Abstract

ABSTRACT. Black garlic is an aged product derived from fresh garlic and exhibits potential antibacterial properties. This study aims to identify the antibacterial compounds in black garlic against Salmonella typhimurium using HPLC fingerprinting and chemometrics. The antibacterial activity of the n-hexane fraction, methanol fraction, and water fraction were analyzed using the well diffusion method. Only the water fraction exhibited an inhibitory zone against S. typhimurium of 11.96 ± 1.86 mm. The chemical profile of each fraction, analyzed by HPLC-MWD, was correlated with antibacterial activity data using Orthogonal Projection to Latent Structure. A retention interval of 2.50 – 2.99 and 2.00 – 2.49 minutes of the water fraction at a wavelength of 214 nm demonstrated a correlation with antibacterial activity. The peaks identified in HPLC-MWD at a retention interval of 2.50 – 2.99 and 2.00 – 2.49 minutes exhibited a similar pattern to the peaks appearing at a retention time of 1.12 and 1.06 minutes on the UHPLC-Q-Orbitrap-HRMS chromatogram. The resultant peak in the UHPLC-Q-Orbitrap-HRMS analysis of the water fraction indicated that it was attributed to S-allyl cysteine and S-allyl mercaptocysteine. Abstract

The consumption of fresh vegetables at the stalls needs serious attention. This research aimed to estimate the exposure probability due to fresh vegetables consumption of street food consumers, to measure sanitation level of street stalls that serving fresh vegetables, and to recommend a mentoring program for the stalls at four locations in Bogor City. This research was conducted at 16 stalls located at four locations in Bogor City. The number of respondents surveyed was 293 people and determined by stratified sampling method. Food frequency questionnaire was used as a tool in the survey. The survey showed that men consumed more fresh vegetables than women with the average of 47.12 g/person/consumption and frequency 3.37 times/week (p<0.05). Adults consumed fresh vegetables at the stalls most often with 3.05 times/week and the average consumption of 44.59 g/person/consumption (p<0.05). All street stalls were at level IV of sanitary practices. Thus, there is a risk of food safety in consuming fresh vegetables at the stalls at four locations in Bogor City. Men and adults had high exposure probability to microbiological hazards due to fresh vegetables consumption at the stalls. Mentoring program for all locations involving socialization and implementation of street stall management and sanitation practices regulations by government, full awareness of street vendors in complying with the prevailing regulations, and firm action for the vendors who violate the rules.

Abstract

Shrimp belongs to type of food that oftenly contaminated with Listeria spp. Bacteriophage as an antimicrobial agent represents an attractive way to destroy bacterial contamination on food. This study presents the proof of lytic phage of Listeria (FL03) usage and influenced storage temperature to infect Listeria grayi on food matrix of shrimp. Shrimp samples were inoculated with 1.3 X 107 CFU mL-1 L. grayi (BA03) and then treated with 1.5 X 108 PFU mL-1 lytic phage of Listeria (FL03). The total number of L. grayi was counted at storage times of 0, 1, 24 and 48 hours at room temperature (±28 oC) and cold temperature (±4 oC). The results showed that shrimp treated with phage FL03 showed a decrease in the number of L. grayi compared to the control, namely 0 - 0.15 log10 CFU g-1 at room temperature (±28 oC) and 0.93 - 1.37 log10 CFU g -1 at cold temperatures (±4 oC). Treatment at cold temperatures (±4 oC) can inhibit the growth of L. grayi by 2.01 – 2.98 log10 CFU g-1. The growth inhibition of L. grayi increased further when treatment at cold temperatures (± 4 oC) was combined with FL03 phage treatment, namely to 3.38 – 3.76 log10 CFU g-1. These findings illustrated that phage FL03 could infect and cause lysis of L.grayi on food matrix of shrimp samples.
