Abstract

The use of natural materials for medical purposes is still being conducted by the public. One of the natural substances that are believed to play a role in health is Tamarindus indica. In Indonesia, the material is referred to as "asam jawa". The usefulness of Tamarindus indica in health have been widely trusted by the world community that led to the development of various researches, ranging from in order to know the content and active ingredients of Tamarindus indica  that may have potential until the mechanism of its role in a variety of conditions in the body. The studies even developed on Tamarindus indicas possible role in the inhibition of cancer. The purpose of this paper is to add our knowledge regarding the use and role of Tamarindus indica in a variety of therapeutic efforts.

Abstract

Obesity is a health problem whose prevalence tends to increase in Indonesia. Obesity becomes a concern, because it is one of the parameters that often followed by impaired of glucose tolerance in the metabolic syndrome. In Indonesia, Tamarindus indica is believed as a body slimming substance so it has possibility to become an alternative therapy that is easily available and cheap for obesity. The purpose of this study was to determine the ability of Tamarindus indica in inhibiting obesity. This experiment used 28 male Wistar strains in 2-3 months age that were divided into groups: group 1 (negative control, normal diet without Tamarindus indica), group 2 (positive control, obese diet without Tamarindus indica), group 3 (obese diet + Tamarindus indica 2700mg/kg BW), and group 4 (obese diet + Tamarindus indica 3600mg/kg BW). Measurement were performed in the first day after acclimization before treatment (day 8), and repeated weekly afterwards, ie at day 15, 22, 29, 36, and 43. Anova and Tukey Simultaneous statistics test were performed to see the differences in wight gain per week compared with the first week of study, then compared between groups.  Furthermore, a virtual screening that called insilico bioinformatics analyzes was performed to test the binding activity between quercetin and rutin (compounds in Tamarindus indica) with fatty acid synthase, an enzym that plays a role in de novo synthesis of fatty acids. Tamarindus indica was shown to significantly inhibit weight gain in rats with obese diet food (P=0.000), but differences dose of Tamarindus indica did not appear to have significant effect to inhibit obesity (P=0.321). The ability of Tamarindus indica is due to the compounds quercetin and rutin, where these two ingredients estimated to have the ability to inhibit fatty acid synthase with high strength.

Abstract

Fatty acid synthase is known to participate in the occurrence of malignancies, so fatty acid synthase inhibition is expected can restrain malignancy. In this research, virtual screening is done by molecular docking between the active ingredients in Tamarindus indica with three domains of FAS. In thioesterase domain, it turns out all of the active ingredients in Tamarindus indica can bind thioesterase domain right in the place where orlistat (as a reference inhibitor) bonded, with a higher strength than the orlistat. In enoyl [acyl-carrier-protein] reductase domain, it turns out the binding affinity of quercetin, rutin, catechin and epicatechin against fatty acid synthase are greater than the reference inhibitor, triclosan. In malonyl-CoA / acetyl-CoA-ACP-transacylase domain, it turns out the binding affinity of quercetin, rutine, proanthocyanidin, and catechin against fatty acid synthase is greater than the natural substrate, Malonyl-CoA. The high binding affinity of the active ingredients in Tamarindus indica against the two domains of fatty acid synthase that may also can be occupied by reference inhibitors, showed the ability of Tamarindus indica as an inhibitor of fatty acid synthase. While the high ability of active ingredients in Tamarindus indica to bind to a domain that should be occupied by the natural substrate of fatty acid synthase (malonyl-CoA) demonstrated the ability of Tamarindus indica to inhibit fatty acid synthase's work in a way to compete with the natural substrate. This study shows that Tamarindus indica may serve as anti-malignancy through its ability to inhibit fatty acid synthase.

Abstract

Objective: Tamarindus indica is one of the famous natural plant that has various effects on health. Recent research has begun to examine the effect of Tamarindus indica on cell malignancy. One of the things that happens in cell malignancy is an increasing ability of cell migration, which is characterized by increasing expression of Snail2 and MMP-9. This study was conducted to determine the effect of extract of Tamarindus indica Fruit Extract (TIFE) towards Snail2 and MMP-9 expression in JEG-3 cells.Methods :.TIFE were extracted by using methanol. JEG-3 cells were grown in culture medium treated with TIFE (at 0, 25, 50 and 100 µg/ml). Immunofluorescence staining was carried out, and observed under Confocal Laser Scanning Microscope.Result: Snail2 expression after treatment with TIFE 50 and 100 µg/ml higher than control- (P=0.015 ; P=0.001) whereas at MMP-9 expression there was a biphasic pattern, in which the MMP-9 expression peaked at concentration of 50 µg/ml (P=0.002), then MMP-9 expression decreased at 100 µg/ml concentration. Conclusion : The expression of Snail2 influence MMP-2 expression after treat with TIFE, so it might increase the potential for JEG-3 cell migration Keywords : Methanolic extract, Tamarindus indica, Snail2, MMP-9

Abstract

Malignant disease or cancer progression burden the community after decades. This tumor formation or tumorigenesis involves cell proliferation. Protein Kinases (MAPK9s) is key proteins of regulating the growth and viability of cells physiologically and pathologically. Rhizomes of Curcuma zedoaria or zedoary or white turmeric is used as a health supplement. The aim of this study was to obtained candidate proteins for anti-proliferative using the docking method between the protein MAPK9 and the active compound obtained from the crude extract of white turmeric i.e. demethoxycurcumin, curcumenol and germacrone. The result s hows that complex MAPK-demethoxycurcumin have the lowest binding affinity -8.4 Kkal/mol, while MAPK-curcumenol was -8 Kkal/mol, and MAPK-Germacrone was -6.2 Kkal/mol, it determined the potential activity cell proliferation.

Abstract

Introduction: Cervical cancer is a type of malignant tumor characterized by the appearance of abnormal cells in the cervix. According to WHO data, Indonesia has the highest number of cervical cancer patients in the world. One of the efforts to reduce the high incidence of cervical cancer is early prevention. The purpose of this study was to determine the relationship between the level of knowledge of women of childbearing age about cervical cancer and the act of pap smear examination. Method: The research method used is a literature study and analyzed through the results of research that has been done before and has been published in the form of scientific journals and other official sources. Result: The literature review show that (100%) of the journals reviewed showed a sig value < 0,05. Conclusion: The result of the literature study research on journals that examine the relationship between the level of knowledge of women of childbearing age about cervical cancer with pap smear examination actions can be concluded that all journals mention there is a significant relationship between the level of knowledge of women of childbearing age about cervical cancer with pap smear examination actions.

Abstract

Osteoarthritis merupakan suatu kondisi dimana terjadi gangguan pada persendian terutama sendi penumpu, yang merupakan penyakit degeneratif yang banyak diderita pada orang dengan usia tua sehingga menjadi penyebab utama terjadinya kecacatan pada populasi lanjut usia di seluruh dunia. Menurut Riskesdas tahun 2018, prevalensi terjadinya penyakit sendi (termasuk osteoarthritis) berdasarkan diagnosis dokter pada penduduk dengan usia ?15 tahun yang ada di kota Surabaya mencapai 7,67%, dengan prevalensi tertinggi pada kelompok umur 55 tahun ke atas (Kemenkes RI, 2018). Penyakit ini dikarenakan beberapa faktor, antara lain gangguan metabolik, proses inflamasi, dan penuaan sendi. Salah satu cara pencegahan terjadinya osteoarthritis adalah dengan melakukan senam. Pengabdian masyarakat yang kami laksanakan untuk masyarakat di lingkungan sekitar Universitas Wijaya Kusuma Surabaya dan Puskesmas Dukuh Kupang adalah dengan memberikan pelatihan Senam Pencegahan Osteoarthritis. Pelatihan dilakukan dengan diadakannya senam bersama peserta pelatihan dipimpin oleh 3 instruktur, disertai dengan media power point. Setelah acara senam, dibagikan brosur berisi gerakan senam agar peserta dapat melaksanakan sendiri di rumah. Dengan pelatihan ini peserta pelatihan dapat memahami dan mempraktekkan gerakan senam yang dapat bermanfaat menguatkan otot dan mencegah terjadinya osteoarthritis. Pelatihan ini direncanakan dilakukan secara berkala dan diperluas cakupan wilayahnya agar kebermanfaatannya dapat lebih luas.

Abstract

Olahraga saat kehamilan diketahui dapat mengurangi kelelahan dan nyeri saat kontraksi persalinan karena adanya pelepasan β-endorfin yang berperan sebagai inhibitor respon nyeri (analgesik). β-endorfin juga terbukti mengurangi kelelahan pada otot dan meningkatkan uptake glucose. Sementara itu, insulin telah diketahui juga berperan dalam uptake glucose ke dalam sel. Penelitian experimental menggunakan uji insiliko ini bertujuan untuk mengetahui apakah kerja β-endorfin dalam hal ini melalui ikatan pada reseptor insulin dan untuk mengetahui nilai kekuatan dari ikatan β-endorfin pada reseptor insulin dibandingkan dengan ikatan insulin pada reseptor insulin, sehingga dapat mengetahui efek sinergitas ikatan β-endorfin pada reseptor insulin, dan perannya dalam meningkatkan uptake glucose. Hasil penelitian menunjukan terdapat dua model ikatan β-endorfin pada reseptor insulin dengan asam amino yang sama terlibat dalam ikatan insulin, dengan kekuatan ikatan β-endorfin dengan reseptor insulin yang lebih kuat dibandingkan ikatan antara insulin dengan reseptornya.
