Abstract

An understanding of the underpinning physiology and biochemistry of animals is essential to properly understand the impact of anthropogenic changes and natural catastrophes upon the conservation of endangered species. An observation on the tissue location of the key respiratory protein, myoglobin, now opens up new opportunities for understanding how hypoxia tolerance impacts on diving lifestyle in turtles. The respiratory protein, myoglobin has functions other than oxygen binding which are involved in hypoxia tolerance, including metabolism of reactive oxygen species and of the vascular function by metabolism of nitric oxide. Our work aims to determine whether myoglobin expression in the green turtle exists in multiple non muscle tissues and to confirm the hypothesis that reptiles also have a distributed myoglobin expression which is linked to the hypoxiatolerant trait. This initial work in turtle hatch Chelonia mydas confirms the presence of myoglobin transcriptin brain, heart and liver tissues. Furthermore, it will serve as a tool for completing the sequence and generating an in situ hybridization probe for verifying of cell location in expressing tissues.
The aim of this research was to gain the profile of blood gasses of green turtle (Chelonia mydas) hatch. Blood gas of the green turtle was analysed after exposuring them at 28 oC and 50% of humidity  for 24 hours in a pvc tube and at 34 oC under sunlight exposured with 47% of humidity for 30 minutes. The result showed the different values of blood gas contents. This result showed indication of metabolism activities and poikilothermic adaptation of green turtle hatch. Abstract

Green sea turtle – Chelonia mydas is a lung respiration animal that is able to dive and stay uder sea water for hours without needing to surface for oxygen. As oxygen supply in muscles is assured by myoglobin, we propose to study some characteristic of this muscle protein in green sea turtles. To achieve this objective, pure green sea turtle myoglobin has to be made available. Therefore, our first task is to purify this muscle protein from green sea turtles. Skeletal muscles from 3 green sea turtle hatchlings were studied microscopically and biochemically. Microscopy observation showed a general structure of striated muscle. Biochemical studies revealed that green sea turtle myoglobin could be more purely isolated to a certain degree by Sephadex G-75 gel filtration and purified by immunoaffinity gel chromatography rather than direct purification by DEAE-Sepharose ion exchange chromatography technique. SDS-PAGE analysis showed that green sea turtle myoglobin ran together with horse myoglobin as 17 kDa molecular weight proteins

Abstract

The aim of this research was to obtain the different characteristics of haemoglobin molecules in subjects under hypoxic condition, namely eel, catfish, suckermouth fish, green sea turtle using an electrophoresis technique. We used human umbilical cord blood and thalassemia patient blood, as well as a normal adult-human blood as controls. The proteins obtained after electrophoresis process were stained with two different colouring techniques, each based on different principles. Both staining techniques gave practically identical results. Subject that live in hypoxic condition has a different haemoglobin in comparison to the one found in adult human live in normal oxygen condition (normoxia). These hypoxia-adapted or -needed hemoglobin migrate slower than adult human hemoglobin from normoxia. This observation suggests that hemoglobin which is needed to live in hypoxic condition or environment is a different molecule. Whether this hemoglobin from hypoxic condition has a higher affinity to oxygen is not yet known.

Abstract

Mitochondrial gene mutation plays a role in the development of type two diabetes mellitus (T2DM). A point mutation in the mitochondrial gene Nicotinamide adenine dinucleotide dehydrogenase 1 (mtDNA ND1) gene mainly reported as the most common mutation related to T2DM. However, several studies have identified another SNP (single-nucleotide polymorphisms) in the RNA region of mtDNA from patients from specific ethnic populations in Indonesia. Building on those findings, this study aimed to use PCR and DNA sequencing technology to identify nucleotides in RNA and ND1 fragment from 20 Gorontalonese and 20 Javanese T2DM patients, that may trigger T2DM expression. The results showed successful amplification of RNA along 294 bp for all samples. From these samples, we found two types of point  mutation in Javanese patients in the G3316A and  T3200C points of the rRNA and ND1 gene. In samples taken from Gorontalonese patients, no mutation were found in the RNA or ND1 region. We conclude that T2DM was triggered differently in our two populations. While genetic mutation is implicated for the 20 Javanese patients, T2DM pathogenesis in the Gorontalonese patients must be traced to other genetic, environmental, or behavioral factors.

Abstract

ABSTRACT The research was aimed to analyzing the impact of 7 days arobic exercise in elevating rat cardiac muscle myoglobin. Experimental in vivo study on 20 male Wistar rat induced by 20m/ min treadmill aerobic exercise for 1, 3 and 7 days. Research was done in Animal house and protein laboratory, Biochemistry and Molecular Biology Department, FK UI and laboratory of Biochemistry and Molecular Biology, Biology Department FMIPA UNJ for 10 months. Myoglobin concentration was analyzed by spectrofotometry techique based on myoglobin Soret absorbance in ?503nm.  Myoglobin protein expression was assesed with Western blot technique. Standard curve measurement on myoglobin content with horse myoglobin shown that there was an increase on the absorbance value. The result  showedanthat the standar curve was able to be used as standard curve to calculate myoglobin concentration on cardiac muscle within 503 nm. Spectrophotometry myoglobin concentration measurement on Soret absorbance has shown increasing concentration on myoglobin since the first treatment day. Mean myoglobin concentration value has increased  on the 3rd and 7th day, with highest value on the 7th treatment day (0,0656 ± 0,0001). Western blot analysis showed ng on darker band on treatment group compared   on  to control. When aerobic exercise was given longer, the band shown darker means that myglobin content was higher. Myoglobin concentration can be measured by spectrophotometry on 503 nm. Aerobic physical exercise increased myoglobin on rat cardiac muscle since the first treatment day and  still increase oduring the treatment day. Highest level was shown on the 7th treatment day. Abstract
