Abstract

Tujuan penelitian tahap I adalah untuk menentukan level terbaik penggunaan fruktosa dalam pengencer Sitrat-Kuning Telur (S-KT) terhadap persentase motilitas, viabilitas, abnormalitas serta keutuhan membran plasma semen cair babi Landrace yang disimpan pada suhu 18oC dan dievaluasi setiap 6 jam. Rancangan penelitian pada tahap ini terdiri dari 5 perlakuan dan 4 ulangan yaitu sitrat-kuning telur (S-KT) tanpa Fruktosa (P0), S-KT + Fruktosa 2,5% (P1), S-KT + Fruktosa 5% (P2), S-KT + Fruktosa 7,5% (P3) dan S-KT + Fruktosa 10% (P4). Hasil penelitian menunjukkan bahwa kualitas semen segar cukup baik karena motilitas dan viabilitas spermatozoa masing-masing adalah 83,75±4,79% dan 92,70±0,14%. Tahap in vitro selama 24 jam dalam pengencer S-KT yan g suplementasi fruktosa 5% (P2) menunjukkan motilitas (40,00±4,08%) dan viabilitas (54,10±4,35%) lebih tinggi sehingga menunjukan perbedaan nyata (P<0,05) dibandingkan dengan bahan pengencer lainnya. Abnormalitas dan keutuhan membran plasma spermatozoa dalam keempat pengencer tidak berbeda (P>0,05). Penelitian tahap II untuk mengetahui persentase angka kebuntingan dan S/C. Persentase tertinggi angka kebuntingan dalam penelitian ini adalah P1 dengan nilai 80%, terendah pada P0 dan P3 yaitu 60%. Sedangkan nilai S/C terendah adalah P1 dengan nilai 1,25% dan tertinggi pada P0 dan P3 yaitu 1,67%. Kesimpulan dari penelitian ini adalah suplementasi fruktosa 5% dalam pengencer sitrat-kuning telur mampu mempertahankan motilitas dan viabilitas spermatozoa babi landrace di atas 40% selama 24 jam pengamatan dengan angka kebuntingan adalah 67,50%.Kata kunci: Semen babi, sitrat-kuning telur, motilitas, inseminasi buatan, kebuntingan.

Abstract

Tujuan dari penelitian ini adalah untuk mempelajari pengaruh penggunaan albumin dengan tingkatyang berbeda untuk memisahkan spermatozoa X dan Y sebagai media untuk memisahkan spermababi jantan. Sperma dipisahkan menggunakan tingkat yang berbeda dari albumin dengan 4perlakuan: rasio antara fraksi atas dan fraksi bawah: 10 dan 30%, 10 dan 40%, 10 dan 50%, 10 dan60% selama 1 jam. Hasil penelitian menunjukkan bahwa proporsi terbaik sperma X oleh fraksi disemua pelakuan, dan juga proporsi terbaik dari sperma Y oleh fraksi bawah pada semua lokasi.Proporsi X:Y dari semen segar (51,68: 48,32 ± 2,66) berbeda (P <0,05) dibandingkan dengan setiapperlakuan sampai fraksi atas (T1 – 74,02: 25,98 ± 5 , 65); T2 – 64,14: 35,86 ± 3,41); T3 – 62,29:37,71 ± 3,98), dan T4 – 59,26: 40,74 ± 4,33)) dan juga fraksi bawah (T1 – 38,13: 61,87 ± 7,69); T2– 35,48: 64,52 ± 6,11); T3 – 30,37: 69,63 ± 4,93); dan T4 – 26,41: 73,59 ± 4,54)). Viabilitas dankelainan sperma setelah dipisahkan tidak berbeda dalam setiap perlakuan. Konsentrasi spermasetelah perpisahan di fraksi atas lebih dari fraksi bawah di semua perlakuan. Sperma babi tidakterpisah selama serangkaian perlakuan dari proses pencucian hingga proses pemisahan dalammedia albumin yang mengakibatkan penurunan motilitas sperma dari 73 ± 3% menjadi 6,5 ± 0,58%pada fraksi atas pada perlakuan pertama sampai 3 ± 0,82% di fraksi bawah pada perlakuan keempatdi akhir pemisahan. Disimpulkan bahwa perlakuan terbaik untuk mendapatkan sperma X olehkonsentrat 10 dan 30% sedangkan sperma Y yang oleh konsentrat 10 dan 60%, sperma setelahperpisahan baik fraksi atas maupun fraksi bawah memiliki motilitas yang sangat rendah.

Abstract

A research was carried out to know the carrot influence cider different, as a source of antioksidant in citric thinner egg yolks against motility, viability and degrees acidity ( ph ) spermatozoa pigs and landrace, consisting of 5 treatment and 4 the test is as follows; P0 citric = 80 % + 20 % egg yolks, P1= citric egg yolks 99 % + 1.00 % cider carrot, P2= citric egg yolks 98 % + 2.00 % cider carrot, P3= citric egg yolks 97 % + 3.00 % cider carrot P4 = citric egg yolks 96 % + 4.00 % cider carrot. This research using random design complete (RDC), who analyzed by Analysis of variance (ANOVA) and followed by test duncan on the SPSS 13.0 version. The result showed that motility, viability and pH spermatozoa after dilution markedly dissimilar (P>0,05) between treatment.To motility viability best and treatment on p1. Treatment is capable of sustaining to observation to 28 hours worth motility 43,75 ± 2,50 %, viability 45,17 ± 5,21 %. On the parameter best ph treatment on P0 where until observation to 28 pH is 7,13 ± 0,13%. ABSTRAK Suatu penelitian ini telah dilaksanakan untuk mengetahui pengaruh level sari wortel yang berbeda, sebagai sumber antioksidan dalam pengencer sitrat kuning telur terhadap motilitas, viabilitas, dan derajat keasaman (pH) spermatozoa babi peranakan landrace, yang terdiri dari 5 perlakuan dan 4 ulangan tersebut adalah Sebagai berikut; P0 = sitrat 80 % + 20 % kuning telur, P1 = sitrat kuning telur 99 % + 1.00 % sari wortel, P2 = sitrat kuning telur 98 %+ 2.00 % sari wortel, P3 = sitrat kuning telur 97 % + 3.00 % sari wortel P4 = sitrat kuning telur 96 % + 4.00 % sari wortel. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL), yang dianalisis dengan Analysis of variance (ANOVA) dan dilanjutkan dengan uji Duncan menggunakan program SPSS versi 13.0. Hasil penelitian menunjukkan bahwa motilitas, viabilitas dan pH spermatozoa setelah pengenceran berbeda nyata (P>0,05) antar perlakuan. Untuk motilitas dan viabilitas perlakuan terbaik pada P1. Perlakuan ini mampu mempertahankan sampai jam pengamatan ke-28 dengan nilai motilitas 43,75±2,50%, viabilitas 45,17±5,21%. Pada parameter pH perlakuan terbaik pada P0 dimana sampai jam pengamatan ke-28 pHnya adalah 7,13±0,13.

Abstract

The aims of this study were to determine the best level of additional extracts Noni juice in the egg yolk citrate diluent on motility and viability of spermatozoa of landrace pig. The study was conducted following a completely randomized design with 5 treatments (P0 = 0%; P1 = 1%; P2 = 2%; P3 = 3%; P4 = 4%) and 4 times of ejaculated from two different landrace pigs. The treatments were placed on a cool box at a temperature of 17-19oC for further obserrvation of the motility and viability in every 4 hour intervals until the spermatozoa landrace pigs was<40%. Data were analyzed using SPSS software version 17. Additional of extracts noni juice in the egg yolk citrates diluent at level of 3% was significantly different for the average volume (212.50 ± 47.87 ml), motility (78.75 ± 4.79) and viability (84.23 ± 3.90) compared with the other treatments. In addition, a significant different between treatments were also observed for the concentration of spermatozoa (29.44 ± 250) and abnormality (14.99 ± 3.25). Motility of spermatozoa was observed after 32 hours of storage at the level of 3% of additional extracts noni juice in the egg yolk citrate diluent. However, there was no different between treatments on pH values. ABSTRAK Penelitian ini bertujuan untuk mengetahui level terbaik dari penambahan sari buah mengkudu dalam pengencer sitrat kuning telur terhadap motilitas dan viabilitas spermatozoa babi landrace. Penelitian mengunakan rancangan acak lengkap dengan 5 perlakuan dengan 4 kali ejakulat dari 2 ekor babi landrace yang berbeda. Perbedaan perlakuan dari pengencer sari buah mengkudu antara lain P0 (0%), P1 (1%), P2 (2%), P3 (3%), P4 (4%). Simpan perlakuan pada coolboox dengan suhu 17-19oC selanjutnya diamati setiap 4 jam sampai motilitas dan viabilitas spermatozoa babi landrace < 40%. Data dianalisis menggunakan software SPSS versi 17. Hasil penelitian semen segar yang diperoleh menunjukan volume rata-rata 212,50±47,87 ml pH 7,28±0,15 motilitas 78,75±4,79 viabilitas 84,23±3,90 konsentrasi 250±29,44 abnormalitas 14,99±3,25 yang digunakan, pengaruh penambahan sari buah mengkudu terhadap motilitas dan viabilitas nyata berbeda dengan perlakuan lainnya, namun tidak terdapat perbedaan yang nyata terhadap nilai pH. Pada penelitian level terbaik dengan penambahan sari buah mengkudu 3% dalam pengencer sitrat kuning telur dan motilitas dicapai pada jam ke 32 penyimpanan.

Abstract

The purpose of the present study was to determine the reproductive performance of landrace crossbreed sows which being maintained at the village level. Eighteen crossbreed landrace sows and one landrace-male pigs as a source of semen were used in this study following a randomized complete block design with three treatments and six replications. The treatments were: P1 (first and second parity); P2 (third and fourth parity); and P3 (fifth and sixth parity). The parameters observed were: length of estrus, service per conception rate (S/C), conception rate (CR) and litter size. Data were analyzed using Anova. There were no significant difference (P>0.05) between treatments on length of estrus, service per conception rate (S/C), conception rate (CR) and litter size. However, the results indicate that length of estrus of landrace crossbreed sows were: 3.16 days; 3.33 days and 3.16 days for P1, P2 and P3, respectively. The services per conception obtained were: 1.16 times; 1.33 times and 1.16 times for P1, P2, and P3, respectively. Furthermore, conception rates were: P1 = 83.33%; P2 =100% and P3 = 100%. Litter size obtained were: 8.4; 8 and 8.6 for P1, P2 and P3, respectively. ABSTRAK Suatu penelitian terhadap kinerja reproduksi babi betina yang berbeda paritas telah dilaksanakan di Desa Penfui Timur, Kecamatan Kupang Timur, Kabupaten Kupang selama (4) bulan. Tujuan dari penelitian ini adalah untuk mengetahui kinerja dari babi–babi induk betina peranakan landrace yang sedang dipelihara ditingkat peternakan rakyat. Penelitian ini menggunakan 18 ekor babi betina bangsa peranakan landrace dan satu ekor babi jantan bangsa landrace sebagai sumber semen untuk keperluan inseminasi. Penelitian menggunakan rancangan acak kelompok dengan tiga perlakuan dan 6 ulangan.Perlakuan tersebut adalah: P1 (paritas pertama dan kedua); P2 (paritas ketiga dan keempat); dan P3 (paritas kelima dan keenam). Parameter yang diamati yaitu: lama estrus, service per conception rate (S/C), conception rate (CR) dan litter Size. Data dianalisis dengan menggunakan anova. Hasil penelitian menunjukkkan bahwa lama berahi babi betina peranakan landrace yaitu P1= 3,16 hari; P2= 3,33 hari dan P3= 3,16 hari. S/C yang diperoleh adalah: P1= 1,16 kali; P2= 1,33 kali dan P3= 1,16 kali. Selanjutnya angka kebuntingan P1 = 83,33 %; P2= 100% danP3= 100%. Litter size P1= 8,4 ekor; P2= 8 ekor dan P3= 8,6 ekor. Hasil analisis statistik menunjukan bahwa perlakuan berpengaruh tidak nyata terhadap lama berahi, angka kebuntingan (CR), service per conception (S/C) dan litter size (P>0.05).

Abstract

This study aims to determine the effect of adding various doses of carrot juice into citrate-egg yolk diluents (C-EY) on bligon buck semen quality. Semen was collected twice a week using an artificial vaginal method of three buck with normal body condition and reproductive organs. Semen was 70% of motility, konsentration ≥1000x106mL and abnormality ≤ 15 diluted with C-EY added with carrot juice at a concentrations: 0% (CJ-0), 10% (CJ-10), 12.5% (CJ-12,5), 15% (CJ-15), 17.5% (CJ-17,5), and 20% (CJ-20), and stored in a refrigerator at 3-5o C. The quality of spermatozoa was evaluated every 24 hours. The Variables measured were spermatozoa motility, viability, and abnormalities. The results showed that spermatozoa preserved by C-EY supplemented by 17.5% carrot juice (CJ-17.5) had a higher quality (P<0.05) compared to the other four treatments, with motility (49.04 ± 1.0%), viability (55.70 ± 1.22%), dan abnormalities (4.57 ± 0.88 %) on the 4th day of storage. This study concludes that 17.5% carrot juice into citrate eggyolk extender improves the bligon buck semen

Abstract

The purpose of this study was to examine the effect of adding various concentrations of dragon fruit peel extract (DFPE) in Tris-egg yolk diluent (TEY) on the quality of bligon goat spermatozoa. Semen was collected twice a week using the artificial vaginal method from three year old bligon male goats with normal body conditions and reproductive organs. After the macroscopic and microscopic evaluation, good quality semen with a sperm motility of > 70%, a concentration of > 800 x 106 mL-1, and abnormality of < 15 %, was diluted with TEY + DFPE at a concentration of 0% (P0), 2% (P1), 4% (P2), 6% (P3), 8% (P4), dan 10% (P5). The semen which still had a motility ≥ 70% was then stored at a temperature of 3-5oC. Evaluation of the quality of spermatozoa was carried out every 24 hours until the motility decrease to 40%. The results showed that from day 1 to day 5 of preservation, spermatozoa preserved in TEY + 6% DFPE diluent (P3) had higher motility and viability (P<0,05) than other treatments. The abnormalities of spermatozoa until the 5 day of preservation were below 6%, but showed a significant difference (P<0,05) between treatments that received DFPE suplemention with control (P0). It was concluded that the addition of DFPE in TEY up to 6% could reduce the decrease in sperm quality of bligon goats during in vitro preservation.

Abstract

An experiment was carried out to recognize the effect of long equilibration time on the quality of spermatozoa and to obtain optimal equilibration time in the process cryopreservation of landrace pig semen. Semen was obtained from a landrace boar aged 2-3 years using glove hand method. The semen was than evaluated macroscopically and microscopically, good quality semen was diluted in a basic diluent, than kept for two hours at a temperature of 27-28oC. The semen were then centrifuged at 2000 rpm for 15 minutes, the supernatant was discarced and the pellets was re-diluted into basic diluent with a ratio of 1:1. Then, semen was diluted in treatment diluent with modified durasperm + 6% lactose + 4% glycerol with a concentration of 500 x 106/0.5 mL. Furthermore, the semen was packaged in 0.5 ml straws, then equilibrated at a temperature of 3-5oC appropriate to treatment, with 1 hour equilibration (T1), 2 hours equilibration (T2) and 3 hours equilibration (T3). After equilibration, the straws was frozen horizontally 5 cm above the liquid N2 vapor for 10 minutes at a temperature of ± -110oC and then stored in a liquid N2 container (-196oC). To test frozen semen, it was done by stored frozen semen at 37oC for 30 seconds. The results showed that the highest quality of spermatozoa (P <0.05) was found in T2 and T3 when compared to T1 (P> 0.05), with the motility value was T2: 28.13 ± 2.39%; T3: 25.00 ± 4.08%; and T1 18,15±3,23%. It can be concluded that 2 hours and 3 hours equilibration time was able to maintain the quality of frozen semen landrace boars and the statistically was not significant.

Abstract

The purpose of this study was to test the influence of the equilibration time on the frozen semen quality of duroc pig in modified tris-egg yolk with the addition of extracellular cryoprotectants. Semen is collected twice per week using massage methods from two duroc males aged ±2.5 years which in good health and have been trained in semen colection. The design used is a completely randomized design with 4 tests and 3 treatments T1: equilibration time of 1 hour, T2: equilibration time of 2 hours, T3: equilibration time of 3 hours at a temperature of 50C. Observed parameters were motility, viability, abnormality, MPU, and recovery rate (RR). The results showed that motility, viability, abnormalities, MPU, and RR were higher (P<0.05) at 2 hours of equilibration (25.63±3.15%, 55.86±5.41%, 4.92±0.53%, 56.69±5.64%, 36.61±4.49%) compared to 1 and 3 hours (20.00±3.54%, 39.28±9.89%, 7.93±0.76%, 40.21±9.39%, 28.57±5.05%) and (15.00±4.08%, 32.19±7.0%, 8.41±0.70%, 32.71±7.07%, 21.43±5.83%). It was concluded the motility of duroc pig spermatozoa in the tris-modified diluents after 2 hours of equilibration reached 64.375% and 25.63% post thawing, but did not reach the requirement of 40% spermatozoa motility for artificial insemination. Abstract

This study determined whether the administration of progesterone and hCG after insemination increased pregnancy rate in Bali Timor cows. A total of 30 Bali Timor cows (BW ± 200-225 kg; aged 3-7 years old) with different parity were used in this study. Cows were inseminated with progesterone and hCG. Cows were grouped based on body weight and number of parities following a Complete Randomized Design with 3 treatments and 10 replications. The treatments were: P1 = did not receive any treatment (control); P2 = estrus induced by PGF2α 125 mg on day 5, 7 and 9 (n = 10); and P3 = estrus induced by hCG 1000 IU on day 5 after artificial insemination (AI). Blood samples were collected from the jugular vein of the animals on day 0 and continued for a period of 33 days after AI using 5 mL heparinised pre-set syringes Blood samples were then centrifuged at 1500 rpm for 10 minutes. Serum was harvested and stored at -20oC until serum concentrations of progesterone and hCG were determined using ELISA procedures. Pregnancy was determined by rectal palpation. The results showed that the AI pregnancy rates was greater in group treated with hCG (70%) than cows induced with progesterone (50%) or the control group. The concentrations of both progesterone and hCG were increased simultaneously on day 5 to 11 post AI. In conclusion, progesterone and hCG application in Bali Timor cows after artificial insemination could increase pregnancy rates.
