Abstract

Objective: to know the result of Hematoxyllin-eosin and immunostainning D2-40 Podoplanin examination in detecting lymph vessel invasion (LVI) and to analyze correlation between lymph vessel invasion with pelvic lymph node metastasis in stage IB and IIA cervical cancer. Design of this research was cross-sectional study. The clinicopathological data of 41 patients with stage IB and IIA cervical cancer who underwent radical hysterectomy and pelvic lymphadenectomy (RHPL) at Dr. Soetomo Hospital, Surabaya, from January 2008 to June 2013 were retrospectively reviewed. Age, histology type, clinical stage, tumor size, number of removed pelvic lymph node, parametrial invasion and LVI were analyzed to define their correlation with pelvic node metastases. Immunohistochemical staining using MAb-D2-40 Podoplanin was used to detect lymph vessel invasion on the corresponding HE slides. Eligible criteria are paraffin wax embedded tissue from cervix cancer patients was diagnosed as cervix cancer, and patient’s medical record can be found and filled completely. Ineligible criteria are patients which is had chemotherapy or radiotherapy before surgery or patient’s paraffin wax embedded tissue was damaged. The correlation among variables was analyzed using Phi correlation test. Results: among of total 41 patients , lymph vessel invasion (LVI) was detected in 7 (17.07%) D2-40 Podoplanin slides, and 9 (21.95%) HE slides. Agreement rate with Kappa statistic between two method was significant, k = 0,381, p = 0,014. Lymph node metastasis was found in 6 (14.63%) samples. Positive LVI detected by D2-40 Podoplanin was significantly correlated with lymph node metastasis (? = 0.362; p = 0.02), but positive LVI detected by HE was not significantly correlated (? = 0.281; p = 0.072).

Abstract

Abstract

Sea cucumbers are deep-sea-dwelling invertebrate animals that are rich in nutrients and have been empirically used to prevent various degenerative diseases, especially due to their ability to dampen free radicals. The purpose of this study was to identify, and determine antioxidant activity, total phenolic content, and toxic properties in extracts of several sea cucumber species originating from Tegal Mas Island, Lampung. The identification results of the three sea cucumber species were Stichopus vastus Sluiter (SV), Stichopus monotuberculatus Quay & Gaimard (SM), and Stichopus quadrifasciatus Massin. (SQ). The extraction was performed by kinetic maceration against all parts of the sea cucumber using a 70% ethanol solvent. Antioxidant activity test using the DPPH (2,2-diphenylpicrilhydrazyl) free radical suppression method and determination of the total phenolic content using the Folin-Ciocalteu method. The results showed that SV had the highest total phenolics and antioxidants with the lowest toxicity. However, optimisation of extraction between wet and dry sea cucumbers is still required. Extraction optimisation with various solvents can also be done to gain secondary metabolites more effectively.

Abstract

BACKGROUND: Tumor markers such as human epididymis protein 4 (HE4), cancer antigen 125 (CA-125), and risk of ovarian malignancy algorithm (ROMA) are frequently utilized for diagnostic and prognostic purposes. Lymphocytes, essential immune cells, play a significant role in eliminating cancer cells. However, the precise correlation between tumor markers and lymphocytes remains incompletely elucidated. The aim of this study was to explore the correlation between tumor markers and lymphocyte subtype profiles in differentiating ovarian cancer and benign tumors.METHODS: This was a cross-sectional study involving 12 ovarian cancer and 17 benign ovarian tumor patients. Blood samples were collected for the characterization of T lymphocytes, B lymphocytes, natural killer (NK), and T regulatory (Treg), which were analyzed using flowcytometry. Additionally, tumor markers HE4 and CA-125 were measured from patient serum using the chemiluminescent microparticle immunoassay (CMIA) method.RESULTS: Benign ovarian tumors and ovarian cancer can be distinguished by a significant increase in HE4 levels (p=0.004), ROMA (p=0.004), and Treg cells (CD4+/CD25+/FOXP3+, p=0.017). Furthermore, the correlation between tumor markers and lymphocytes indicates that an increase in ROMA was weakly correlated with an increase in the percentage of T regulatory cells (CD4+/CD25+/FOXP3+, r=0.553, p=0.006) and B lymphocytes (CD19+, r=0.528, p=0.010), accompanied by a decrease in the number of T lymphocytes (CD3+, r=-0.598, p=0.003), T helper lymphocytes (CD3+CD4+, r=-0.594, p=0.003), and cytotoxic lymphocytes (CD3+CD8+, r=-0.510, p=0.013).CONCLUSION: The elevation of serum tumor markers (HE4 and ROMA) accompanied by an increase in Treg cells can distinguish between benign ovarian tumor patients and ovarian cancer patients.KEYWORDS: tumor marker, CA-125, HE4, ROMA, subtype of lymphocytes, ovarian cancer
