Abstract

Mare’s milk has good nutrient composition for human being in the form of natural milk or milk products. It can be used to replace cow’s milk, especially in the regions outside of Java Island which are rarely found dairy cow. This study had an objective to develop fermented mare’s milk by using mixed cultures of probiotic bacteria i.e. Lactobacillus acidophilus (A), Bifidobacterium longum (B), and Lactobacillus casei (C). The cultures of two probiotic bacteria AB and BC had been developed as well as three probiotic bacteria ABC culture. The mixed cultures (AB, BC and ABC) were prepared in single culture then cultivated in mixed culture as total 10% v/v of mare’s milk used and was incubated at 39°C for 9 h. The pH, acidity and bacterial count, each of them was analyzed in every 3 h of incubation time, while the organic acid and sensory tests were conducted at the end of 9 h of incubation. The results showed that the growth of mixed probiotic bacteria culture BC and ABC had better pH value that was around 3 compared with AB around 4.5. This showed that the cooperation between bacteria was different in each different combinations. The number of bacteria also increased sharply between 3-6 h of incubation time and 6-9 incubation time (AB and BC) along with the increased lactic acid, but the acetic acid decreased from 1750 mg/L to 1500 mg/L. The result of sensory test showed low acceptability of trained panelists. It is concluded that mixed cultures, using two or three probiotic bacteria (L. acidophilus, B. longum, and L. casei) could grow in mare’s milk. The change of the biochemical patterns indicated a commensalism cooperation among bacteria used. It was therefore the fermented mare’s milk using these probiotic bacteria were not well accepted by entrained panelists.

Abstract

Kajian kasus ini ditujukan untuk mengungkap fenomena klinis dan reproduktif yang dialami 5 ekor kambing Bligon betina umur 2-3 tahun yang dipelihara oleh petani di Yogyakarta. Menurut informasi dari pemilik, kambing mengalami kekurusan dan tidak pernah menunjukkan gejala berahi. Hewan sudah diobati dengan ivermectin secara berkala. Pada hewan tersebut selanjutnya dilakukan kajian manajemen pemeliharaan serta observasi dan pemeriksan klinis (pemeriksaan fisik, uii berahi, pemeriksaan sitologi vagina terhadap organ reproduksi)dalam kurun waktu sekitar 60 hari (akhir Juli- awal September 2019). Pada akhir periode observasi hewan diberi perlakuan gertak berahi dengan pemberian injeksi PGF2-alfa dua kali dengan selang 11 hari dan pemeriksaan USG. Hasil pemeriksaan klini hewan tidak bunting, tidak ditemukan adanya ekto dan endoparasit, tidak ditemukan adanya perubahan fisik, dan tanda-tanda penyakit infeksi. Selama masa pengamatan hewan mendapat pakan berupa jerami kangkung pada level sekitar 2% bobot badan, pertambahan bobot badan harian yang negatif atau rendah, BCS buruk (1-1,5 dalam skala 1-5), tidak ditemukan berahi, gambaran sitologi bagina didominasi sel parabasal dan transisional, respon terhadap pemberian preparat PGF2-alfa tidak menunjukkan adanya perubahan fisik alat kelamin, perilaku berahi, dan gambaran sitologi apus vagina. Hasil pemeriksaan USG tidak menunjukkan adanya status ovarium yang aktif. Hewan didiagnosa mengalami Undernutrition dan anestrus. Berdasarkan pemeriksaan fisik dan pendukung dapat disimpulkan faktor lingkungan berupa asupan pakan yang rendah dan kondisi tubuh yang buruk berpotensi mengakibatkan gangguan reproduksi pada kambing Bligon betina di masa usia produktif.

Abstract

The study aimed to describe lactation characteristic of Etawah Crossed Breed goats raised under intensive management. The study located in BBPTU HPT Baturraden, Purwokerto, used 27 female pregnant goats, at 24 to 27 months old. The goats were managed followed standard operating procedure of the research institute. The data consisted of body size and body weight, colostrum and milk yield, lactation period and reproductive performance. The results showed the average of daily milk yield, lactation period, total milk and colostrum production in first lactation were 960±340 mL/day, 157±41 days, 114,720±68,900 mL and 415±240 mL/day. The proportion of monthly milk production from initial lactation to dry off were 21.27, 22.17, 20.18, 17.29, 12.52, 9.13, and 5.65% of total lactation yield, respectively. Peak production ranged from first to second months of lactation, the highest was in the week 4th, averaged of 1080 mL/day. Production persistency was 66.39%. The rate of increase toward peak was 5.60%/week and rate of decline from peak was -3.92% of milk yield in the previous week. Trend equation of lactation curve was Y = 8412.7 –233.65 x. Means period of postpartum mating, service per conception and body condition score were 88.7±39.2 days, 1.2 and 3.45, respectively. There was no correlation between postpartum mating, body condition score and milk production. The study concluded that Etawah Crossed Breed goat under intensive management had short lactation period, early peak production, low persistency. The dry period was estimated 3 months and pregnancy occurred after peak production.

Abstract

Susu kambing memiliki komponen protein salah satunya protein β dan secara umum terjadi polimorfisme pada level protein. Perubahan urutan asam amino akibat polimorfisme memungkinkan adanya potensi dihasilkannya peptida bioaktif penghambat enzim pengubah angiotensin (ACEi). Penelitian ini bertujuan untuk menyaring peptida bioaktif yang berpotensi sebagai ACEi dari kasein β kambing beserta polimorfismenya. Penelitian ini dilakukan dengan teknik in silico terhadap sekuen kasein β kambing serta struktur tiga dimensi human testicular ACE. Langkah yang dilakukan dalam penelitian ini meliputi simulasi pemotongan peptida dengan enzim pencernaan (pepsin, tripsin dan kimotripsin), peninjauan karakteristik peptida lalu simulasi docking ligan-reseptor. Tampilan parameter Lipinski’s Rule of Five (Ro5), bioaktivitas dan energi afinitas dipertimbangkan untuk memilih peptida bioaktif. Hasil yang didapat menunjukkan bahwa ditemukan peptida bioaktif yakni INK (Ile-Asp-Lys) yang memiliki kemampuan hampir setara dengan lisinopril (afinitas energi -8,2kkal/mol vs. -8,3kkal/mol). Peptida INK dapat ditemukan dari hasil hidrolisis dari alel A, C, D dan E, sehingga polimorfisme tidak menyebabkan perbedaan produksi peptida bioaktif. Kesimpulan yang dapat diambil yakni kasein β susu kambing jika dicerna dengan enzim pencernaan dapat menghasilkan peptida bioaktif ACEi yakni INK.Identification of Angiotensin Converting Enzyme-inhibitor (ACEi) Bioactive Peptide from Goat Milk β-Casein with It's Polymorphism by In Silico TechniqueAbstractPolymorphism eventually may be occurred at the protein level. Changes in the amino acid sequence due to polymorphism may exhibit a potential action to generate of the angiotensin-converting enzyme inhibitors (ACEi) bioactive peptide. This study is aimed to assess bioactive peptides that have a great potent value as ACEi from goat β casein along with its polymorphism. The research was done by in silico technique on goat β-casein sequence and three-dimensional structure human testicular ACE. Peptide-cutting simulations with digestive enzymes (pepsin, trypsin and chymotrypsin), peptide properties review, then ligand-receptor docking simulations was applied in this research. Appearance of Lipinski's Rule of Five (Ro5), bioactivity and affinity energy were considered for selecting bioactive peptides. The results show that bioactive peptide found as INK (Ile-Asp-Lys) which had similar ability as lisinopril (energy affinity –8.2kcal/mol vs. –8.3kcal/mol). The INK peptides could be found from the hydrolysis resulted in alleles A, C, D and E, therefore polymorphism did not affect the differences of production of bioactive peptides. A conclusion, processed goat milk β casein with digestive enzymes could produce ACEi of INK as bioactive peptide.

Abstract

This study aimed to evaluate the body weight (BW), morphometry, and physiological status of Etawah Crossbred Doeling (ECD) under different farms management. This study was performed in two farms, Garage (Tlogoadi village, Mlati subdistrict) and Bumiku Hijau (Margomulyo village, Seyegan subdistrict), both located in Yogyakarta and each farm used 6 ECD as the replications (12 animals in total). Parameters of the study consist of body weight (BW), morphometry (chest girth, hip height, body length, chest depth, ear length, ear width, and withers height), physiological status (respiration, pulse, and rectal temperature). The parameter was measured weekly for four consecutive weeks of observation. The results showed that the difference in farm management between Garage and Bumiku Hijau influenced on the BW, morphometry, and physiological status of ECD. The study showed that the BW and all morphometry from the Garage farm was higher than Bumiku Hijau farm (P<0.05). However, the physiological status (except for rectal temperature) from Garage farm was lower than Bumiku Hijau farm (P<0.05). It can be concluded that Garage farm implemented better good farming practices compared to Bumiku Hijau farm.

Abstract

Butter is a dairy product that is susceptible to oxidation, which causes a rancid taste and a short shelf life. The quality of the butter depends on the quality of milk. The aim of the study is to determine the quality of butter made from goat and cow reared by small-scale farmers in Yogyakarta with the traditional manufacturing process. The milk of Saanen-Peranakan Ettawa (Sapera)  goats and Fresian Holstein were taken from farmers in Sleman district, Yogyakarta, Indonesia. Butters were sampled after 0, 15, 30 days storage. This study used the Nested Design method , namely the nested quality of butter in the type of milk. The result showed that butter made from cow milk produced a higher fat content compared to goat milk butter (P<0.05), namely 85.29 ± 0.04% and 80.10 ± 0.27%. The acid value increased while pH value was decreased during storage (P<0.05). There was no increase in peroxide value in cow butter but there was an increase in goat butter on day 0 to day 15 (P<0.05). It can be concluded that the goat and cow milk reared by small-scale farmers in Yogyakarta have good quality so it produced good quality butter which is still good for consumption until 30 days of storage in the refrigerator.

Abstract

Beta-casein in milk is known to be a bioactive peptide producer because of its amino acid sequence. Bioactive peptides have prospected molecules that can adhere with SARS-CoV-2 spike protein, so they can inhibit the virus from hooking up with human cell receptor protein. The research is aimed to find any peptides from goat’s milk beta-casein that are prospective candidates as SARS-CoV-2 spike protein inhibitors. Goat’s milk beta-casein was simulated as being digested by the digestive tract. Pepsin, trypsin, and chymotrypsin enzymes cut the beta-casein amino acids sequence into small peptides. Then, their bioavailability was predicted by Lipinski’s Rules of 5 (Ro5), any most fitted peptides to the rules will be simulated to dock to SARS-CoV2 spike protein besides Curcumin as the control ligand. Peptides with the best bind activity with the spike protein will be selected as inhibitor candidates. Peptide QPK is selected as a SARS-CoV-2 inhibitor candidate because it has better affinity energy than Curcumin or other selected peptides.

Abstract

Many players in the halal industry do not have access to formal finance. As a result, they do not have access to capital, their business does not develop, and gradually goes out of business. This article aims to develop the concept of financing with the Rahn contract scheme. So far, the Rahn contract is only understood as an agreement in a pawnshop to obtain a certain amount of loans for consumptive purposes. This article uses a type of literature review where data is obtained from a number of publications derived from journals, books, proceedings, review results, magazines or newspapers, and other sources that are considered informative. This article argues that the Rahn contract can be developed as a capitalization solution for halal industry business units in the micro, small, and medium sectors. It further suggests the development of the function of the Rahn contract as a business capitalization contract by issuing shares as Marhun for financing media. The capitalization model with this scheme is intended to develop collateral as a means of investment.

Abstract

Cheese coagulants from plant origin have emerged as promising alternatives to animal rennet in cheese production. Among them, latex from Ficus carica L. has shown potential as a natural milk-clotting agent for goat cheese production. This study aims to determine the optimal incubation time and fig tree latex extract concentration on the quality of goat cheese. The optimization of the production process of the goat cheese was carried out using Response Surface Methodology (RSM) with Central Composite Design (CCD). The independent variables were the concentration of the Ficus carica L. latex extract (0.2-0.5 mL/100 mL of milk) and the time of incubation (60-120 min). The goal was to achieve maximum protein content (%), ash content (%), and calcium content (%). The results of this study showed that the Ficus carica L. latex extract used in the study could coagulate milk with a strength of 354.69 MCU and a dominant protein molecular weight of about 20 kDa. The optimization results using RSM method with CCD design showed that the goat cheese was produced optimally in 0.5 mL/100 mL of milk for the concentration of Ficus carica L. latex extract, with 120 min incubation time. These optimal conditions resulted in a cheese with protein content of 18.56%, ash content of 2.32%, calcium content of 803.93 mg/100g, and desirability level of 0.93.
