Abstract

Makanan yang bergizi, pengendalian berat badan, dan aktivitas fisik merupakan program terapi untuk pasien diabetes melitus. Namun, banyak pasien yang sulit untuk menjalankan gaya hidup sehat tersebut, sehingga pasien harus bergantung pada obat-obatan untuk terapi. Selain pengobatan modern, pengobatan tradisional telah digunakan sejak lama dan berperan penting sebagai pengobatan alternatif. Berbagai jenis senyawa metabolit sekunder seperti tannin dan flavonoid telah diidentifikasi sebagai penghambat enzim hidrolitik α-amilase dan α-glukosidase yang membantu mempertahankan fungsi sel serta menurunkan kadar glukosa darah. Salah satu tanaman yang memiliki potensi tersebut yaitu tanaman landep (Barleria prionitis L.). Penelitian ini mengevaluasi ekstrak daun landep (Barleria prionitis L.) sebagai kandidat antidiabetes dengan metode ekstraksi sokletasi dan maserasi. Simplisia daun daun landep diekstraksi menggunakan etanol 70%. Pengujian potensi sebagai antidiabetes dilakukan dengan metode Nelson Somogyi. Hasil uji fitokimia ekstrak daun landep metode maserasi (ELM) dan sokletasi (ELS) mengandung alkaloid, flavonoid, dan tannin. Efektivitas penurunan kadar glukosa berupa EC50 ELS sebesar 18,28±0,04 mg/L dan ELM sebesar 22,63±0,01 mg/L. Perbandingan kedua metode ekstraksi ini menunjukkan adanya perbedaan kualitatif dalam kandungan fenolik dan flavonoid serta efektivitas penurunan kadar glukosa. Secara keseluruhan, metode sokhletasi untuk ekstraksi daun landep dalam memperoleh zat sebagai kandidat antidiabetes lebih efektif dibandingkan dengan maserasi.

Abstract

This study evaluated kirinyuh leaf extract as a natural antioxidant using Soxhlet extraction and maceration extraction methods. Dried leaves of kirinyuh were extracted with 96% ethanol. The total phenolic content was measured spectrophotometrically according to the Folin Ciocalteu method, calculated as gallic acid equivalent (GAE), while the antioxidant activity was measured using ABTS. The results of the phytochemical test of kirinyuh leaf extract using maceration (EKM) and Soxhletation (EKS) methods revealed alkaloids, flavonoids, saponins, steroids, and tannins. The total phenolic content of EKM was 39.85 ± 0.20 mgGAE/gram and EKS was 46.95 ± 0.15 mgGAE/gram extract. Antioxidant activity in the form of IC50 EKM was 46.10 ± 0.09 mg/L and EKS was 45.27 ± 0.07 mg/L, both of them have a very strong category. There were significant differences in the total phenolic content and antioxidant activity of the ethanol extract of the Kirinyuh leaf using maceration and soxhletation methods based on the results of the T-test. Abstract

Secondary metabolites found in oyong leaves (Luffa acutangula (Linn.) Roxb.) include  phenolics, flavonoids, saponins, terpenes and tannins. Phenolic compounds have an important role as antioxidants. Antioxidant be able to protect the body from degenerative diseases caused by cell damage due to free radicals. Extract that undergo a purification process will give the different levels of total phenolic to the crude extract. In this research, the total phenolic content of ethanol extract and purified extract of oyong leaves will be determined. Maceration with 96% ethanol was used as an extraction technique. The liquid extraction technique was employed for extract purification, with warm water, n-hexane and ethyl acetate serving the solvent. The phenolic content was detected by Thin Layer Chromatography (TLC), the stationary phase of silica gel , the geraneous phase of n-hexane : ethyl acetate : methanol (2:7:2). Phenolic content was measured using UV-Vis Spectrophotometry, Folin-Ciocalteau reagent and standard gallic acid. The total phenolic levels obtained in the ethanol extract and purified extract of oyong leaves were (34,000 ± 0,350) mg GAE/gram extract and (40,117 ± 0,195) mg GAE/gram extract. Purified extract have higher levels because pass through a purification stage which produce pure extract with higher compound content.

Abstract

The Covid-19 pandemic, according to PHEOC data from the Ministry of Health as of July 31, 2020, in Indonesia from 866,539 specimens examined, 108,376 confirmed cases, 5,131 cases (4.7%) died and 65,907 cases (60.8%) recovered and 37,316 cases in care. One of the government's programs to prevent the spread of Covid-19 is to use masks properly. The purpose of this study was to determine the level of compliance and accuracy of the use of masks for traders in traditional markets in Surakarta as an effort to prevent Covid-19 disease. This research was conducted using a descriptive survey of 5 markets in the city of Surakarta including the Gede market, Nusukan, Jongke, Kadiplo, and Sidodadi markets. The research sample was taken by random sampling as many as 200 traders in the traditional market of Surakarta. The results of this study indicate that the compliance of traders who interact and conduct transactions in traditional markets in Surakarta City is quite good, because 85% of traders obey using masks as a health protocol to prevent the transmission of the Covid-19 virus. However, the level of accuracy in using masks is still poor, because of the 200 respondents, only 25.24% traders use masks correctly. Suggestions, it is better for the Surakarta City Government, especially the Trade Office, to make regulations and increase socialization and supervision of traders and buyers who interact actively in traditional markets so that new clusters of Covid-19 spread in traditional markets are not formed.
