Abstract

A research was perfomed to find the proper of coconut water and natural ekstracts combined with Hyponex medium that compared with Vacin & Went medium. This research has been done at Tissue Culture Laboratory, Agricultural Faculty, Universitas Muhammadiyah Yogyakarta. The method of this research was arranged in Randomized Complete Design with 9 treatments and 8 replications. The treatments were VW + NAA 0,5 ppm + BAP 3 ppm, green Hyponex 3 g/l, green Hyponex 3 g/l + bean sprouts 150 g/l, green Hyponex 3 g/l + tomatoes 150 ml/l, green Hyponex 3 g/l + avocado 150 g/l, red Hyponex 3 g/l, red Hyponex 3 g/l + bean sprouts 150 g/l, red Hyponex 3 g/l + tomatoes 150 ml/l, red Hyponex 3 g/l + avocado 150 g/l. The each of Hyponex mediums were given with bananas 150 g/l and coconut water 150 ml/l. The result of this research showed that Hyponex mediums combined with natural ekstract was not signicicantly with Vacin & Went medium to high and shoot growth. Red Hyponex medium 3 g/l combined bananas 150 g/l and coconut water 150 ml/l resulted the best high, leaf and shoot in tissue culture Cattleya pastoral innocence orchid.

Abstract

Kepel (Stelechocarpus burahol [Bl] Hook F. & Th.) merupakan buah asli Indonesia berbiji banyak dengan ukuran yang besar. Bagian buah yang dapat dikonsumsi hanya 49% dengan bagian lain berupa biji. Perbanyakan kepel secara konvensional masih sulit dilakukan dengan hasil yang rendah. Kultur endosperma secara in vitro adalah metode perbanyakan yang tepat untuk memperoleh tanaman triploid dengan buah tanpa biji. Sterilisasi merupakan tahap awal yang menjadi kunci keberhasilan kultur in vitro. Hingga kini belum dilaporkan metode sterilisasi endosperma kepel secara in vitro yang tepat. Penelitian ini bertujuan untuk memperoleh metode sterilisasi eksplan yang tepat untuk kultur endosperma kepel. Penelitian ini telah dilaksanakan pada bulan September 2018 – Januari 2019 di Laboratorium Kultur In Vitro, Fakultas Pertanian, Universitas Muhammadiyah Yogyakarta. Penelitian ini menggunakan metode eksperimen disusun dalam Rancangan Acak Lengkap (RAL) faktor tunggal dengan perlakuan berbagai konsentrasi bahan sterilan yang terbagi 8 aras: H2O2 (3%10’, 3%15’, 5%10’, 5%15’) NaOCl (5%5’, 5%10’, 10%5’, 10%10’) dengan 3 kali ulangan dan 3 sempel. Parameter yang diamati yaitu: persentase kontaminasi, browning, hidup, vitrifikasi, jenis kontaminasi, waktu kontaminasi dan waktu browning. Hasil penelitian diperoleh bahwa perlakuan NaOCl 10% selama 10 menit merupakan metode sterilisasi paling tepat dengan presentase ekplan hidup 44,44%, persentase eksplan vitrifikasi 66,66%, serta tidak terjadi kontaminasi dan browning

Abstract

Porang (Amorphophallus muelleri B.) is a tuberous plant with the potential to be the main source of carbohydrates and is rich in benefits. Porang proliferation is limited by a 6-month dormancy period per year, and generative propagation is unlikely due to the seeds being apomictic triploid. The aimed of my research to analyze the application of BAP and NAA in culture media for direct propagation of porang shoots. The explant used in this research was young leaves. The research was arranged in a completely randomized design with a combination of BAP and NAA hormones added to the MS medium. There were three BAP treatments, namely 1.0 mg/L, 2.0 mg/L, and 3.0 mg/L, while NAA treatments consisted of 2 levels, namely 2.0 mg/L and 4.0 mg/L. The addition of 1.0 mg/L BAP combined with 4.0 mg/L NAA was the best treatment that produced seven shoots with an average shoot length of 2.14 cm and root length of 3.6 cm, with the earliest bud emergence (9.7 weeks after planting).

Abstract

The production of chrysanthemum plants in large quantities can be obtained by in vitro growth of chrysanthemums. The growth media that is often used is MS media which has a relatively high price. Therefore, a substitution medium that has a relatively affordable price is needed in the form of foliar fertilizer media with coconut water and ambon bananas added. This study aims to examine the best medium that can be used for in vitro chrysanthemum growth. The research was carried out in June-September 2020 at the In Vitro Culture Laboratory, Faculty of Agriculture, University of Muhammadiyah Yogyakarta. The research was carried out by a single-factor experimental method with 7 treatments and each had 5 replicates and each replicate consisted of 3 samples arranged with a Complete Random Design (RAL). The treatment that was experimented with was the use of foliar fertilizers in the form of Growmore and Hyponex Green with the addition of coconut water and bananas. The concentrations of Growmore foliar fertilizer given were 3 g/l, 4 g/l, and 5 g/l. The concentrations of Green Hyponex foliar fertilizer given were 3 g/l, 4 g/l, and 5 g/l. The results of the research showed that foliar fertilizer added with coconut water and banana ambon could be used as a substitution medium for MS media in chrysanthemum growth in vitro. The use of Growmore 4 g/l foliar fertilizer shows the best hail against the tall growth of chrysanthemum shoots in vitro

Abstract

Orchid Ph. amabilis is one of the plants designated as "Indonesian National Flower" and designated as "Puspa Pesona Indonesia". Orchid Ph. amabilis has a diversity that is not widely known, so that it is necessary to identify it molecularly. DNA isolation is the first step in molecular testing. This study aims to determine the sample weight, incubation time and the appropriate isolation method for DNA isolation from samples of Ph.amabilis orchid roots. In this study, the DNA isolation stage used two experimental methods, namely the CTAB method (Murray and Thompson) and the mini kit. The research method used was a single factor experiment with a combination of sample weight and incubation time consisting of 6 treatments with 3 replications, so 18 experimental units were obtained. The experimental treatment used was from the roots of the orchid Ph. amabilis which has a sample weight of 0.25 g, 0.5 g, and 0.75 g and each sample weight treatment was incubated for 30 minutes and 60 minutes. In addition, DNA isolation using the kit method had a sample weight of 0.25 g, 0.5 g and 0.75 g and an incubation period of 10 minutes. The results showed that the DNA isolation of Ph. amabilis with a sample weight of 0.75g, incubation time of 30 minutes using the CTAB method showed the best treatment in producing DNA with a concentration of 384 ng/µl and a purity of 1.9. This study is expected to provide information on sample weight, incubation time and the appropriate method for DNA isolation of Ph. amabilis orchid roots.
