Abstract

This study was conducted to determine the condition of immunological response of and the presence of bacterial diseases in Anabas Testudineus fish was transported from pond culture towards Segiri Market. Transport system carried out by the fishmonger supposed to influence the health conditions of fish as well as the presence of bacteria on the fish's body. The sample was 30 fish from Muara Kaman pond and Markets Segiri as a fish marketing. Some parameters were observed among others: changes in swimming behavior, pathology anatomy organs, aemotology and inventory of bacterial diseases. The results showed that the samples of fish from the pond aquaculture generally have normal conditions of organ anatomy, symptom behavior swim also normally, otherwise fish from marketed in the Market Segiri, as much as 60% of fish have abnormal behavior of swimming, 73% have pathology on external anatomy and 53% fish have abnormalities in organs. Results of the isolation and identification of bacterial disease found that there are 5 types of bacteria Aeromonas, Pseudomonas, Staphyllococcus, Streptococcus and Basillus, and only bacteria Staphyllococcus and Basillus not found in fish pond aquaculture origin. The health condition of the fish pond aquaculture origin betook better, judging from the hemoglobin, hematocrit, total leukocyte and total erythrocyte that is within the normal range. While the blood picture fish from Segiri market is generally higher in all parameters that indicate health problems. It can be concluded that the transport system carried out by the wet etching fishmonger effect on the occurrence of abnormalities in behavioral symptoms, anatomic pathology outer and inner organs, the presence of bacterial disease and the decline in fish health conditions.Keywords : Betok fish, bacterial, transportation

Abstract

Aeromonas hydrophila is a causative agent of Haemoragic Septicemia on Clarias gariepinus. The aim of this research was to investigate the effect of Acanthus ilicifolius leaf extract toward survival rate of C. gariepinus infected by A. hydrophila. Tested fish C. gariepinus 10-12 cm and weight ±25 gr. This reasearch was conducted by 4(four) treatments namely, A (treatment with no leaf ectract A. ilicifolius), B (300 ppm), C (500 ppm), and D (700) 30 minutes immersion. The challenge test was done by injecting 0,1 mL A. hydrophila suspensions with dosage 108 cell/mm3 intra-muscular by experiment fishes. Observation was performed for 5 days after infection such as blood profile, clinical symptoms, and survival rate of C. gariepinus. The results of blood profile after the infection showed that hematokrit of A, B, C, and D treatments was 17,33%, 14,67%, 15,00%, 19,67% respectively. Percentage of the highest erythrocyte was at D treatment 1,79x106 sel/mm3, B 1,72 x106 sel/mm3, A 1,64x106 sel/mm3, C 1,52x106 sel/mm3. Percentage of the highest leuchocyte was at C treatment 5,43x104 sel/mm3, D 3,11x104 sel/mm3, B 2,63x104 sel/mm3, and A 2,32x104 sel/mm3. The result showed that clinical symptoms of A. hydrophila infected fish swam abnormally, injured and haemoragic on the skin along with damaged on the body. Immersion with A. ilicifolius extract leaf past infection indicated that they were not significantly different on C. gariepinus survival rate. Therefore the dosage of A. ilicifolius leaf extract did not sufficient to protect C. gariepinus from A. hydrophila infection.

Abstract

This research aimed to study the administrasion of B. pandurata extract mixed with crab feed on the percentage of molting, growth, and survival, and feed conversion ratio of mud crabs reared in silvofishery ponds. This research was conducted in Salo Palai Village, Muara Badak District, Kutai Kartanegara Regency. This study used a completely randomized design, with 4 treatments and 3 replications. The extract of B. pandurata in the form 3 In BIOIMUN® drug there are 4 treatment dose dasnamely (0 mL/kg, 20 mL/kg, 25 mL/kg, 30 mL/kg feed). The crabs used came from the catch of the Muara Badak community with a size of 200-280 g, and were culture for 4 weeks (30 days). The crabs were fed fresh fish with the addition of extract 2 times a day, in the morning (07.00-08.00) and afternoon (16.00-17.00), but feeding was done every 2 days. The feed given was weighed as much as 5% of the weight of the crab. The results showed that the administration of 3 In 1 BIOIMUN® in treatment P1 (20 mL/kg) affect the percentage of molting, growth, survival, and feed conversion ratio.

Abstract

Tilapia is one type of freshwater fish that has high economic value. With the increasing demand by the community, tilapia is cultivated in a semi-intensive system so that bacterial diseases caused by Aeromonas hydrophila arise. One way to treat disease in fish safely is the use of lactic acid bacteria. The purpose of this study was to examine the isolates of lactic acid bacteria in controlling diseases caused by Aeromonas hydrophila in tilapia. This study used a completely randomized design experimental method with 4 treatments and 3 replications. The treatments consisted of feed mixed with Phosphate Buffered Saline (PBS), Enterococcus faecalis isolate, Lactobacillus plantarum isolate and Lactococcus lactis isolate at a dose of 0.1 mL/g of feed at a concentration of 106 CFU/mL and mixed with 0.1 mL of egg white. The results showed that the treatment with lactic acid bacteria affected the condition of clinical symptoms and anatomical pathology of tilapia, but was better than the control treatment. The feed mixed with lactic acid bacteria also had an effect on increasing the immune response and showed higher survival than the control treatment. The condition of water quality in the rearing media showed the normal range for the life of tilapia.

Abstract

Shrimp is a leading commodity traded in local and international markets. The process of deterioration of shrimp quality can be caused by an autolysis reaction. This problem is the rationale for looking for an alternative natural preservative that is harmless and easy to obtain, namely using pedada (S. alba) leaves. This study aimed to determine the effect of pedada leaf extract (S. alba) on the post-harvest quality of fire shrimp and to determine the best concentration in inhibiting bacterial growth during cold storage (5oC). The test parameters observed were TPC, pH, and organoleptic tests. This study used a completely randomized design (CRD) with 5 treatments and 3 replications. The treatments consisted of P1 (seawater extract + 800 ppm S. alba); P2 (seawater extract + S. alba 1200 ppm); P3 (freshwater extract + S. alba 800 ppm); P4 (freshwater extract + S. alba 1200 ppm); and P5 (control/without extract). The results showed that the immersion of fire shrimp in pedada leaf extract did not affect the TPC and pH values during cold storage, but did affect the organoleptic test for the appearance, smell, and texture specifications of the shrimp. Pedada leaf extract with 1200 ppm seawater as a solvent during cold storage (5°C) obtained the best results with a TPC value of 2.88 log cfu/g, a pH value of 7.12, and an organoleptic value (appearance 7.3, odor 7.3 and texture 7,3).

Abstract

Present research aimed to examine the pathogenicity opportunities of some bacteria which were isolated from freshwater aquaculture ponds to some fish Oreochromis niloticus, Clarias gariepinus, and Pangasius pangasius. The bacteria were isolated from the water and feces of O. niloticus and C. gariepinus aquaculture in Samarinda, which were identified as Escherichia coli, Enterobacter cloacae, and Enterobacter amnigenus. An 0.1 mL-1 of each bacteria was intramuscular injection and evaluated the clinical signs, eksternal pathology, and fish mortality. The freshwater fish using in this research were Oreochromis niloticus, Clarias gariepinus, and Pangasius pangasius with size 10-15 g from Loa Kulu, Kutai Kartanegara Regency, East Kalimantan, Indonesia. The result showed that each bacteria caused mortality in fish: E. coli bacteria caused fish mortality 23.33-66.67%; E. cloacae 10.00 to 90.00%; and E. amnigenus by 3.33-56.67%. The average death time of E. coli and E. amnigenus in all three fish were under 48 hours, while E. cloacae caused mortality less than 48 hours in C. gariepinus and P. pangasius. In conclusion, E. coli, E. cloacae, and E. amnigenus are putative pathogenic bacterium in O. niloticus, C. gariepinus, and P. pangasius.

Abstract

Penggunaan berbagai jenis mangrove efektif sebagai bahan alami untuk menghambat pertumbuhan bakteri pada ikan. Buah Xylocarpus granatum mengandung metabolit sekunder yang berpotensi sebagai antibakteri alami. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dan mengetahui nilai Minimum inhibitory concentration (MIC) ekstrak metanol biji buah X. granatum. Tahapan penelitian yaitu : preparasi dan ekstraksi biji buah X. granatum, isolasi bakteri insang ikan nila, pengujian daya hambat, pengujian MIC dan pengujian Total Plate Count (TPC). Preparasi dilakukan dengan cara mengiris tipis biji buah X. granatum dan dikeringanginkan selama 30 hari. Biji buah yang sudah kering (simplisia) dikecilkan ukurannya menggunakan blender selanjutnya diesktraksi dengan cara maserasi menggunakan pelarut methanol 98% selama 24 jam, selanjunya disaring. Filtrat dipisahkan dari pelarut methanol menggunakan rotary evaporator pada suhu 600C hingga didapat ekstrak biji buah X. Granatum. Kandungan metabolit sekunder ekstrak methanol biji buah X. granatum ditemukan seyawa alkaloid, triterpenoid, flavonoid, saponin, fenol, dan tanin. Perlakuan dalam penelitian ini adalah ekstrak metanol biji buah X. granatum dengan konsentrasi 100 ppm, 500 ppm, dan 1000 ppm, kontrol positif (Chloramphenicol) 1000 ppm dan akuades steril sebagai kontrol negatif. Parameter yang diamati yaitu daya hambat, MIC, dan TPC. Hasil penelitian menunjukan bahwa ekstrak metanol biji buah X. granatum memiliki kemampuan dalam menghambat pertumbuhan isolat bakteri dari insang ikan nila dengan kategori sedang (9-9.8 mm) pada konsentrasi 100 ppm dan 500 ppm, konsentrasi 1000 ppm dengan kategori kuat (11,5 mm) pada pengamatan jam ke 48. MIC ekstrak metanol biji buah X. granatum terhadap isolat bakteri insang ikan nila adalah pada konsentrasi 40 ppm. Hasil pengujian TPC pada konsentrasi 40 ppm adalah 7 CFU/ml atau terjadi penghambatan pertumbuhan bakteri sebesar 91,67%.

Abstract

Penelitian ini menggunakan pendekatan deskriptif kualitatif dengan tujuan menganalisis pelaksanaan dan dampak program tersebut terhadap lingkungan, ekonomi, dan sosial. Hasil penelitian menunjukkan keberhasilan program mencapai 100% dari kriteria yang ditetapkan dalam rencana pascatambang. Populasi sapi meningkat dengan kesehatan yang baik, menggunakan pakan seperti rerumputan dan batang pisang. Pola pemeliharaan sapi mayoritas dilakukan secara ekstensif. Dampak sosial positif terlihat dari hubungan baik antar peternak, sementara dampak ekonomi terwujud melalui peningkatan pendapatan dari penjualan sapi. Dampak lingkungan diminimalkan dengan memanfaatkan kotoran sapi sebagai pupuk. Partisipasi peternak aktif, didukung komunikasi baik antar peternak, PPL, dan perusahaan, dengan minat tinggi dalam menerapkan teknologi ternak seperti inseminasi buatan.

Abstract

The initiative to aid in the formulation of herbal medication for fish, targeting disease management and enhancing freshwater fish farming output alongside "KOPER BASAH," is implemented to instruct fish farmers in the creation of herbal remedies from indigenous flora for disease control in tilapia aquaculture. This activity involves detecting issues such as elevated death rates owing to pathogen assaults; facilitating the preparation of herbal medicine; assessing the efficacy of the produced herbal medicine; and monitoring and analyzing its impact on tilapia output. Knowledge pertaining to the signs of fish afflicted by infections and the management of diseased fish has augmented by 25-50%. Knowledge of plant kinds with antibacterial and immunostimulant properties increased by 30-50%, while abilities in herbal medicine preparation improved by up to 60%. This activity enhances the value of locally accessible raw materials and optimizes tilapia farming production in a secure, cost-effective, and sustainable manner. This research is anticipated to be a prudent advancement for the community in tilapia aquaculture by adhering to Good Aquaculture Practices and guaranteeing food safety of the resultant fisheries items.

Abstract

This study aims to determine the pathogenicity of lactic acid bacteria from repang fish on clinical symptoms, anatomical pathology, hematology and survival rate of tilapia. This study used a completely randomized design with 5 treatments and 3 replications, namely Phosphate Bufferet Saline (PBS) 0.1 ml/fish as a negative control, A. hydrophila 106 CFU/ml as a negative control, Enterococcus sp. 106 CFU/ml, Lactobacillus sp. 106 CFU/ml and Lactococcus sp. 106 CFU/ml, fish given intramuscularly. Parameters observed included clinical symptoms, anatomical pathology. Hematology and survival. Tilapia with an average weight of 30 g/fish were reared for 7 days in an aquarium measuring 50×40×30 cm3 with a volume of 40 L of water, at a density of 10 fish/aquarium. Fish are fed 3 times a day. Administration of lactic acid bacteria Lactobacillus sp. showed the highest survival compared to the treatment with other isolates. Hematological parameters were in the normal range in all treatments except for the treatment with A. hydrophila bacteria. Changes in clinical symptoms of tilapia treated with injection with A. hydrophila showed clinical symptoms of swimming patterns, decreased appetite and also moved more hyperactively. In fish treated with Enterococcus sp., Lactococcus sp., and Lactobacillus sp. generally shows normal condition. Anatomical changes in fish treated with A. hydrophila bacteria showed changes in the form of excess mucus, redness on the back, pale body and easily detached scales. The three lactic acid bacteria from the intestines of repang fish are safe for tilapia and can be used in further probiotic tests.
