Abstract

Vanda tricolor Lindl. var. suavis is an Indonesian wild orchid that has been rare in nature,  so it needs attention to take care and conserve them.  The objective of the research was to investigate the effect of coconut water and / or tomato juice on the growth of protocorm of V. tricolor Lindl. var suavis from Bali grown in vitro. The experiment was laid out in the factorial design, with two factors (coconut water/CW and tomato juice/TJ), each contained three concentration (CW: 0, 100, and 200 cc L-1; TJ: 0, 100 and 200 g L-1) resulted in nine combination of treatments and replicated four times.  The results showed that tomato juice with concentration of 100 gL-1 or 200 gL-1 promotes growth of protocorms of Vanda tricolor Lindl. var. suavis from Bali regardless the presence of  coconut water.

Abstract

Penelitian ini bertujuan untuk mengetahui kualitas susu kambing Peranakan Etawa selama penyimpanan suhu ruang.Sampel susu diambil dari 5 ekor kambing Peranakan Etawa masing-masing sebanyak 500 ml. Penelitian ini menggunakan rancangan acak kelompok (RAK), dengan pola 5x5 yaitu 5 ekor kambing Peranakan Etawa sebagai kelompok dan diamati 2, 5, 8, 11 dan 14 jam sebagai perlakuan, sehingga sampel yang diperiksa sebanyak 25 sampel. Hasil Penelitian menunjukkan bahwa kualitas susu kambing Peranakan Etawa menurut uji rasa, pH dan uji alkohol setelah 5 jam penyimpanan suhu ruang mengalami penyimpangan. Kemudian dilanjutkan dengan uji statistik yang memperoleh hasil untuk uji rasa setelah 5 jam terjadi penyimpangan yang sangat nyata (P < 0,01). Pada pengukuran nilai pH menunjukkan lama penyimpanan suhu ruang berpengaruh nyata (P < 0,05) terhadap kualitas susu kambing dilanjutkan dengan uji Duncan yang memberikan hasil bahwa selama 2 sampai 5 jam tidak terjadi penurunan pH yang nyata (P > 0,05), sedangkan pada jam ke- 8, 11 dan 14 terjadi penurunan pH yang sangat nyata (P < 0,01). Uji alkohol menunjukkan bahwa pada jam ke- 2 susu dalam keadaan normal dan belum menunjukkan adanya butiran pada dinding tabung reaksi, sedangkan mulai jam ke - 5 sampai jam ke - 14 terjadi penyimpangan sangat nyata (P<0,01), sehingga dapat disimpulkan dari parameter rasa, pH dan uji Alkohol susu kambing hanya bertahan sampai 5 jam dalam penyimpanan suhu ruang.

Abstract

Telah dilakukan penelitian untuk mengetahui beban cemaran bakteri Escherichia coli pada daging se’i babi yang dipasarkan di kota Kupang Provinsi Nusa Tenggara Timur (NTT) yang diambil dari enam tempat pembuatan daging se’i babi secara tradisional yang tersebar di kelurahan Oebufu, Oebobo dan Baun. Se’i merupakan daging asap khas kota Kupang yang diasapi menggunakan kayu Kosambi (Schleichera oleosa, Merr). Hasil penelitian untuk setiap lokasi diperoleh sebagai berikut (1) Bambu Kuning–Oebobo sebesar 210 MPN/gr (2) Green Garden–Oebufu sebesar 150 MPN/gr (3) Baun sebesar 210 MPN/gr (4) Petra-Oebufu sebesar 7,2 MPN/gr (5) Pondok Sawah-Oebufu sebesar 3,6 MPN/gr dan (6) Aroma-Oebobo sebesar 14 MPN/gr. Jumlah kandungan bakteri Escherichia coli dari keenam sampel, sudah melebihi batas maksimum cemaran bakteri Escherichia coli pada daging asap.

Abstract

African swine fever (ASF) is a highly contagious and lethal disease of pigs for which no effective vaccine is currently available. This study evaluated the pathogenicity of field African swine fever virus (ASFV) isolates from Bali and East Nusa Tenggara and assessed their potential for inactivated vaccine development using spleen-derived antigens. Based on the qPCR results, among all ASFV isolates tested, only the Bali isolate, designated B1, exhibited the highest DNA concentration, suggesting that this sample contained the highest ASFV titer. Consequently, the B1 isolate was selected and processed into chemically treated (CT-ASFV) and non-treated (NCT-ASFV) formulations. In the pathogenesis trials, pigs inoculated with NCT-ASFV developed acute ASF and died within 12 days, whereas those receiving CT-ASFV exhibited no clinical signs of ASF and showed no evidence of viral replication. In the vaccination trial, pigs immunized with CT-ASFV emulsified in Montanide™ ISA 50 V2 demonstrated survival in two out of three animals (67%) following challenge with a virulent strain, while all control animals succumbed to infection. Despite the absence of detectable humoral responses as determined by ELISA, the observed protection suggests a potential role for cell-mediated immunity. These findings indicate that the B1 isolate was highly virulent and represents a promising candidate for the development of an inactivated ASF vaccine. Further evaluation in larger-scale field trials is warranted.
