Abstract

Inflammation is a normal protective response to tissue injuries caused by physical trauma, destructive chemicals, or microbiological substances. The purpose of this study was to determine the anti-inflammatory activity of papaya leaf water extract (Carica papaya L.) on the feet of male Wistar rats which were induced by carrageenan 1%. The research method used is experimental. The stages of this study include plant identification, making dry simplicia powder, simplicia characteristics, extracting papaya leaves using water, simplicia phytochemical screening and papaya leaf water extract (EADP), as well as testing for anti-inflammatory activity. Anti-inflammatory testing was carried out on 20 male Wistar rats induced with 1% carrageenan sub plantar. The test group was divided into five groups for the induction, comparison, EADP 50, 100, 200 mg/kg BW groups. The parameters observed were the volume of mouse leg udema measured by a pletismometer every 30 min for 5 h. Data obtained then calculated percent inflammation and percent inflammation inhibition. Furthermore, it was analysed by ANOVA with a confidence level of 95%. The results of the simplicia characteristics showed that the water content, water content of the sea water and soluble extracts of ethanol were 8.9%, 51.4% and 10.2% respectively. The results of simplicia and EADP phytochemical screening showed the content of flavonoids, tannins, triterpenoids, glycosides, and alkaloids. Observations of the percentage of inflammation in the induction group had the highest percentage of inflammation compared with the other test groups at time of 300 min (p 0.05). Whereas the percentage of inflammatory inhibition of all groups of test extracts has effectiveness comparable to the comparison group. EADP group 200 mg/kg BW is the best dose of extract as anti-inflammatory, where flavonoid compounds are thought to be responsible for decreasing the volume of inflammation in the legs of test animals.

Abstract

Cinnamon bark (Cinnamomum burmanni) is one of the cheap and easy traditional medicinal herbs and is widely used to treat toothache. The largest content of cinnamon bark is essential oil which contains the main compounds cinnamaldehyde (60.72%), eugenol (17.62%) and coumarin (13.39%). The content has potential as an antibacterial. Cinnamon bark extract affects the growth of Streptococcus mutans which is the main bacterium that causes dental caries (Puspita et al., 2013). This research was continued to the toothpaste formulation stage where people can use this plant as an anti-plaque comfortably. The development of extracts into paste preparations requires safety or toxicity data within a certain period of time. This study uses an experimental design. The test rats were divided into 4 groups consisting of the normal group (CMC Na 0.5%), the EEKM group at doses of 300 mg/kg BW, 2000 mg/kg BW and 5000 mg/kg BW. Toxic symptoms observed included behavioral changes such as tremors, slow walking, urination, defecation, motor ability tests such as stage tests, catalepsy, hanging and death. Changes in toxic symptoms were observed intensively every 30 minutes in the first 2 hours after administration of the test preparation and followed up to 14 days after administration. Based on the results of the study showed that EEKM at doses of 300 mg/kg BW, 2000 mg/kg BW and 5000 mg/kg BW did not show a toxic effect on the physical behavior of the test animals. EEKM is included in the practically non-toxic criteria, where at a dose of 5000 mg/kg bb there is no death, the LD50 value is greater than 5000 so it is included in the practically non-toxic criteria.

Abstract

Oral health has an important role in overall body health. This study aims to evaluate the antibacterial efficacy of toothpaste with cinnamon ethanol extract against Streptococcus bacteria in vivo, analyze changes in the number of bacterial colonies before and after using toothpaste, and identify the type of Streptococcus bacteria in dental plaque. This pure experimental research was conducted at the Pharmacy Laboratory of the Poltekkes Kemenkes Medan and the Microbiology Laboratory of the Faculty of Medicine, University of North Sumatra. Samples included cinnamon extract, toothpaste formulation with 12.5% extract concentration, and dental plaque from 40 respondents. Data analysis used descriptive univariate test with SPSS 22.0 application. Results showed Gram-positive bacterial infections, especially Rothia dentocariosa (24%), dominated dental plaque, followed by Streptococcus mitis (16.4%). The use of cinnamon-active toothpaste was able to significantly reduce the number of bacterial colonies, especially at high concentrations. In addition, active compounds in cinnamon extracts such as cinnamaldehyde and eugenol showed the ability to damage bacterial cell walls and inhibit biofilm formation. Gargling was also shown to be effective in reducing bacterial colonies by clearing plaque and food debris. The conclusion of this study suggests that Gram-positive bacterial infections in dental plaque require special attention in management with appropriate antibiotics, while gargling and the use of cinnamon-based antimicrobial agents can be an effective preventive strategy. These findings support the development of natural ingredient-based oral health products to improve infection prevention and the quality of oral health care. Abstract

The imbalance between high production and low excretion of uric acid in the body causes hyperuricemia, which can lead to gout. This disease is the second largest after stroke in Indonesia. Conventional therapy is not free from adverse effects. The tendency of society to go back to nature and the government's "Saintifikasi Jamu" program are efforts to explore safer alternative therapies. One of the plants that has the potential as an antihyperuricemia is bay leaves (Syzygium polyanthum Walp.). The purpose of this study is to optimize the use of scientifically based bay leaves as an antihyperuricemia agent. The research method used is experimental with a pre-posttest design . A total of 20 mice were divided into 5 test groups (induction, comparison, bay leaf extract group (EEDS) with doses of 100 mg/kgBW, 200 mg/kgBW, and 400 mg/kgBW. The effectiveness of antihyperuricemia was tested by inducing mice with 1% chicken liver juice and potassium oxonate. Uric acid levels were measured at T1, T7 and T14 days of the study using a Blood Uric Acid Meter. All groups of test extract doses showed effectiveness as agents to lower uric acid levels significantly compared to the induction group (p<0.05). The potential effectiveness of the antihyperuricemia extract showed an effect that was not statistically different from the comparison (allopurinol) (p>0.05) at all measurement points. The conclusion of this study shows that bay leaf ethanol extract has the potential as an antihyperuricemia agent, where a dose of 100 mg/kgBW showed the best effectiveness as an antihyperuricemia Keywords: Bay Leaves, Extract, Chicken Liver, Potassium Oxonate, Uric Acid
