekuensi tertinggi disusul alel 250 (0,21), alel 258 (0,21), alel 242 (0,12), alel 246 (0,04) dan alel 238 (0,04). Heterozigositas populasi monyet ekor panjang Pura Pulaki menggunakan lokus D13S765 sebesar h=0,78. Dari hasil penelitian dapat disimpulkan bahwa lokus D13S765 bersifat polimorfik pada populasi monyet ekor panjang Pura Pulaki

Abstract

The purpose of this study was to know the resistant pattern of Salmonella typhi (S. typhi) isolated from wolf fish (Hoplias malabaricus) to antibiotics in Bali Safari and Marine Park. S. typhi was isolated from fecal swab collected from the illness fish. The S. typhi was confirmed with MicrogenTM GN A ID test’s method. The antibiotic sensitivity of the S. typhi was tested using streptomycin, gentamicin, penicillin, vancomycin, and chloramphenicol according to the Kirby bouer method. The results showed that the isolate of S. typhi was resistant to penicillin and vancomycin, but sensitive to streptomycin, gentamicin, and chloramphenicol.

Abstract

This study aims to determinate the effect of combination golden snail powder and fermented coconut cake on quality of duck eggs. We were using experimental feeds combination form golden snail powder and fermented coconut cake as raw material for livestock rearing for duck. This research was conducted in July - September 2018 in Mangkoso Village, Barru Regency and continued with sample testing at the Laboratory Animal Husbandry Faculty of Livestock Product Technology University of Hasanuddin. This study used 16 female duck who are 20 weeks old with four types of treatments consists of four replications of each treatment. The egg quality was carried out using a sample of 16 eggs from Ducks which was maintained and treated with different rations. The data obtained were analyzed using variance analysis with a Completely Randomized test for significant different results. The results of the combination of golden snail powder and fermented coconut cake on egg color, egg white index, egg yolk index, haugh unit (HU) value of egg units. All treatments (P1, P2, P3) gave effect to three parameters, namely eggshell quality, egg yolk index and egg yolk color, while the other three parameters namely egg weight, egg white index and Haugh Unit (HU) value did not differ significantly from negative controls. The best treatment was shown by treatment P3 which gave an effect on the heavier egg weight than other treatments, eggshell quality grade A, Egg White Index and Egg Yolk Index showed good quality and bright golden egg yolks.

Abstract

ABSTRACT: A pet such as a domestic cat (Felis Catus) with a sterile male sex aged 1 year 7 months with a body weight of 3.7 kg where the animal is affected by urinary system disorders namely Nephritis. Nephritis is kidney inflammation that can occur in the glomerulus, pyelum or tubules. Clinical symptoms shown in patients include polyuria, decreased appetite, and vomiting. For the diagnosis of Nephritis clinically evaluated based on changes in urination behavior, palpation of the dorsal abdominal caudal thoracic is temporarily labored based on a urine dipstick test containing protein ++ (± 100 mg / dL), leukocytes valued at +, urine specific gravity of 1,015 while based on ultrasound examination thickening of the renal cortex, medullary and cortical border is not apparent, renal size is 4.21 cm, kidney capsules that appear hyperechoic, and ascites in the abdomen. Cats treated with isotonic fluid infusion fluids, wet and dry diet diets, antibiotics, multivitamins, and anti-inflammatory. Inpatient animals for 4 days showed improvement in clinical symptoms, namely drinking quite well, starting to stand up, there was movement for activities, and urine was clearer.

Abstract

Neutrophils are one of the most abundant cells of the immune system and are very active during the wound healing process. This study aimed to determine the description and comparison of neutrophil cells after treated sugar and honey in the incision wounds of domestic cats (F. domestica). 15 samples were used with treatment for 14 days and the preparations were given topically. The treatment group consisted of 5 groups, namely Negative control group (NCG) with normal wound healing (without treatment), Positive control group (PCG) given Bioplacenton ointment, treatment group 1 (TG1) used sugar, treatment group 2 (TG2) used honey, Group Treatment 3 (TG3) used combination of sugar and honey. Skin samples were taken on the 4th, 9th, and 14th day of each group to make histopathology preparations and observation under a microscope by assessing the number of neutrophil cells with an indicator score of 0 - 5. Treatment Group 1 (TG1 ) used sugar shown the best decrease in the number of neutrophil cells with the development of the same healing process like as Treatment Group 3 (TG3) used a combination of sugar and honey, while Treatment Group 2 (TG2) used honey shown also a decrease in the number of neutrophil cells. The three treatment groups showed a better healing process than Negative control group (NCG) without treatment and showed almost equivalent results to the Positive control Group (PCG) used Bioplacenton ointment. Abstract

The reality at SMA Negeri 1 Tondano shows that there are still many TEACHERS who don't even know what the Mendeley or Turnitin applications are, even though as a teacher they also have to be involved in the world of literacy, especially regarding scientific publications which are also needed for promotion. The activity method that will be carried out in the Community Partnership Program (PKM) is practicum and direct demonstrations using Mendeley and Turnitin applications. The results of the service show that for the assessment tool there is an increase in data interpretation by 60.9%. This activity must be carried out every year to facilitate the creation of the right tools for teachers to assess their students, so that they continue to increase the professionalism of teachers in developing learning assessments. At the end of this training activity, participants gave positive feedback on the activity with a data interpretation of 69.6%.

Abstract

This service is carried out to improve the competency of students in the Unima Sociology Education Study Program, especially as well as students outside Unima who are interested in taking part in this training with the aim of being able to understand how to use the Filmora editing application to create learning videos and in fact this competency can also be used as a basis for becoming a content creator. The methods used in this activity include lectures, discussions, questions and answers and of course direct practice. After carrying out the training, it can be concluded that there are several steps in implementing this training, namely making and distributing electronic pamphlets, several things directed by the lecturer regarding tools and materials that must be prepared by participants are (bring a laptop, install the Filmora application on the participant's laptop, prepare a learning video will be edited), implementation of training (checking participants' software and hardware completeness, opening the Filmora video editing application, uploading videos, adding videos to projects, adding video text, dividing videos, adding filters, adding overlays, adding elements, adding split screens, adding sound background, exporting video).

Abstract

This study aims to develop students' podcast skills as part of teaching variations to enhance digital communication capabilities in the industrial revolution 4.0 era. The method employed a structured approach consisting of four main stages: preparation, implementation, practice, and evaluation. The program was conducted at the Sociology Education Study Program, Manado State University, involving 25 students as participants. The preparation stage included needs assessment and learning module design. The implementation stage focused on theoretical understanding and technical skills development. The practice stage involved group projects where students created their podcast episodes, while the evaluation stage assessed both technical and content aspects of the produced podcasts. Results showed significant improvement in students' podcast skills, demonstrated by the increase in test scores from an average of 31.72 in the pretest to 80.56 in the posttest. The evaluation revealed that students successfully developed competencies in podcast production, from conceptualization to final editing. Additionally, participants showed enhanced abilities in digital communication, teamwork, and content creation. This program proved effective in developing students' digital communication skills while providing practical experience in modern content creation.

Abstract

Background: Hemorrhagic shock is a condition that causes rapid and significant loss of intravascular blood volume, further leading to hemodynamic instability, decreased oxygen delivery, decreased tissue perfusion, cellular hypoxia, organ damage, and death. In conditions of hemorrhagic shock, measures are needed to prevent death in both humans and animals, one of which is by providing fluid therapy such as lactate ringer and gelatin. The purpose of the study: to see the effect of resuscitation of lactate and gelatin ringer fluid from the aspect of animal blood profile includes total erythrocytes, hematocrit (HCT) and hemoglobin (Hb). Research method: the sample in the study using male rabbits as many as 12 rabbits was divided into four groups: The negative control group (NC) was a healthy group without bleeding, the positive control group (PC) was the group given the bleeding treatment, the treatment group 1 (TG1) was the bleeding group and was treated with lactate ringer, and the treatment group 2 (TG2) was the bleeding group and treated a combination of lactate ringer and gelatin. Rabbits are bleeding with a presentation of 30% until they reach a state of hemorrhagic shock then shock is maintained for 45 minutes before resuscitation of fluid and for 60 minutes given fluid resuscitation. The results of the study: showed that the NC, PC, TG1, and TG2 groups showed a decrease in HCT values of 1.6%, 11.3%, 9% and 12%, respectively. Meanwhile, Hb values in rabbits decreased on average to 22 gr / dl, 13 gr / dl, 24.4 gr / dl and 20.9 gr / dl. Meanwhile, the total erythrocyte value showed a decrease in the average value reaching 2.48 x 106 / μL, 3.16 x 106 / μL, 4.2 x 106 / μL, and 3.9 x 106 / μL. The decline that occurs in the NC group is normal because the decrease that occurs is influenced by blood sampling, but in the PC, TG1, and TG2 groups are affected by the bleeding process. Based on the results of the study, it shows an increase in the indicators of Hb and Total Erythrocytes, this shows that giving fluid resuscitation to rabbits who experience hemorrhagic shock gives better results to the blood profile picture. Abstract

Feline Lower Urinary Tract Disease (FLUTD) is a term used to describe a condition that can affect the urinary bladder and lower urinary tract. Cystitis is an inflammatory disease of the urinary bladder caused by infectious and non-infectious agents, namely urine crystals. The occurrence of crystals in the urinary tract often occurs in cats that consume high-mineral and calcium feed without adequate water intake. The cat case complained of not urinating for three days, abdominal distension, and pain in the urinary bladder. Examination methods with routine hematology, ultrasonography, native tests, urine dipsticks, bacterial cultures, and native tests were carried out at the Unhas Veterinary Teaching Hospital. The diagnosis of Cystitis was confirmed by laboratory examination results found struvite crystals found >10 crystals/HPF, ultrasound examination showed hyperechoic appearance in the urinary bladder, urine dipstick test pH 7.5 and protein +++, hematology results found an increase in MCV values ​​and a decrease in Hb, MCHC and PCT indicators. . Handling of this case with catheter installation and fluid therapy. Treatment with antibiotics, anti-inflammatories, and supportive drugs vitamins and immune boosters.
