Abstract

Result of research Dian Indratmi previously (2000, 2001, and 2002) indicated among antagonisfungi that shown potential inhibition Colletotrichum gloeosporioides better were fructoplant yeastinoculan especially Debaryomyces sp. For evaluation of efficacy yeast Debaryomyces sp.as apotential biokontrol agent of chilli anthracnose or to develop this agent as biopesticide, it is importantto have available culture media that maximize production at low cost.The aimed of this research to find out a low cost media culture formulation for yeast fructoplantbiomass production for their application on field to suppressed anthracnose disease on pepper.The laboratory experiment with Complete Randomized Design and 6 replication. There were11 treatment, it?s A = malt extract liquid media; B = malt extract liquid media + pepper extract; C= corn meal liquid media; D = corn meal liquid media + pepper extract; E = sugar potato extractliquid media; F = Alioshina liquid media; G = sugar mixture of rice and bran liquid media; H =sugar pepper extract liquid media; I = maize solid media; J = waste water rice media; K = ipomoealiquid media.Results of the experiment indicated 11 of the yeast media culture test can as a substrate foryeast biomass production. Yeast Debaryomyces sp that cultured on the substrate corn meal liquidmedia that added with or without pepper extract resulted better weight and quantity cell of yeastDebaryomyces sp. The yeast for their growing needed a rather acid substrate, it?s about pH 4.79-6.07.

Abstract

The aimed of this research to find out the effectifity some adjuvant to enhance Debaryomycessp. and Schizosaccharomyces sp. actifity as agens biocontrols against anthracnose pathogen C.gloeosporioides on mango.The laboratory experiment included testing adjuvant conferral for enhance anthracnose diseaseobstruction on mango. The experiment with Complete Randomized Design and 3 replication. Therewere 13 treatment,it’s A1 = control (only pathogen); A2 = yeast Debaryomyces sp. ; A3 = yeastSchizosaccharomyces sp.; A4 = yeast Debaryomyces sp.+ Schizosaccharomyces sp.(mixture); A5 =Debaryomyces sp. + Tween 1 %; A6 = Schizosaccharomyces sp. + Tween 1 %; A7 = mixture yeast+ Tween 1 %; A8 = Debaryomyces sp. + CaCl2 2%; A9 = Schizosaccharomyces sp. + CaCl2 2%;A10 = mixture yeast + CaCl2 2%; A11 = Debaryomyces sp. + Trehalose 2%; A12 =Schizosaccharomyces sp. + Trehalose 2%; A13 = mixture yeast + Trehalose 2%.Results of the experiment indicated Debaryomyces sp. and Schizosaccharomyces sp. yeast insingle or mixtures with or without adjuvant to be able to grow on mango fruit surface, and effectiveinhibited development of the anthracnose disease caused of C. gloeosporioide on mango fruit thatshown with significantly reduced lesion size and incubation periods 4 days longer.

There were 10 treatment, it’s M1 (rain tree pod extract + potato extract); M2 (rain tree pod extract + chilli extract ); M3 (corn meal+ chilli extract); M4 (sugar potato extract); M5 (sugar chilli extract); M6 (potato dextrose broth); M7 (corn solid media); M8 (100% chilli extract); M9 (sugar corn meal); dan M10 (100% rain tree pod extract) Results of the experiment indicated from 10 of the yeast media culture test 100%Chilli extract medium and rain tree pod extract + potato extract medium resulted better spore weight and cell density of Rhodotorula sp. yeast. Its shown with spore weight 0.4833 g/ 10 ml to 100% chilli extract and cell density 59.67 x 103 cell/ ml. At rain tree pod extract + potato extract medium, spore weight 0.4845 g / 10 ml with cell density 70.67x103 cell/ml. Production Rhodotorula sp. yeasy cell on solid medium can used corn medium.Key word : Colletrotichum gloeosporioides, Rhodotorula sp. , Biocontrol
